Methicillin-resistant Staphylococcus aureus (MRSA) is a major global health threat because of its ability to adapt. In North America, the USA300 lineage ST8 has become the predominant MRSA clone, whereas the ST72 lineage has emerged as an important MRSA in East Asia. Here, we compare USA300 and SAWL001 from the ST72 at the phenotypic, genomic, and transcriptomic levels. Phenotypic assays assessed antibiotic susceptibility, intracellular invasion, oxidative stress survival, biofilm formation under β-lactam exposure, blood-induced cell clumping, persister formation, and virulence in a mouse seps

UNLABELLED: Clinical microbiology laboratories are increasingly adopting automation to meet testing demands, improve throughput, and accelerate diagnostic turnaround. This study evaluated optimal culture read times using Copan WaspLab microbiology laboratory automation (MLA) to determine if earlier read times affect accuracy and turnaround times as they pertain to microbial identification and antimicrobial susceptibility testing (AST). A total of 374 patient specimens were cultured simultaneously using a conventional incubation method and MLA, with MLA images captured from 10 to 72 h of incuba

Ticks and tick-borne pathogens are expanding their geographic ranges to novel suitable habitats. Together with the World Health Organization (WHO) and the United Nations (UN), Saudi Arabia's government has joined efforts to prevent the development of tick-associated pathogens. Here, we investigated the prevalence and diversity of antimicrobial-resistant (AMR) bacteria in ticks parasitizing camels in Al-Jouf province. A total of 60 ticks were sampled and identified as Hyalomma excavatum (n = 41) and Hyalomma dromedarii (n = 19), infesting 11 camels. Altogether 70 bacterial isolates were isolate

BACKGROUND: Enterococcal infections represent 10% of intensive care unit (ICU)-acquired infections and are associated with adverse outcomes, particularly in the case of E. faecium infections. Some studies focusing on non-critically ill patients have cast doubt on the non-inferiority of daptomycin compared to other antibiotics for these infections. We aimed to describe antibiotic treatment practices for monomicrobial E. faecium bloodstream infections (BSI) in patients admitted to the ICU, and to identify factors associated with treatment failure.
METHODS: This retrospective multicenter study i

Background Neonatal sepsis is a major cause of mortality in low- and middle-income countries (LMICs) such as India, where delayed diagnosis and antimicrobial resistance (AMR) challenge effective management. Conventional blood cultures, taking 2-5 days for pathogen identification, delay targeted therapy, worsening outcomes, and fueling AMR. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) (bioMérieux, USA) offers rapid diagnostics, potentially transforming neonatal sepsis care. This study assesses the diagnostic speed of MALDI-TOF MS versus conventiona

BACKGROUND: Staphylococcus aureus (S. aureus) was a prevalent pathogenic bacterium among children. Due to the extensive use of antibiotics, the sensitivity of S. aureus to these drugs has gradually declined. Since the 1960s, methicillin-resistant Staphylococcus aureus (MRSA) has emerged and spread worldwide, becoming a primary cause of both healthcare-associated (HA) and community-acquired (CA) infections. This retrospective study aimed to highlight the significance of S. aureus among bacteria isolated from children in Beijing, China, and to elucidate its antimicrobial resistance patterns.

INTRODUCTION: Non-aureus Staphylococcus (NAS) species are currently the most commonly identified microbial agents causing sub-clinical infections of the udder and are also deemed as opportunistic pathogens of clinical mastitis in dairy cattle. More than 10 NAS species have been identified and studied but little is known about S. haemolyticus in accordance with dairy mastitis. The present study focused on the molecular epidemiology and genotypic characterization of S. haemolyticus isolated from dairy cattle milk in Northwest, China.
METHODS: In this study, a total of 356 milk samples were coll

Enterococci, especially Enterococcus faecium, are one of today's leading causes of multidrug-resistant infections in hospital settings. The marine environment may harbour enterococci, but its role as an evolutionary niche and as a vector for the spread of enterococci is sparsely investigated. Hence, by applying enterococci in bivalves as a sentinel tool, this study aimed to describe the prevalence of enterocooci along the Norwegian coast and in addition the phylogeny of E. faecium in particular. Enterococci in batch samples of marine bivalves, harvested from 86 different locations, were quanti

BACKGROUND: Mycobacterium chimaera is a slowly growing non-tuberculous mycobacterium associated with outbreaks of fatal infections in patients after cardiac surgery, and it is increasingly being detected in patients with chronic lung conditions. M chimaera can cause disseminated disease, osteomyelitis, and chronic skin or soft-tissue infections. We aimed to find new inhibitory compounds and drug repurposing opportunities for M chimaera, as current therapeutic options often result in poor outcomes.
METHODS: In an open drug discovery approach, we screened the Medicines for Malaria Venture (MMV)

Enterococci, which are commonly found in the environment, cause serious infections despite the absence of well-defined virulence factors and toxins. Knowing the virulence properties of enterococci is important to understand the complex pathogenic structures. In this study, we aimed to investigate the virulence factors (asa1, hyl, cylA, efa, ebp, ace, esp, gelE, sprE, fsrA, fsrB, fsrC genes, gelatinase activity, hemolysin, hydrogen peroxide and biofilm production) and antibiotic resistance of Enterococcus faecium and Enterococcus faecalis strains isolated from clinical specimens. Central nervous system (CNS) infections are associated with high mortality rates. The clinical presentation of many CNS infections by different pathogens is difficult to distinguish, but the definite diagnosis of the etiology is critical for effective therapy and prognosis. The aim of this study was to explore the etiology of CNS infections with definite diagnoses based on data from a clinical microbiology laboratory in Tongji Hospital, a teaching hospital in China, obtained over a six-year period. We conducted a retrospective study on all cerebrospinal fluid (CSF) specimens submitted to our c

BACKGROUND AND OBJECTIVES: Neonatal septicemia can be rapidly fatal if not treated promptly. A speedy laboratory diagnosis would improve the outcome. The BacT/ALERT 3D system (bioMerieux, Durham, North Carolina) is currently being used for laboratory diagnosis of blood stream infections. In the present study, a modified protocol was employed in which the broth was subcultured into two nutrient broth tubes and these tubes were used for biochemical tests and antimicrobial susceptibility testing to decrease the turnaround time.
MATERIALS AND METHODS: A prospective study was conducted in the Depa

Background: Methicillin-resistant Staphylococcus aureus (MRSA) colonization is a risk factor for potential staphylococcal infection and outbreaks. Although it is recommended to obtain a swab culture to detect nasal colonization its necessity in low-prevalence countries is debated. The aim of this study was to determine the prevalence of MRSA nasal colonization, the rate of invasive infection development, and the risk factors for invasive infections in patients admitted to the intensive care unit. Materials and Methods: This retrospective study included patients who were followed up in one of t
