Effects of ethanol and varenicline on female Sprague-Dawley rats in a third trimester model of fetal alcohol syndrome.

Perinatal ethanol exposure disrupts a variety of developmental processes in neurons important for establishing a healthy brain. These ethanol-induced impairments known as fetal alcohol spectrum disorder (FASD) are not fully understood, and currently, there is no effective treatment. Further, growing evidence suggests that adult females are more susceptible to ethanol, with the effects of perinatal ethanol exposure also being sexually divergent. Female models have been historically underutilized in neurophysiological investigations, but here, we used a third-trimester binge-ethanol model of FAS

Fetal alcohol syndrome surveillance: age of syndrome manifestation in case ascertainment.

BACKGROUND: Fetal alcohol syndrome (FAS) is a leading cause of developmental disability (Abel & Sokol, ). Active public health surveillance through medical record abstraction has been used to estimate FAS prevalence rates, typically based on birth cohorts. There is an extended time for FAS characteristics to become apparent in infants and young children, and there are often delays in syndrome recognition and documentation. This methodological study analyzes the age at case ascertainment in a large surveillance program. METHODS: The Fetal Alcohol Syndrome Surveillance (FASSLink) Project, funde

Effects of all three trimester moderate binge alcohol exposure on the foetal hippocampal formation and olfactory bulb.

OBJECTIVE: Pre-natal alcohol exposure results in injury to the hippocampus and olfactory bulb, but currently there is no consensus on the critical window of vulnerability. This study tested thehypothesis that pre-natal exposure to a moderate dose of alcohol during all three trimesterequivalentsalters development of the hippocampal formation and olfactory bulb in an ovinemodel, where all brain development occurs pre-natally as it does in humans.Research design and methods: Pregnant sheep were divided into saline control and abinge drinking groups (alcohol dose 1.75 g kg(-1); mean peak blood alco

Calcium-mediated repression of β-catenin and its transcriptional signaling mediates neural crest cell death in an avian model of fetal alcohol syndrome.

Fetal alcohol syndrome (FAS) is a common birth defect in many societies. Affected individuals have neurodevelopmental disabilities and a distinctive craniofacial dysmorphology. These latter deficits originate during early development from the ethanol-mediated apoptotic depletion of cranial facial progenitors, a population known as the neural crest. We showed previously that this apoptosis is caused because acute ethanol exposure activates G-protein-dependent intracellular calcium within cranial neural crest progenitors, and this calcium transient initiates the cell death. The dysregulated sign

Beta-endorphin neuronal cell transplant reduces corticotropin releasing hormone hyperresponse to lipopolysaccharide and eliminates natural killer cell functional deficiencies in fetal alcohol exposed rats.

BACKGROUND: Natural killer (NK) cell dysfunction is associated with hyperresponse of corticotropin releasing hormone (CRH) to immune challenge and with a loss of beta-endorphin (BEP) neurons in fetal alcohol exposed animals. Recently, we established a method to differentiate neural stem cells into BEP neurons using cyclic adenosine monophosphate (cAMP)-elevating agents in cultures. Hence, we determined whether in vitro differentiated BEP neurons could be used for reversing the compromised stress response and immune function in fetal alcohol exposed rats. METHODS: To determine the effect of BE

Design, construction, and testing of a stereo-photogrammetric tool for the diagnosis of fetal alcohol syndrome in infants.

Stereo-photogrammetry provides a low cost, easy to use, and noninvasive alternative to traditional facial anthropometry for the diagnosis of fetal alcohol syndrome (FAS). We describe such a system for use in obtaining 3-D facial information in infants. The infant is photographed using three high resolution digital cameras simultaneously while seated in a car seat. The subject's head is enclosed in a control frame during imaging. Technical system tests, namely control frame interpolation, camera calibration reliability, and camera synchronization delay assessments were performed. Direct and ste

What human service professionals know and want to know about fetal alcohol syndrome.

