Biofilm-host immunometabolic crosstalk in Staphylococcus aureus orthopedic implant infection: mechanisms and translational constraints.

Orthopedic implant biofilms are not only antibiotic-tolerant bacterial communities but metabolically active niches that reshape host immunity. Compared with planktonic infection, mature biofilms impose spatial and metabolic constraints that can uncouple leukocyte recruitment from sterilizing activity. Neutrophils can penetrate and phagocytose portions of Staphylococcus aureus biofilms, but clearance becomes incomplete as biofilms mature and may coexist with degranulation and neutrophil extracellular trap-associated tissue injury. Macrophages show impaired access and bactericidal programming, a

PARP inhibitors and the tumor micro-environment: Bidirectional crosstalk, resistance mechanisms, and therapeutic opportunities.

Poly (ADP-ribose) polymerase inhibitors (PARPi) have revolutionized in precision oncology, particularly in tumors harboring homologous recombination deficiency (HRD), such asovarian and breast cancers. Despite their clinical success, both primary and acquired resistance substantially limit the durability and breadth of therapeutic benefit, indicating that tumor-intrinsic DNA repair defects alone are insufficient to fully explain treatment outcomes. Accumulating evidence highlights the tumor microenvironment (TME) as a critical determinant of PARPi sensitivity and resistance. This review provid

Dose-dependent effects of dimethyl sulfoxide (DMSO; Me2SO) for platelet cryopreservation: 10% Me2SO provides superior cryoprotection.

The most widely used method of platelet cryopreservation involves the addition of 5-6% dimethyl sulfoxide (DMSO; Me2SO), followed by pre-freeze centrifugation and hyperconcentration to minimize the content of Me2SO transfused. However, several aspects of platelet quality are negatively affected by cryopreservation, indicating the process could be optimized. The aim of this study was to identify the concentration of Me2SO that provides maximal cryoprotection for platelets. Scaled-down titration experiments were performed in tubes. Me2SO (5% to 20% v/v) was added to apheresis platelets, followed

Metabolite-Mediated Crosstalk Between Neutrophils, Commensal Bacteria, and Dendritic Cells Is Associated with Intestinal Immune Responses.

The intestinal immune system relies on coordinated interactions between innate immune cells and commensal bacteria. How metabolites integrate host-microbe communication in the gut remains incompletely understood. Using neutrophil activation assays, bacterial chemotaxis analysis, short-chain fatty acid profiling, and imaging of inflamed intestinal tissue, we examined how neutrophil-derived metabolites regulate commensal behavior and immune cell function. Activated neutrophils released lactate, which was associated with selective chemotaxis of commensal Escherichia coli but not pathogenic Acinet

Identification of NETs and inflammation-related prognostic genes in breast cancer and PCR experimental validation.

BACKGROUND: Neutrophil extracellular trap-driven necrosis-related genes (NRGs) and inflammation-related genes (IRGs) are crucial in mitigating or inhibiting cancer progression in breast cancer patients. The interaction between NRGs and IRGs in breast cancer remains unclear. Thus, identifying prognostic genes associated with NETosis and inflammation in breast cancer may offer a novel approach to improving the outcome of breast cancer. METHODS: Genes with statistically significant expression levels and correlations with IRGs and NRGs scores were selected as candidate genes, and the protein-prot

Exploring the mechanism of polymorphonuclear neutrophils against sepsis based on immune model.

BACKGROUND: Sepsis is a life-threatening organ dysfunction caused by a dysregulated host response to infection and remains a major global health challenge. Polymorphonuclear neutrophils (PMNs), as major effectors of innate immunity, are essential for antimicrobial defense but can also contribute to immune dysregulation, tissue injury, and organ failure during sepsis. METHODS: We conducted a narrative review of the literature by searching PubMed and Web of Science from database inception to November 2025. Search terms included sepsis, septic shock, neutrophils, polymorphonuclear neutrophils, P

The shared biomarkers and molecular mechanisms of systemic lupus erythematosus and type 2 diabetes.

Systemic lupus erythematosus (SLE) and type 2 diabetes mellitus (T2DM) share inflammatory and metabolic disturbances, yet the molecular mechanism of their overlap remains unclear. This study used integrated bioinformatics to identify transcriptomic signatures and potential biomarkers common to both conditions. Gene expression profiles from publicly available datasets (SLE: 38 patients/32 controls; T2DM: 6 patients/6 controls; validation cohorts: 79/30 and 41/15, respectively) were analyzed to detect shared differentially expressed genes and co-expression modules. Impact of dipeptidyl peptidase I and neutrophil serine proteases on neutrophil functional responses.

Dipeptidyl peptidase-1 (DPP1) is a lysosomal cysteine protease essential for activating neutrophil serine proteases (NSPs), including neutrophil elastase, cathepsin G, and proteinase 3, during neutrophil differentiation in the bone marrow. Because NSP-mediated tissue damage contributes to chronic inflammatory and autoimmune diseases, targeting NSPs has emerged as a therapeutic strategy. In this regard, small molecule drugs have been developed such as brensocatib-a competitive, reversible DPP1 inhibitor. Active DPP1 is expressed in mature neutrophils, which raises the possibility that DPP1 inhi

An overview of current research on exercise interventions in aging and aging-related disease.

