Global incidence of childhood cancer by subtype in 2022: a population-based registry study.

BACKGROUND: Childhood cancers are an important cause of premature mortality and long-term disability worldwide, yet most global estimates stop at broad diagnostic groups. A clearer picture by subtype and its social determinants is needed to guide the World Health Organization (WHO) target of at least sixty percent survival for all children with cancer by 2030. METHODS: In this population-based registry study, we analysed incidence and mortality data for 185 countries in 2022 using the GLOBOCAN database, and redistributed unspecified tumours according to subtype proportions derived from 670 hi

Early CD8+-recovery independently predicts low probability of disease relapse but also associates with severe GVHD after allogeneic HSCT.

In this single-center study we retrospectively evaluated the impact of early reconstitution of different lymphocyte subsets on patient outcomes after allogeneic hematopoietic stem cell transplantation (allo-HSCT). We found that CD8+ T-cell counts exceeding 50x10(6)/l as early as on day 28 post-transplantation correlated significantly with decreased relapse risk, with three-year relapse rates of 17.0% and 55.6% (P = 0.002), but were also associated with severe acute and chronic GVHD. Incidence of grade III-IV acute GVHD was 30.5% for those with early CD8+ T-cell recovery compared to 2.1% for th

A review of the first year of Medicare coverage of erythropoietin.

Recombinant human erythropoietin (rHuEPO) is a new drug for treating anemia associated with end stage renal disease (ESRD). In a study of rHuEPO diffusion, costs, and effectiveness, we analyze ESRD program data and all claims submitted to Medicare for reimbursement of rHuEPO administered to ESRD dialysis patients. Access to rHuEPO was rapid and extensive during the first year of Medicare coverage. Dosing of rHuEPO and achieved hematocrit were lower than expected based on the results of clinical trials. rHuEPO cost Medicare $144 million in its first year. The analysis of insurance claims data a

Adoptive cell therapies in solid tumors: current clinical landscape, challenges, and future directions.

Adoptive cell therapy (ACT) has emerged as a transformative strategy in cancer immunotherapy, offering durable clinical benefit in hematologic malignancies and expanding therapeutic potential in solid tumors. However, the translation of ACT to solid malignancies remains constrained by biological, immunological, and logistical challenges. This narrative review provides an evidence based overview of the current clinical landscape of ACT in solid tumors, with a focus on chimeric antigen receptor T cell (CAR-T), tumor-infiltrating lymphocyte (TIL), and T cell receptor-engineered T cell (TCR-T) the

Osteoid cell-derived chemokines drive bone-metastatic prostate cancer.

One of the greatest challenges in improving prostate cancer (PCa) survival is in designing new therapies to effectively target bone metastases. PCa regulation of the bone environment has been well characterized; however, bone-targeted therapies have little impact on patient survival, demonstrating a need for understanding the complexities of the tumor-bone environment. Many factors contribute to creating a favorable microenvironment for prostate tumors in bone, including cell signaling proteins produced by osteoid cells. Specifically, there has been extensive evidence from both past and recent

Lost in Translation (LiT): IUPHAR Review 6.

Translational medicine is a roller coaster with occasional brilliant successes and a large majority of failures. Lost in Translation 1 ('LiT1'), beginning in the 1950s, was a golden era built upon earlier advances in experimental physiology, biochemistry and pharmacology, with a dash of serendipity, that led to the discovery of many new drugs for serious illnesses. LiT2 saw the large-scale industrialization of drug discovery using high-throughput screens and assays based on affinity for the target molecule. The links between drug development and university sciences and medicine weakened, but t

Research Advances on Targets and Mechanisms in Cancers Complicated by Cardiovascular Diseases.

PurposeCancers complicated by cardiovascular diseases (CVDs) are increasingly becoming major limiting factors affecting patients' long-term quality of life and clinical outcomes. Systematic identification of therapeutic targets and their clinical development status is crucial for optimizing treatment strategies. Therefore, this study aimed to establish a target-based analytical framework to systematically map the distribution, developmental stage, maturity, and mechanistic characteristics of clinical trials investigating tumors co-occurring with CVD, thereby identifying potential therapeutic t

Infection risk associated with talquetamab in relapsed/refractory multiple myeloma: a systematic review with meta-analysis of talquetamab monotherapy and descriptive analysis of combination therapy.