BACKGROUND: Although human service professionals are critical to prevention of primary and secondary disabilities among those who are or could be affected by prenatal exposure to alcohol surprisingly little information is available on their knowledge, attitudes, or beliefs about this problem. This article presents the results from a statewide survey (2005) undertaken in the United States to gain such information. OBJECTIVE: The purpose of the study was to : a) measure the knowledge, attitudes, and beliefs about Fetal Alcohol Syndrome (FAS) of professionals working in the fields of child welfa

Fetal alcohol syndrome in association with Rett syndrome.

Fetal alcohol syndrome in association with RETT syndrome: We report on a girl with neonatal dystrophy, microcephaly, heart defect, and the characteristic features of alcohol embryopathy. Later, she developed distinctive features of RETT syndrome including loss of early acquired developmental skills and presented typical symptoms of RETT syndrome as reduction of communication skills, reduction of hand function, hyperventilation, and grinding of teeth. Molecular analysis of the MECP2 gene revealed the c.808T>C (R270X) mutation located in the nuclear localisation signal sequence of the gene. Our

Children with Fetal Alcohol Syndrome are impaired at place learning but not cued-navigation in a virtual Morris water task.

We employed a computerized (virtual) Morris water task (VMWT) to measure place learning and cued-navigation in eight adolescent males (9.5-16.5 years old) diagnosed with Fetal Alcohol Syndrome (FAS). Eight adolescent males matched for age and ethnicity with no history of prenatal alcohol exposure served as controls. Participants were trained to navigate to a hidden platform in a fixed location relative to a set of four conspicuous extramaze cues. After 20 hidden platform trials, a single no-platform probe trial was conducted, followed by 8 trials during which the platform was visible (cued-nav

Emotion-related learning in individuals prenatally exposed to alcohol: an investigation of the relation between set shifting, extinction of responses, and behavior.

The association between deficits in emotion-related learning, conceptual set shifting, and behavioral problems was investigated in individuals with substantial prenatal alcohol exposure. Twenty subjects with confirmed prenatal alcohol exposure (10 of whom were diagnosed as having Fetal Alcohol Syndrome) and 20 normal controls matched for age, gender, and ethnic background participated. The two groups were administered a battery of tests including two tests of emotion-related learning (visual discrimination reversal and extinction of reward-response associations), tests of conceptual set shifti

In utero ethanol exposure increases proenkephalin, a precursor of a neuropeptide that is inhibitory to neuronal growth.

BACKGROUND: One of the effects of fetal alcohol syndrome (FAS) is altered motor function. In an attempt to elucidate a potential cause of this ethanol-associated damage, we investigated the effects of in utero ethanol exposure on the production of proenkephalin (PE). METHODS: PE is the precursor of met- and leu-enkephalin, two neuropeptides that inhibit the proliferation of neurons and astrocytes. PE mRNA and PE peptide were measured in the striatum and nucleus accumbens because of the importance of these brain regions to motor function, their sensitivity to the effects of in utero ethanol ex

Electrophysiological characterization of cerebellar neurons from adult rats exposed to ethanol during development.

The purpose of this study was to investigate the spontaneous activity of mature rat cerebellar neurons that had been exposed to ethanol (EtOH) during postnatal days 4 to 10, which corresponds to the third trimester in humans. Newborn Sprague-Dawley rats were implanted with gastric feeding tubes and were artificially reared from postnatal days 4 to 10 with two different diets. The experimental group received 4.5 g/kg/day of EtOH delivered in a milk solution. Controls received similar feeding with an isocaloric supplement replacing the EtOH. Electrophysiological evaluations were performed after

Effects of prenatal ethanol exposure on brain region NMDA-mediated increase in intracellular calcium and the NMDAR1 subunit in forebrain.

