Vinca Extract Infused Silk Fibroin Hydrogel for Treatment of Artificially Induced Diabetic Foot Ulcer in Murine Model.

Diabetic foot ulcers are challenging to manage and require intensive treatment as they heal slowly and greatly affect quality of life due to reduced mobility, infection and sepsis. The aim of current study was to develop novel hybrid hydrogels of silk fibroin containing 0.5%, 1.0% and 1.5% of Catharanthus roseus extract. Prepared hydrogels were characterized and GC-MS analysis of C. roseus extract was done to assess the component composition. Swiss albino mice were injected with alloxan to develop diabetic models and foot ulcer was induced. Hydrogels were administered every other day and wound

Phytochemistry and pharmacology of Melaleuca alternifolia: Bridging aboriginal heritage with contemporary science.

Melaleuca alternifolia (Australian tea tree) sits at a rare intersection of ethnomedicine and modern pharmacology. Rooted in Bundjalung Aboriginal practice for respiratory, dermatologic, and wound care, its essential oil (TTO) has since been validated as a multi-target agent. We synthesize advances spanning cultivation ecology, chemistry, mechanisms, and translation. Chemotyped oils (ISO 4730) are dominated by terpinen-4-ol-supported by γ-/α-terpinene, 1,8-cineole, and selected sesquiterpenes-whose coordinated actions destabilize microbial membranes, impair energy metabolism, modulate redox an

Staphylococcal proliferation on skin models to investigate novel anti-infective treatments against dysbiosis.

Inflammatory skin conditions like Acne inversa are characterized by dysbiosis, an imbalance of commensal and pathogenic bacteria, posing challenges for specific treatments. Consequently, we investigated how biofilm formation, low-nutrition skin environments, and air interfaces influence susceptibility to anti-infective treatments in mixed bacterial cultures. To achieve this in a cost-effective and reproducible manner, we developed a simplified substrate made of gelatin, hyaluronic acid, chondroitin sulphate, and alginate (=Gel-Alg). This in vitro model simulates biofilm cultivation on skin sur

Reprogramming ligand-receptor interactions via a self-powered molecular engine for proton-responsive regulation of cellular motility.

Manipulation of ligand-receptor interactions (LRIs) in a user-defined manner is one of the effective ways to regulate cell motility and disease progression. Molecular engines orchestrate LRIs to regulate cell motility by harnessing various forms of energy within physiological surroundings. However, engineering chemical engines remains largely unexplored, primarily due to the insufficient chemical cue density, uncertain conformational changes of scaffolds, and a limited repertoire of environment-responsive switchable scaffolds. Enzymatic biofuel cells (EBFCs) are electrochemical devices that co

Inorganic materials to prevent liquefactive necrosis and improve wound healing: A role of infection control across ischemic models.

Liquefactive necrosis, clinically manifesting as wet gangrene, is a catastrophic complication of ischemic wounds driven by bacterial overgrowth, proteolytic tissue lysis, and loss of structural integrity; current treatments remain largely limited to surgical debridement or amputation. Here, we show that inorganic biomaterial films releasing hydroxide ions, with or without oxygen and hydrogen peroxide, can markedly inhibit the development and spread of liquefactive necrosis in ischemic skin and cartilage. A calcium peroxide (CaO2) loaded film in alginate hydrogel was evaluated in ischemic rat d

Biomimetic Hydrogels with Nucleus Pulposus-like Viscoelasticity and ECM Peptides for Discogenic Differentiation of Stem Cells.

Intervertebral disc (IVD) degeneration is a leading cause of low back pain (LBP), primarily originating in the nucleus pulposus (NP). Regenerative strategies combining mesenchymal stem cells (MSCs) with biomaterials offer great potential for NP repair by replenishing cells and restoring extracellular matrix (ECM). However, key translational challenges remain, including limited stem cell differentiation, poor cell survival in the harsh degenerative niche, and insufficient biomaterial support. While matrix viscoelasticity has been shown to influence adipose-derived stem cell (ASC) discogenic dif

Development of Patient-Derived Conditionally Reprogrammed 3D Breast Cancer Culture Models for Drug Sensitivity Evaluation.

BACKGROUND: Therapeutic responses of breast cancer vary among patients and lead to drug resistance and recurrence due to the heterogeneity. Current preclinical models, however, are inadequate for predicting individual patient responses towards different drugs. This study aimed to investigate the patient-derived breast cancer culture models for drug sensitivity evaluations. METHODS: Tumor and adjacent tissues from female breast cancer patients were collected during surgery. Patient-derived breast cancer cells were cultured using the conditional reprogramming technique to establish 2D models. T

Current Insights into Plausible Mechanisms of Chromium (VI) Neurotoxicity in the Brain and Future Perspectives.