Global declines in physical activity have contributed to an acceleration in immune aging, characterized by systemic inflammation (inflammaging) and impaired immune regulation (immunosenescence). This narrative review provides an overview of the evidence in both preclinical and clinical models supporting exercise as a critical intervention to counteract immune aging and its related diseases. Regular physical activity modulates systemic inflammation, reduces neutrophil extracellular trap (NET) formation, and promotes favorable shifts in immune cell populations, including T cell and natural kille

Giant cell myocarditis: from immune pathogenesis to contemporary management.

Giant cell myocarditis (GCM) is a rare but devastating inflammatory cardiac disease characterized by rapid hemodynamic deterioration, malignant arrhythmias, and high rates of death or heart transplantation. Without immunosuppressive treatment, median transplant-free survival is approximately three months. Current evidence identifies GCM as a predominantly T cell-mediated autoimmune disorder driven by a breakdown of immune tolerance, with contributions from macrophage-derived multinucleated giant cells and neutrophil extracellular trap formation. Viral infections and environmental factors have

Empagliflozin in GSD-Ib: Long-term safety and sustained recovery of neutrophil function including NET formation.

Glycogen storage disease type Ib (GSD-Ib) engenders neutropenia and severe neutrophil dysfunction, leading to recurrent infections and inflammatory complications. Recent studies have identified intracellular accumulation of 1,5-anhydroglucitol-6-phosphate (1,5-AG6P) as a key mechanism underlying neutrophil impairment and have suggested therapeutic benefits of sodium-glucose cotransporter 2 (SGLT2) inhibitors, which lower plasma levels of its precursor 1,5-AG. In this study, we performed a four-year longitudinal evaluation of empagliflozin therapy in a genetically confirmed GSD-Ib infant, contr

Stage-specific elevation of MPO and NE mRNA expression in diabetic kidney disease and their independent associations with albuminuria.

BACKGROUND: Myeloperoxidase (MPO) and neutrophil elastase (NE) are key neutrophil enzymes involved in oxidative stress, inflammation, and neutrophil extracellular trap formation, all of which have been implicated in the pathogenesis of diabetic kidney disease (DKD). However, MPO and NE mRNA expression in peripheral blood leukocytes (PBL) across DKD stages remains insufficiently characterized. This study evaluates the association between DKD severity and MPO and NE mRNA expression in PBL. METHODS AND RESULTS: Seventy patients with type 2 diabetes were categorized into A1 DKD (normoalbuminuria)

Transcriptomic Profiling of Orbital Fat Tissue and Ocular Surface Wash in Active Thyroid Eye Disease Requiring Urgent Orbital Decompression.

PURPOSE: Thyroid eye disease (TED) is an autoimmune disorder characterized by orbital inflammation and tissue remodeling. Although most patients with active TED respond to medical therapy, a subset develops sight-threatening complications which require urgent orbital decompression when conservative treatment fails. This study aimed to elucidate the molecular mechanisms underlying active, moderate-to-severe TED in patients requiring urgent orbital decompression during the active disease stage. METHODS: Transcriptomic profiling was performed on retro-orbital fat samples from 13 patients and ocu

Neuroprotective, anti-inflammatory, and analgesic activity of palmitoylethanolamide in sickle cell mice.

Chronic and acute pain are major comorbidities of sickle cell disease (SCD). Both peripheral and central nervous system mechanisms underlie SCD pain. We investigated the potential roles of endogenous fatty acyl derivatives with endocannabinoid-like activity; using targeted lipidomics of fatty acid ethanolamides and monoacylglycerols, we observed significant reduction of spinal palmitoylethanolamide (PEA) in the humanized, transgenic sickle (HbSS)-BERK mouse model of SCD when compared with healthy HbAA control mice. PEA is a paracannabinoid lipid mediator that acts as a putative mast cell stabi

Regenerative potential of PRP-based scaffolds in chronic wound healing: Mechanisms, advances, and therapeutic insights.

INTRODUCTION: Chronic wounds such as diabetic foot ulcers, venous leg ulcers, and pressure ulcers often remain trapped in the inflammatory phase due to oxidative stress, protease overactivity, and impaired cellular responses, particularly in diabetic conditions. These wounds require advanced therapeutic strategies beyond conventional care. Regenerative medicine-especially platelet-rich plasma (PRP)-based interventions-has emerged as a promising approach for enhancing wound repair. METHODS: This review examines recent developments in PRP-loaded scaffolds, focusing on their biological mechanism

Neutrophil-to-lymphocyte ratio predicts a poor prognosis for penile cancer with an immunosuppressive tumor microenvironment.

BACKGROUND: Chronic inflammation related to poor genital hygiene is a well-recognized pathogenic trigger for penile cancer (PC). The neutrophil-to-lymphocyte ratio (NLR) is a simple, reproducible systemic inflammatory marker and has been reported to indicate unfavorable outcomes. However, previous studies were limited by small sample sizes, confounding prognostic factors and a lack of high-quality evidence demonstrating the significance of the NLR in PC. METHODS: A large multicenter cohort of 582 PC patients who underwent radical inguinal lymphadenectomy with definitive pN stage information w
