Fetal alcohol syndrome, Tourette syndrome, and hyperactivity in nine adopted children.

Much attention has been paid in recent years to the role of prenatal exposure to alcohol. Fetal alcohol syndrome is one of the most severe afflictions resulting from such exposure. The present report documents the cases of adopted children diagnosed with fetal alcohol syndrome who developed both Tourette syndrome and attention deficit-hyperactivity disorder. Out of a population of 138 adopted children with behavior issues whose clinical histories were reviewed retrospectively, 9 children (6.5%) presented this constellation. Epidemiologic data, clinical data, neurologic examination findings, co

Measuring the facial phenotype of individuals with prenatal alcohol exposure: correlations with brain dysfunction.

The purpose of this report is to demonstrate how to measure the magnitude of expression of the fetal alcohol syndrome (FAS) facial phenotype using the new 4-Digit Diagnostic Code and the previously developed D-score and to demonstrate how these two measures of the FAS facial phenotype correlate with brain function and structure; correlations that fail to be identified by the older gestalt method of facial measurement. The D-score and the facial component of the 4-Digit Diagnostic Code quantitatively measure the magnitude of expression of the FAS facial phenotype using three facial features (pa

An e-health transition intervention for youth with brain-based disabilities: Pilot and feasibility results from a Randomized Controlled Trial.

BACKGROUND: Youth with brain-based disabilities (BBD) need tailored support when preparing for adult healthcare. We originally designed a full randomized controlled trial (RCT) to test whether the MyREADY Transition™ BBD App improved transition readiness among youth aged 15-17 years with autism spectrum disorder, cerebral palsy, epilepsy, fetal alcohol spectrum disorder, or spina bifida. Due to slow recruitment, the full RCT was halted and pivoted to a stand-alone pilot and feasibility trial. METHODS: This mixed method, patient-oriented pilot RCT (2019-2022, Canada) evaluated process, resourc

Altered Functional Connectivity Observed at Rest in Children and Adolescents Prenatally Exposed to Alcohol.

Studies of brain structure in fetal alcohol spectrum disorder (FASD) have shown the global and focal effects that prenatal alcohol exposure (PAE) has on the brain, suggesting but not measuring altered function in FASD. This study aimed to (1) identify resting-state functional networks in children and adolescents with FASD, (2) investigate functional connectivity differences compared with healthy controls, and (3) assess the links to cognitive deficits. Participants included 66 children/adolescents with FASD (aged 5.5-18.9 years) and 67 healthy controls (aged 5.8-18.5 years) scanned across four

Maternal drug or alcohol abuse is associated with decreased head size from mid-pregnancy to childhood.

AIM: Maternal alcohol abuse is poorly recognised and causes developmental problems. This study explored the foetal central nervous systems (CNS), head circumference and psychomotor development of children exposed to drugs or alcohol during pregnancy up to 2.5 years of age. METHODS: We recruited 23 pregnant women referred to Kuopio University Hospital, Finland, by their family doctor because of drug or alcohol abuse, and 22 control mothers. Foetal CNS parameters were measured by three-dimensional ultrasonography at the mean gestational age of 20 weeks and the Griffiths Mental Developmental Sca

The effects of prenatal exposure to alcohol and environmental tobacco smoke on risk for ADHD: a large population-based study.

Attention deficit hyperactivity disorder (ADHD) is caused by the interaction of genetic and environmental factors. The objective of this study was to examine the effects of prenatal exposure to alcohol and environmental tobacco smoke (ETS). Among the 30,552 parents who responded to a survey, the answers of 19,940 who replied to questions on prenatal exposure to ETS, alcohol consumption, and completed the DuPaul Rating Scale were analyzed. Results revealed that risk of ADHD significantly increased as a result of exposure to alcohol by 1.55 times (95% CI 1.33-1.82), maternal smoking during pregn

[Low birth weight--additional important factor of diagnosis in children with short stature].

INTRODUCTION: Birth parameters are one of the more important predictors of body height in adult life. Children with low birth weight (LBW) are an essential and heterogenic group of patients diagnosed because of short stature. AIM OF THE STUDY: Evaluation of frequency of low birth mass in children with short stature and evaluation of anthropometric parameters in children with low and normal birth weight. MATERIAL AND METHODS: Analysis of birth data and present somatic parameters in 802 children with short stature who were admitted to the Clinic of Paediatrics and Endocrinology. This group con

Ethanol exposure induces upregulation of specific microRNAs in zebrafish embryos.