BACKGROUND: Talquetamab, a first-in-class G protein-coupled receptor family C group 5 member D (GPRC5D) × CD3 bispecific antibody, demonstrates substantial clinical activity in relapsed/refractory multiple myeloma (RRMM). However, immune dysfunction associated with advanced disease, extensive prior therapies, and bispecific antibody-mediated immune modulation may increase susceptibility to infections. This systematic review and meta-analysis aimed to characterize infection risk associated with talquetamab, with separate evaluation of monotherapy and combination therapy settings. METHODS: PubM

Targeted drug combination therapy design based on driver genes.

Targeted therapies against cancer types with more than one driver gene hold bright but elusive promise, since approved drugs are not available for all driver mutations and monotherapies often result in resistance. Targeting multiple driver genes in different pathways at the same time may provide an impact extensive enough to fight resistance. Our goal was to find synergistic drug combinations based on the availability of targeted drugs and their biological activity profiles and created an associated compound library based on driver gene-related protein targets. In this study, we would like to

ENHANZE® drug delivery technology: a novel approach to subcutaneous administration using recombinant human hyaluronidase PH20

ENHANZE® drug delivery technology is based on the proprietary recombinant human hyaluronidase PH20 enzyme (rHuPH20; Halozyme Therapeutics, Inc.) that facilitates the subcutaneous (SC) delivery of co-administered therapeutics. rHuPH20 works by degrading the glycosaminoglycan hyaluronan (HA), which plays a role in resistance to bulk fluid flow in the SC space, limiting large volume SC drug delivery, dispersion, and absorption. Co-administration of rHuPH20 with partner therapies can overcome administration time and volume barriers associated with existing SC therapeutic formulations, and has been

Epidemiology of Tumor-Induced Osteomalacia in Germany Based on Real World Data.

Tumor-induced osteomalacia (TIO) is an ultra-rare disease caused mostly by benign tumors that secrete fibroblast growth factor-23. Because of nonspecific symptoms, the diagnostic delay is long, and therapy can be challenging. Moreover, epidemiological data on TIO are scarce owing to its rarity. Therefore, this study aimed to quantify TIO's incidence rates and prevalence in Germany. Retrospective longitudinal and cross-sectional analyses were conducted using anonymized German claims data from the statutory health insurance (SHI) database. This database, which comprises the data of approximately

Individualized, Single Session Yoga Therapy to Reduce Physical and Emotional Symptoms in Hospitalized Hematological Cancer Patients.

Objective: Inpatient treatment of hematological cancer is among the most physically and mentally arduous cancer treatments, and it is associated with a number of common physical, emotional, and social symptoms that can negatively affect quality of life (QOL) for years following treatment. While treating symptoms during hospitalization holds promise for improving long-term QOL, successful approaches likely require multidisciplinary interventions. In this article, we describe a 4-year effort in program enhancement that incorporated an adjunctive single yoga therapy session during treatment for h

Fit & strong! Promotes physical activity and well-being in older cancer survivors.

IntroductionPhysical activity reduces fatigue and depression while improving quality of life in cancer survivors. Exercise is generally considered safe and is recommended to survivors of all ages. Despite the high prevalence of cancer in the elderly, few studies address physical activity interventions targeting this older population. Fit & Strong! is an evidence-based physical activity program shown to improve level of physical activity, exercise-self-efficacy, and mood in older adults with osteoarthritis. This study tests the feasibility and short-term impact of the Fit & Strong! exercise pro

Clinical Evidence Linkage From the American Society of Clinical Oncology 2024 Conference Poster Images Using Generative AI: Exploratory Observational Study.

BACKGROUND: Early-stage clinical findings often appear only as conference posters circulated on social media. Because posters rarely carry structured metadata, their citations are invisible to bibliometric and alternative metric tools, limiting real-time research discovery. OBJECTIVE: This study aimed to determine whether a large language model can accurately extract citation data from clinical conference poster images shared on X (formerly known as Twitter) and link those data to the Dimensions and Altmetric databases. METHODS: Poster images associated with the 2024 American Society of Clin
