Blocking Dirofilaria immitis transmission by treatment of heartworm microfilaremic dogs with doxycycline.

BACKGROUND: This study was conducted to determine whether heartworm infective larvae (L3) from mosquitoes fed earlier on blood from dogs during daily administration of doxycycline at various dosages and treatment schedules and a single prophylactic dose of ivermectin could develop normally in dogs. METHODS: A total of 20 heartworm microfilaremic Beagles in a separate study allocated to four groups (n = 5/group) of five dogs were used as blood donors for mosquito infection. These donor dogs in groups 4, 5, and 6 had received doxycycline orally at 5.0, 7.5, or 10.0 mg/kg twice daily (BID) × 14

Evolution of PqsE as a Pseudomonas aeruginosa-specific regulator of LuxR-type receptors: insights from Pseudomonas and Burkholderia.

UNLABELLED: Pseudomonas aeruginosa is a Gram-negative opportunistic pathogen that poses a significant public health threat, particularly in healthcare settings. A key determinant of P. aeruginosa virulence is the regulated synthesis and release of extracellular products, which is controlled by a cell density-dependent signaling system known as quorum sensing (QS). P. aeruginosa uses a complex QS network, including two systems that rely on diffusible N-acylhomoserine lactone (AHL) signal molecules. The LuxR-type receptor RhlR is unique in that it requires not only its cognate AHL but also the a

Isolation and structure elucidation of a steroidal moiety from Withania somnifera and in silico evaluation of antimalarial efficacy against artemisinin resistance Plasmodium falciparum kelch 13 protein.

According to the 2021 Malaria report, 241 million clinical episodes with 627000 deaths penalty was estimated across the worldwide. However, mutation in the propeller domain of Plasmodium falciparum kelch 13 protein resulted in longer parasite clearance time following an artemisinin-based treatment and had a greater survival rate of ring-stage parasites even after a brief exposure to a high dose of artesunate. Clinical manifestations become more complex and worse with the emerging trend of drug resistance against artemisinin derivatives and the poor effectiveness of malaria vaccination drive. S

Beer ethanol and iso-α-acid level affect microbial community establishment and beer chemistry throughout wood maturation of beer.

Sour beers produced by barrel-aging of conventionally fermented beers are becoming increasingly popular. However, as the intricate interactions between the wood, the microbes and the beer are still unclear, wood maturation often leads to inconsistent end products with undesired sensory properties. Previous research on industrial barrel-aging of beer suggests that beer parameters like the ethanol content and bitterness play an important role in the microbial community composition and beer chemistry, but their exact impact still remains to be investigated. In this study, an experimentally tracta

A RNA-seq approach for exploring the protective effect of ginkgolide B on glutamate-induced astrocytes injury.

ETHNOPHARMACOLOGICAL RELEVANCE: There is substantial experimental evidence to support the view that Ginkgo biloba L. (Ginkgoaceae), a traditional Chinese medicine known to treating stroke, has a protective effect on the central nervous system and significantly improves the cognitive dysfunction caused by disease, including alzheimer disease (AD), vascular dementia, and diabetic encephalopathy. Although a number of studies have reported that ginkgolide B (GB), a diterpenoid lactone compound extracted from Ginkgo biloba leaves, has neuroprotective effects, very little research has been performed

Quorum Sensing Influences Burkholderia thailandensis Biofilm Development and Matrix Production.

UNLABELLED: Members of the genus Burkholderia are known to be adept at biofilm formation, which presumably assists in the survival of these organisms in the environment and the host. Biofilm formation has been linked to quorum sensing (QS) in several bacterial species. In this study, we characterized Burkholderia thailandensis biofilm development under flow conditions and sought to determine whether QS contributes to this process. B. thailandensis biofilm formation exhibited an unusual pattern: the cells formed small aggregates and then proceeded to produce mature biofilms characterized by "do

Induction of multiple pleiotropic drug resistance genes in yeast engineered to produce an increased level of anti-malarial drug precursor, artemisinic acid.

BACKGROUND: Due to the global occurrence of multi-drug-resistant malarial parasites (Plasmodium falciparum), the anti-malarial drug most effective against malaria is artemisinin, a natural product (sesquiterpene lactone endoperoxide) extracted from sweet wormwood (Artemisia annua). However, artemisinin is in short supply and unaffordable to most malaria patients. Artemisinin can be semi-synthesized from its precursor artemisinic acid, which can be synthesized from simple sugars using microorganisms genetically engineered with genes from A. annua. In order to develop an industrially competent y

Origin and evolution of genes specifying resistance to macrolide, lincosamide and streptogramin antibiotics: data and hypotheses.

Resistance to macrolide, lincosamide and streptogramin antibiotics is due to alteration of the target site or detoxification of the antibiotic. Postranscriptional methylation of 23S ribosomal rRNA confers resistance to macrolide (M), lincosamide (L) and streptogramin (S) B-type antibiotics, the so-called MLSB phenotype. Several classes of rRNA methylases conferring resistance to MLSB antibiotics have been characterized in Gram-positive cocci, in Bacillus spp, and in strains of actinomycetes producing erythromycin. The enzymes catalyze N6-dimethylation of an adenine residue situated in a highly

Modified QuEChERS for Organophosphorus Pesticide Analysis in Agricultural Soils by Gas Chromatography-Tandem Mass Spectrometry.

Accurate determination of organophosphorus pesticide (OPP) residues in agricultural soils by gas chromatography-tandem mass spectrometry (GC-MS/MS) is often hindered by matrix-induced signal effects caused by co-extracted organic matter, humic substances, and mineral-associated interferents. Conventional Quick-Easy-Cheap-Effective-Rugged-Safe (QuEChERS) cleanup strategies may not sufficiently remove matrix components from complex soil extracts, compromising analytical performance. This study presents a modified QuEChERS protocol that incorporates the adsorbent into mixed-sorbent dispersive sol

Drug repositioning as an antivirulence strategy: Ketoprofen targets quorum sensing and biofilm in Serratia sp.

Serratia sp. are opportunistic Gram-negative bacteria capable of forming robust biofilms and expressing a wide range of quorum sensing (QS)-regulated virulence factors, including motility, protease secretion, and prodigiosin production. The rise of multidrug-resistant strains has emphasized the urgent need for alternative therapeutic strategies targeting bacterial virulence rather than viability. In this context, nonsteroidal anti-inflammatory drugs (NSAIDs) such as ketoprofen have emerged as potential quorum-quenching agents. This study explores the antivirulence activity of ketoprofen agains