Prenatal exposure to ethanol leads to a myriad of developmental disorders known as fetal alcohol spectrum disorder, often characterized by growth and mental retardation, central nervous system damage, and specific craniofacial dysmorphic features. The mechanisms of ethanol toxicity are not fully understood, but exposure during development affects the expression of several genes involved in cell cycle control, apoptosis, and transcriptional regulation. MicroRNAs (miRNAs) are implicated in some of these processes, however, it is not yet clear if they are involved in ethanol-induced toxicity. In

Effect of pre-treatment with aspirin on alcohol-induced neural tube defects in the TO mouse fetuses.

This study was planned to investigate the effects of ethanol on the genetic susceptibility of the TO mouse to neural tube malformations and to determine the ameliorative effects, if any, of aspirin a potent prostaglandin inhibitor. The TO mouse exhibits a spontaneous incidence of 3.6% exencephaly. The mice were exposed to single (i.p.) dose of 0.03 ml/gm body weight of a solution of (25%v/v) of absolute alcohol in physiological saline on day 7 or 8 of gestation. Subteratogenic doses (150 or 200 mg/kg) of aspirin were administered (i.p.) an hour before ethanol exposure. Fetuses were collected o

Binge drinking during pregnancy and risk of seizures in childhood: a study based on the Danish National Birth Cohort.

Seizures are often found in children with fetal alcohol syndrome, but it is not known whether binge drinking during pregnancy by nonalcoholic women is associated with an increased risk of seizure disorders in children. The authors conducted a population-based cohort study of 80,526 liveborn singletons in the Danish National Birth Cohort (1996-2002). Information on maternal binge drinking (intake of > or = 5 drinks on a single occasion) was collected in 2 computer-assisted telephone interviews during pregnancy. Children were followed for up to 8 years. Information on neonatal seizures, epilepsy

Prenatal ethanol exposure increases ethanol reinforcement in neonatal rats.

BACKGROUND: Recent experimental findings indicate that newborn rats are quite susceptible to the reinforcing properties of ethanol (EtOH). The basis seems to be more pharmacological than gustatory, olfactory, or caloric as a single pairing of a surrogate nipple providing water with an intraperitoneal (i.p.) injection of EtOH results in conditioned attachment to an empty surrogate nipple. The present study tested whether prenatal exposure to moderate EtOH during late gestation would alter the neonatal rat's susceptibility to this source of EtOH reinforcement and/or EtOH metabolism. METHODS: Pr

Brief intervention for prenatal alcohol use: the role of drinking goal selection.

The behavioral problems and cognitive deficits resulting from even small amounts of prenatal alcohol exposure can be significant and enduring but completely preventable. The purpose of this study was to examine the impact of a prenatal drinking goal selected during a brief intervention for 115 pregnant women and their partners on subsequent consumption. Higher proportions of women having their first pregnancy chose abstinence as a goal over drinking reduction. Goal selection was highly predictive of subsequent drinking behavior. Interestingly, the participants who were abstinent at enrollment

Neuroanatomical and neurophysiological mechanisms involved in central nervous system dysfunctions induced by prenatal alcohol exposure.

One of the most severe consequences of maternal ethanol consumption is the damage to the developing central nervous system, which is manifested by long-term cognitive and behavioral deficits in the offspring. Prenatal exposure to ethanol affects many crucial neurochemical and cellular components of the developing brain. Ethanol interferes with all of the stages of brain development, and the severity of the damage depends on the amount of ethanol intake and level of exposure. Experimental observations also indicate that the toxic effects of ethanol are not uniform: some brain regions are more a

Prenatal exposure of ethanol induces increased glutamatergic neuronal differentiation of neural progenitor cells.

BACKGROUND: Prenatal ethanol exposure during pregnancy induces a spectrum of mental and physical disorders called fetal alcohol spectrum disorder (FASD). The central nervous system is the main organ influenced by FASD, and neurological symptoms include mental retardation, learning abnormalities, hyperactivity and seizure susceptibility in childhood along with the microcephaly. In this study, we examined whether ethanol exposure adversely affects the proliferation of NPC and de-regulates the normal ratio between glutamatergic and GABAergic neuronal differentiation using primary neural progenito

[Consequences of tobacco, cocaine and cannabis consumption during pregnancy on the pregnancy itself, on the newborn and on child development: A review].

BACKGROUND: Substance use has increased worldwide. Based on these data, we may think that substance use has also increased during pregnancy, but epidemiological data are scarce in this population. The potential consequences of tobacco, cocaine or cannabis use during pregnancy are a major public health concern. The combined use of different substances during pregnancy may have serious consequences on the pregnancy and on child development. METHODS: In this paper, we will describe the potential consequences for the newborn, child and adolescent after being exposed to tobacco, cannabis and cocai