The effects of prenatal ethanol exposure on N-methyl-D-aspartate (NMDA)-mediated calcium entry into neonatal dissociated neurons from hippocampus, forebrain, and cerebellum were investigated. Dissociated cells were isolated from less than 1-day-old pups of prenatally exposed, pair-fed control and ad libitum control groups and loaded with fura-2. Prenatal ethanol exposure significantly reduced the NMDA-stimulated increase in intracellular calcium in all three brain regions compared to the two control groups. These findings are very similar to those previously observed in neonatal dissociated wh

Prenatal alcohol exposure results in hyperactivity of the hypothalamic-pituitary-adrenal axis of the offspring: modulation by fostering at birth and postnatal handling.

Exposure of fetal rats to alcohol results in permanent hyperactivity of the hypothalamic-pituitary-adrenal (HPA) axis. In contrast, postnatal handling or fostering have been reported to restrain HPA activity. Because of the deleterious consequences of a hyperresponsive HPA axis, we thought that the possibility that postnatal manipulations might be able to reverse the influence of prenatal alcohol treatment deserved investigation. To test this hypothesis, we exposed rat dams to alcohol by inhalation during the second week of gestation. At birth, pups were either fostered or remained with their

Maternal drinking during pregnancy: attention and short-term memory in 14-year-old offspring--a longitudinal prospective study.

A large and compelling experimental literature has documented the adverse impact of prenatal alcohol exposure on the developing brain of the offspring. This is the first report of adolescent attention/memory performance and its relationship with prenatal alcohol exposure in a population-based, longitudinal, prospective study (n = 462) involving substantial covariate control and "blind" examiners. Prenatal alcohol exposure was significantly related to attention/memory deficits in a dose-dependent fashion. A latent variable reflecting 13 measures of maternal drinking was correlated 0.26 with a l

Difference in susceptibility to teratogenic effects of alcohol in discordant twins exposed to alcohol during the second half of gestation.

A pair of discordant twins exposed to heavy maternal alcohol consumption only during the second half of pregnancy is reported. Apparently, differences in susceptibility to the dysmorphogenic influence of ethanol caused 1 twin to be more severely affected than the other. One twin had prenatal growth retardation, neonatal withdrawal symptoms, delay in both motor and cognitive function during the first year of life, slowing of background activity on electroencephalography, and cortical and central brain atrophy on computed tomography. Catch-up growth occurred during the postnatal period for the a

Alcohol exposure before pregnancy: effect on GABA levels and turnover in rat offspring.

The GABA levels and turnover rates in various brain areas from 2-month-old rats born to mothers who consumed 20% (v/v) alcohol during 1 month only before pregnancy, were investigated. A decreased level was found in the olfactory tubercules and an increase was observed in the hypothalamus. The turnover rates were reduced in both areas, whereas an increase was observed in the frontal cortex. These results indicate that biochemical alterations may occur in the offspring even if the fetus did not develop under alcoholization.

[Eye manifestations of fetal alcohol syndrome].

Alcohol is the most frequent and most important teratogenic agent causing mental and physical retardation in childhood. The alcohol fetal syndrome is characterized by pre- and postnatal growth retardation, hypotonia, hyperactivity, microcephalus, mental retardation and typical craniofacial malformations. The latter includes short palpebral fissures, a poorly developed philtrum, thin upper lip vermillion, short mandibles, a flattened midface structure and dysplastic ears. Ophthalmological signs occur in 90% and include epicanthus, ptosis, myopia, optic nerve hypoplasia and tortuous retinal vess

Fetal alcohol syndrome in adolescents and adults.

Fetal alcohol syndrome is a specific recognizable pattern of malformation. Manifestations in 61 adolescents and adults suffering from alcohol teratogenesis are presented. After puberty, the faces of patients with fetal alcohol syndrome or fetal alcohol effects were not as distinctive. Patients tended to remain short and microcephalic, although their weight was somewhat closer to the mean. The average IQ was 68, but the range of IQ scores widely varied. Average academic functioning was at the second- to fourth-grade levels, with arithmetic deficits most characteristic. Maladaptive behaviors suc