BACKGROUND: Hexavalent chromium (Cr (VI)) is a known neurotoxin and environmental contaminant. Despite its recognition, the underlying mechanisms by which Cr (VI) induces neurological damage remain insufficiently explored. The complexities of the Central Nervous System (CNS), including the Blood Brain Barrier (BBB) and supporting brain cells, contribute to regions-specific susceptibility within the brain. Understanding Cr (VI) neurotoxicity is crucial for its potential role in neurodegenerative diseases. METHODS: A Systematic Review was conducted using international databases (PubMed, Medline

Molecular epidemiology and antimicrobial resistance profiles of Pseudomonas aeruginosa causing bloodstream infections in neutropenic cancer patients.

BACKGROUND: Bloodstream infections (BSI) in neutropenic cancer patients, particularly those caused by Pseudomonas aeruginosa (PA), are associated with high morbidity and mortality. The increasing prevalence of multidrug-resistant (MDR) and extensively drug-resistant (XDR) PA strains complicates clinical management. This study aimed to characterise PA strains causing BSI in neutropenic cancer patients and assess the association between microbiological features and clinical outcomes. METHODS: We analysed PA strains from 94 BSI episodes in neutropenic cancer patients across five Spanish hospital

Bioreduction of N-(3-oxobutyl)heterocycles with flexible ring by yeast whole-cell biocatalysts.

This study explored the bioreduction of N-(3-oxobutyl)heterocycles with (partially) saturated heterocyclic moieties using whole-cell forms of wild-type yeast strains and commercially available baker's yeast (Saccharomyces cerevisiae). Eleven wild-type yeast strains and baker's yeast were screened for ketoreductase activity on a series of five flexible N-heterocycles with prochiral carbonyl group in the N-(3-oxobutyl) substituent. Among the yeast strains tested, Candida parapsilosis (WY12) proved to be the most efficient biocatalyst in the bioreductions, resulting in the corresponding enantiopu

An anti-FAP-scFv-functionalized exosome-carrying hydrogel delivers SKI mRNA to fibrotic nucleus pulposus cells to alleviate intervertebral disc degeneration by regulating FOXO3.

Rationale: Nucleus pulposus (NP) fibrosis is a contributing factor to intervertebral disc degeneration (IVDD), which lacks an effective treatment. This study focuses on elucidating the role and mechanisms of the TGF-β signaling repressor SKI in NP fibrosis and exploring its therapeutic potential. Methods: Single-cell RNA sequencing (scRNA-seq) was used to investigate fibrotic nucleus pulposus cell (NPC) subsets and assess TGF-β signaling activation. Two recombinant plasmids encoding single-chain variable fragments (scFvs) targeting the fibrotic NPC marker FAP and SKI mRNA were co-transfected i

Amelogenin Null Mice Develop Osteoarthritis, While Its Application Mitigates Disease Phenotypes in a Rat Model.

Previous studies have demonstrated that recombinant human amelogenin protein (rHAM+) promotes healing of injured articular cartilage, subchondral bone, and skeletal ligaments. Therefore, we speculated that amelogenin may play a role in osteoarthritis (OA) development. Aged amelogenin-null and wild-type mice underwent micro-computed tomography (micro-CT) and histological analyses to assess OA-related changes. Additionally, OA was induced in rat knees via destabilization of the medial meniscus, followed by treatment with 0.5 mg/mL rHAM+ dissolved in propylene glycol alginate (PGA) or PGA alone.

Human progenitor T-cell differentiation regulated by the mechanical resistance of thymus-mimetic extracellular matrices.

Therapeutic T-cell engineering ex vivo from human hematopoietic stem cells (HSCs) focuses on recapitulating notch1-signaling and α4β1-integrin-mediated adhesion within the thymic niche with supportive stromal cell feeder-layers or surface-immobilized recombinant protein-based engineered thymic niches (ETNs). The relevant Notch1-DLL-4 and α4β1-integrin-VCAM-1 interactions are known to respond to mechanical forces that regulate their bond dissociation behaviors and downstream signal transduction, yet manipulating the mechanosensitive features of these key receptor-ligand interactions in thymopoi

Bioengineering scalable and drug-responsive in vitro human multicellular non-alcoholic fatty liver disease microtissues encapsulated in the liver extracellular matrix-derived hydrogel.

Non-alcoholic fatty liver disease (NAFLD) is a high-prevalence and progressive disorder. Due to lack of reliable in vitro models to recapitulate the consecutive phases, the exact pathogenesis mechanism of this disease and approved therapeutic medications have not been revealed yet. It has been proven that the interplay between multiple hepatic cell types and liver extracellular matrix (ECM) are critical in NAFLD initiation and progression. Herein, a liver microtissue (LMT) consisting of Huh-7, THP-1, and LX-2 cell lines and human umbilical vein endothelial cells (HUVEC), which could be substit
