A NET-competent neutrophil subpopulation drives early brain injury after subarachnoid hemorrhage.

INTRODUCTION: Early brain injury (EBI) following subarachnoid hemorrhage (SAH) critically determines neurological outcomes, yet the cellular drivers remain unclear. METHODS: To systematically dissect acute pathogenic programs, we performed single-cell RNAsequencing of CD45+ immune cells isolated from a mouse model of SAH, coupled with weighted gene co-expression network analysis (WGCNA). RESULTS: This revealed a 24-h-specific injury module enriched for inflammatory and myeloid-related pathways. Mapping this module onto the single-cell landscape identified neutrophils as the principal cellula

Peripheral immune and vascular inflammatory signatures precede Alzheimer's disease onset: A serum proteomic study.

BackgroundNeuroinflammation plays an important role in the pathogenesis of Alzheimer's disease, but systemic immune alterations preceding clinical onset remain insufficiently characterized. Peripheral immune and vascular inflammatory processes may influence the central nervous system through endothelial dysfunction and blood-brain barrier vulnerability. Serum proteomics offers a minimally invasive approach to identify such early systemic changes.ObjectiveIn this population-based longitudinal cohort study, we analyzed baseline serum proteomic profiles from cognitively normal older adults who we

PAD4-DB: A Curated Structure-Activity Resource Reveals Hub-Organized Activity Cliffs and Scaffold-Dependent SAR Ruggedness in PAD4 Inhibitors.

Peptidylarginine deiminase 4 (PAD4) is an increasingly prominent therapeutic target in oncology, inflammatory disease, and neutrophil extracellular trap (NET)-associated pathologies, yet public bioactivity data for PAD4 inhibitors remain fragmented across multiple repositories with substantial redundancy and inconsistent annotation. Here, we present PAD4-DB, a curated structure-activity relationship resource integrating 3093 unique inhibitors (consensus pIC50 range 2.00-8.52; median 6.84) from PubChem, ChEMBL, and BindingDB through a reproducible pipeline encompassing structure standardization

The landscape of protein post-translational modifications in the pathogenesis of acute respiratory distress syndrome.

Acute respiratory distress syndrome (ARDS) remains one of the most severe forms of acute lung injury, characterized by diffuse alveolar damage and refractory hypoxemia secondary to non-cardiogenic pulmonary edema. After many years of research and incremental improvements in ventilatory and supportive strategies, mortality rates are still disappointingly high and no pharmacological agent has convincingly demonstrated a mortality benefit in large trials. Elucidating the rapid molecular processes driving the acute phase of lung injury, particularly post-translational modifications (PTMs), may be

Vaccine-Induced Immune Thrombotic Thrombocytopenia (VITT): An Immunopathogenic Model of Dysregulated Vaccine-Triggered Immunity.

BACKGROUND/OBJECTIVES: Vaccine-induced immune thrombotic thrombocytopenia (VITT) is a rare but severe immune-mediated adverse event associated with adenoviral vector-based SARS-CoV-2 vaccines. Beyond its clinical relevance, VITT provides a unique human model of vaccine-triggered autoimmunity and immune-thrombosis. This review critically reassesses the immunopathogenic framework of VITT in light of recent evidence. METHODS: We conducted a structured narrative review of studies published between 2021 and 2025, focusing on clinical, epidemiological, and mechanistic data relevant to PF4 immunogen

The expression signature, prognostic significance and immune cell infiltration of the OAS gene family in gastric cancer.

Gastric cancer (GC) is a common malignant tumour and a leading cause of cancer-related deaths worldwide. Although the OAS gene family has been implicated in various tumour-related biological processes, its specific role in GC remains unclear. This study integrated bioinformatics analysis of public datasets with experimental validation using clinical GC specimens to investigate the function of the OAS gene family in GC. We found that OAS family members were significantly upregulated in GC and associated with poor patient prognosis. Furthermore, OAS expression levels correlated with genomic muta

Neutrophil elastase promotes low molecular weight cyclin E1 formation to accelerate osteosarcoma proliferation.

INTRODUCTION: Osteosarcoma (OS) is the most common primary bone malignancy, characterized by aggressive local invasion and a high propensity for metastasis. We previously reported that cyclin E1 is upregulated in osteosarcoma. In this study, we identified a cytoplasmic, low molecular weight cyclin E1 isoform (LMW-cyclin E1) in osteosarcoma that is significantly associated with poor patient outcomes. METHODS: We collected RNA sequencing data to analyze the cyclin E1 (CCNE1) expression and performed Western blot assay, immunofluorescence, and immunohistochemistry staining to validate cyclin E1

Inflammatory gene expression profile of oral plasmablastic lymphoma.

