Abnormalities of the corpus callosum in children prenatally exposed to alcohol.

For 20 years, it has been known that fetal alcohol syndrome (FAS) is associated with abnormal brain development. Early autopsy studies point to the corpus callosum as one area affected by heavy alcohol exposure. Little is known, however, about the integrity of the brain in alcohol-exposed children who survive the perinatal period. This study was designed to assess the corpus callosum in living children exposed to high doses of alcohol prenatally. Thirteen children with histories of significant prenatal alcohol exposure and 12 normal control children were evaluated using magnetic resonance imag

Effect of pre- or postnatal exposure to ethanol on the total number of neurons in the principal sensory nucleus of the trigeminal nerve: cell proliferation and neuronal death.

Early exposure to ethanol reduces the number of neurons in many CNS structures in vivo. The present study determined whether such reductions are caused by the death of neurons. Three groups of ethanol-treated rats were prepared: those exposed to ethanol from gestational day (G) 11 to G19 (during the period of neuronal generation and migration), from postnatal day (P) 4 to P12 (during the period of synaptogenesis), or from P31 to P39 [after the mature structure and function of neurons in the principal sensory nucleus (PSN) of the trigeminal nerve was established]. During these times, pregnant d

Generation of neurons in the rat dentate gyrus and hippocampus: effects of prenatal and postnatal treatment with ethanol.

Neurons in the rat hippocampal formation (the dentate gyrus and the hippocampus) are born over a protracted period, from gestational day (G) 15 into adulthood. Dentate gyral neurons born prenatally are generated from the ventricular zone, whereas those born postnatally are derived from a secondary proliferative zone, the intrahilar zone. In contrast, hippocampal pyramidal neurons are generated only prenatally from the ventricular zone. In the neocortex, ethanol depresses the proliferation of cells in the ventricular zone and stimulates the proliferation of cells in the secondary proliferative

Stage-dependent effects of ethanol on cranial neural crest cell development: partial basis for the phenotypic variations observed in fetal alcohol syndrome.

Fetal alcohol syndrome (FAS) is characterized by growth retardation, mental deficiencies, and numerous craniofacial and neuronal anomalies; the type and severity of these defects may be related to the time and dose of maternal ethanol exposure. Ethanol administered during presomitic stages results in the typical FAS craniofacial phenotype and is accompanied by a loss of cranial neural crest cells (CNCCs) through ethanol-induced cell death. However, the stage-specific effects of ethanol on the CNCC population is unknown. We examined the effects of ethanol on CNCC populations by treating in ovo

Male and female sexual behavior potential of male rats prenatally exposed to the influence of alcohol, stress, or both factors.

Adult sexual behaviors were characterized in male rats prenatally exposed to ethanol, stress, or ethanol combined with stress; 60% to 75% of each group exhibited female-typical lordosis. A substantial proportion of males subjected to alcohol (44%) or to alcohol with stress (54%) failed to ejaculate. The adult genitalia and testicular size appeared normal in all groups. Either alcohol or stress can suppress fetal plasma testosterone. Thus, exposing pregnant dams to alcohol, particularly in association with stress, may alter the hormonal milieu of their male fetuses sufficiently to block full ma

Role of noradrenergic innervation of brown adipose tissue in thermoregulatory deficits following prenatal alcohol exposure.

The development of thermoregulation in newborns is delayed by prenatal alcohol exposure in an animal model of moderate maternal drinking. Newborn mammals generate heat primarily via nonshivering thermogenesis in brown adipose tissue (BAT), which is activated by the sympathetic nervous system. In this study, the effects of prenatal alcohol exposure on the development of the sympathetic innervation of BAT was investigated by assessing the concentration of norepinephrine (NE) in interscapular BAT. Pregnant dams were given either a liquid diet with 35% of the calories derived from alcohol, a liqui

Effects of levamisole on ethanol-induced suppression of lactational immune transfer in rats.

Maternal ethanol consumption in rats has been shown to inhibit lactational transfer of immunity to Trichinella spiralis (T. spiralis) from dams to their neonates. The purpose of this study was to determine if this depressed immune transfer could be altered by treating the dams with a known immunostimulatory drug during pregnancy and lactation. Groups of female rats were fed ethanol-containing or were pair-fed isocaloric control liquid diets for 30 days, infected orally with 1,000 T. spiralis larva, and then continued on diet for 10 days to allow the adult worms to establish. The animals were p

Alcohol exposure in utero results in diminished T-cell function and alterations in brain corticotropin-releasing factor and ACTH content.

The long-term teratogenic effects of prenatal ethanol exposure during the last week of gestation on immune responsiveness and levels of pituitary ACTH and hypothalamic corticotropin-releasing factor (CRF) were examined in Sprague-Dawley rats. Immune responsiveness was measured by T-lymphocyte proliferation in response to mitogenic stimulation with Con A (3 micrograms/ml) in spleen and thymus cells of 21-old-day male rats who were exposed to alcohol in utero. The proliferative response was 8-fold lower in spleen and twofold lower in thymus cells from alcohol-exposed animals compared to response

Effects of prenatal alcohol exposure on behavior of aged rats.

Pregnant rats were intubated with alcohol (3.5 g/kg, twice daily) on gestation days 11-21. Control animals were intubated with an isocaloric sucrose solution and were pair-fed and pair-watered to alcohol-treated dams. At birth, offspring were placed with non-treated surrogate dams. When animals were slightly more than 1 year of age, they were tested for passive avoidance learning, spontaneous alternation and activity. Groups did not differ in passive avoidance learning or spontaneous alteration but animals prenatally exposed to alcohol were more active than controls. Additional studies showed

Gangliosides in axolemmal and synaptic membrane fractions from developing rats: effects of maternal ethanol consumption on offspring.

Previous work from this laboratory has shown that in utero exposure to ethanol significantly alters the synthesis of glycoproteins in synaptic, axolemmal, and myelin membranes from developing rats. In an attempt to determine whether in utero exposure to ethanol similarly alters the synthesis of other glycoconjugates involved with cell-cell interactions, the present study examined the influence of chronic maternal ethanol consumption prior to parturition on the content and synthesis of gangliosides in axolemmal and synaptic plasma membranes from developing rats. The results demonstrate that, in

Effects of Ethanol on the Cerebellum: Advances and Prospects.

Alcohol abuse causes cerebellar dysfunction and cerebellar ataxia is a common feature in alcoholics. Alcohol exposure during development also impacts the cerebellum. Children with fetal alcohol spectrum disorder (FASD) show many symptoms associated specifically with cerebellar deficits. However, the cellular and molecular mechanisms are unclear. This special issue discusses the most recent advances in the study of mechanisms underlying alcoholinduced cerebellar deficits. The alteration in GABAA receptor-dependent neurotransmission is a potential mechanism for ethanol-induced cerebellar dysfunc

The Impact of Paternal Alcohol Consumption on Offspring Brain and Behavioral Development.

BACKGROUND: Fetal alcohol spectrum disorders (FASD) describe the wide array of long-lasting developmental abnormalities in offspring due to prenatal alcohol (ethanol [EtOH]) exposure via maternal gestational drinking. Although the teratogenic consequences of prenatal EtOH exposure, are apparent, the effects of preconception paternal EtOH exposure (PatEE) are still unclear. Previous research suggests that PatEE can induce molecular changes and abnormal behavior in the offspring. However, it is not known whether PatEE impacts the development of the neocortex and behavior in offspring as demonstr…