Plasmablastic lymphoma (PBL) is a rare and aggressive non-Hodgkin lymphoma with a poor prognosis and short survival rates. It is classified as a large B-cell lymphoma subtype, but carries a plasmacytic immunophenotype. Therefore, PBL has pathogenetic overlaps with diffuse large B-cell lymphoma not otherwise specified (DLBCL NOS) and plasma cell neoplasms (PCNs). Although recent studies have characterized important genetic abnormalities in PBL, the precise molecular mechanisms driving its pathogenesis remain elusive. In this study, we analyzed 607 inflammatory genes in 10 PBL, 11 DLBCL NOS/high

KLF7-IT1-ARPC2 axis in macrophages orchestrates caspase-4-driven hypercoagulation in sepsis.

Sepsis is characterized by dysregulated inflammation and coagulation leading to organ dysfunction, yet the molecular mechanisms linking these processes remain elusive. Through transcriptomic profiling of leukocytes from septic and non-septic patients, we identified Kruppel-like factor 7 intronic transcript 1 (KLF7-IT1) as one of the most robustly upregulated long non-coding RNAs in sepsis. Elevated KLF7-IT1 expression correlated strongly with hypercoagulability, inflammatory cytokines, and multi-organ injury, suggesting a pathogenic role in septic coagulopathy. Functional studies revealed that

Combined antiresorptive and new anabolic drug approach in osteogenesis imperfecta zebrafish models.

Osteogenesis imperfecta (OI) is a family of heritable collagen I-related skeletal disorders for which, to date, no definitive cure is available. Individuals with OI are mainly treated with bisphosphonates that enhance bone mass by inhibiting bone resorption. However, new strategies combining antiresorptive molecules with bone anabolic drugs are likely to provide valid alternatives for skeletal health, protecting physiological bone turnover. Recently, cellular stress has been identified as a therapeutic target in both dominant and recessive forms of OI characterized by overmodified collagen I.

Identification of anti-resorptive GPCRs by high-content imaging in human osteoclasts.

Osteoporosis diagnoses are increasing in the ageing population, and although some treatments exist, these have several disadvantages, highlighting the need to identify new drug targets. G protein-coupled receptors (GPCRs) are transmembrane proteins whose surface expression and extracellular activation make them desirable drug targets. Our previous studies have identified 144 GPCR genes to be expressed in primary human osteoclasts, which could provide novel drug targets. The development of high-throughput assays to assess osteoclast activity would improve the efficiency at which we could assess

Computer-aided repositioning and functional in vitro assessment of novel PAD4 inhibitors.

Peptidyl arginine deiminase 4 (PAD4) is a protein that catalyzes both normal and abnormal citrullination of interacting protein partners, affecting gene regulation and being associated with diseases such as Alzheimer's, cancer, and rheumatoid arthritis (RA). As a result, PAD4 has emerged as a potential therapeutic target; however, no inhibitors have been approved to date. In this study, the REFRAME and ZINC15 drug databases were virtually screened. The approach used molecular docking and dynamics simulation techniques to identify compounds with high-predicted binding affinity to PAD4 (PDB ID:

Inflammatory neutrophil responses and T cell activation in ART-treated SIVmac239-infected rhesus macaques.

Modern antiretroviral therapy (ART) regimens have revolutionized the management of human immunodeficiency virus (HIV) and transformed it from a life-threatening disease to a manageable chronic condition. Despite the durable viral suppression associated with ART adherence, people with HIV (PWH) continue to experience chronic immune activation and inflammation, which has been linked with increased risk of developing non-acquired immunodeficiency syndrome (AIDS) comorbidities, including cardiovascular disease, liver disease, or neurocognitive disorders. Importantly, the mechanisms underlying esta

Pathogenic mechanisms of disease in idiopathic inflammatory myopathies: autoantibodies as clues.

Idiopathic inflammatory myopathies (IIMs) encompass a spectrum of autoimmune diseases characterized by muscle inflammation and systemic involvement. This review aimed to synthesize current evidence on the clinical significance and pathogenic mechanisms underlying autoantibodies associated with IIMs. Autoantibodies targeting aminoacyl-tRNA synthetases (ARS) play a pivotal role in antisynthetase syndrome (ASS), highlighting associations with interstitial lung disease (ILD) and distinctive clinical features. Anti-Mi-2 antibodies in dermatomyositis (DM) are hallmarked by characteristic cutaneous m

pH-Responsive, Charge-Reversing Layer-by-Layer Nanoparticle Surfaces Enhance Biofilm Penetration and Eradication.

Microbes entrenched within biofilms can withstand 1000-fold higher concentrations of antibiotics, in part due to the viscous extracellular matrix that sequesters and attenuates antimicrobial activity. Nanoparticle (NP)-based therapeutics can aid in delivering higher local concentrations throughout biofilms as compared to free drugs alone, thereby enhancing the efficacy. Canonical design criteria dictate that positively charged nanoparticles can multivalently bind to anionic biofilm components and increase biofilm penetration. However, cationic particles are toxic and are rapidly cleared from c
