Gene therapy via canalostomy approach preserves auditory and vestibular functions in a mouse model of Jervell and Lange-Nielsen syndrome type 2.

Mutations in voltage-gated potassium channel KCNE1 cause Jervell and Lange-Nielsen syndrome type 2 (JLNS2), resulting in congenital deafness and vestibular dysfunction. We conducted gene therapy by injecting viral vectors using the canalostomy approach in Kcne1-/- mice to treat both the hearing and vestibular symptoms. Results showed early treatment prevented collapse of the Reissner's membrane and vestibular wall, retained the normal size of the semicircular canals, and prevented the degeneration of inner ear cells. In a dose-dependent manner, the treatment preserved auditory (16 out of 20 mi

Subjective body vertical: a promising diagnostic tool in idiopathic normal pressure hydrocephalus?

Postural instability is a frequent symptom of patients with idiopathic normal pressure hydrocephalus (iNPH), and might be due to the misperception of body verticality. The objective of this study was to assess the usefulness of the subjective body vertical (SBV) as a potential tool for diagnosing iNPH. Twenty patients with iNPH underwent tests of SBV in the pitch and roll planes before and after cerebrospinal fluid (CSF) drainage. Ten patients with other central gait disorders served as controls and also underwent tests for SBV. Before CSF drainage, patients with iNPH showed an impaired vertic

Lower extremity joint position sense in runners with and without a history of knee overuse injury.

Kinematic and kinetic analyses are routinely implemented to determine if gait differences exist between runners with and without a history of knee injury. Hip and knee kinematic differences have been reported between knee injured and non-injured runners. Yet, there is no consensus on whether these differences are the primary variables contributing to knee injury. Furthermore, there may be additional underlying factors that contribute to the development of injury that cannot be determined by gait analysis. The purpose of this investigation was to determine if joint position sense differences ex

Upper limb motor strategies in persons with and without shoulder impingement syndrome across different speeds of movement.

BACKGROUND: Activities requiring repetitive arm movements, including high velocity actions, have been identified as a risk factor for shoulder impingement. However, the effect of speed on upper limb motor strategies has yet to be evaluated for individuals with shoulder impingement. The aims of this study were to characterize upper limb motor strategies in individuals with and without shoulder impingement during reaching at natural speed and to evaluate their adaptation to higher speeds of movement. METHODS: Twenty healthy individuals and 33 individuals with shoulder impingement took part in o

What to expect from immersive virtual environment exposure: influences of gender, body mass index, and past experience.

For those interested in using head-coupled PC-based immersive virtual environment (VE) technology to train, entertain, or inform, it is essential to understand the effects this technology has on its users. This study investigated potential adverse effects, including the sickness associated with exposure and extreme responses (emesis, flashbacks). Participants were exposed to a VE for 15 to 60 min, with either complete or streamlined navigational control and simple or complex scenes, after which time measures of sickness were obtained. More than 80% of participants experienced nausea, oculomoto

Motor inhibition and excitation are independent effects of magnetic cortical stimulation.

We administered magnetic cortical stimulation (MCS) during voluntary contraction of intrinsic hand muscles to 8 patients with motor neuron disease (MND), 5 patients with pure lower motor neuron syndromes (LMN), a patient with severe subacute sensory neuropathy (SSN), and 10 healthy volunteers. Patients with MND had clinical evidence of upper MND and elevated thresholds for (3 patients) or absence of (5 patients) motor evoked potentials (MEPs). MCS during sustained contraction inhibited electromyographic activity in 6 of 8 patients with MND, without preceding MEPs. MCS had no effect on the elec

Simulation of Space-Adaptation Syndrome on Earth.

The three Spacelab D-1 Scientist Astronauts were exposed to a 1.5 h centrifuge run in the supine position, resulting in a linear 3 g acceleration. They used their space experience to evaluate their readaptation to normal gravity and compared their observations with 'Space Adaptation Syndrome'. After the centrifuge runs, the vestibular visual system appeared to be modified in a very specific and reproducible manner. Readaptation to the normal 1 g environment took at least 6 h. During this period there was a striking similarity to the astronauts' experience during adaptation to weightlessness in

Electromyographic patterns and masticatory muscles activity changes in patients with dentoskeletal deformities before and after orthognathic surgery: a pilot study.

PURPOSE: Orthognathic surgery involves repositioning the skeletal bases to correct dentoskeletal deformities and malocclusions. While its impact on occlusion is well documented, its effects on masticatory muscle activity remain under investigation. This pilot study evaluates changes in static electromyographic (sEMG) patterns of masseter and temporal muscles, and explores correlations with TMJ-related muscular symptoms before and after surgery. METHODS: A surface electromyography (sEMG), by employing TeethanTM, based on occlusal contacts, was performed recording the activation patterns of mas

Muscle-Derived BMP4 Regulates Morphology and Function of Endplates on Extrafusal and Intrafusal Muscle Fibers in Adult Mice.

Understanding factors contributing to neuromuscular junction (NMJ) stability postdevelopment will shed light on how this stability is lost during aging and in neuromuscular diseases. Previous work in Drosophila suggests that morphogens within the bone morphogenic protein (BMP) family are potential candidates because the BMP homolog, gbb, along with its receptor, wit, have key roles in NMJ structure, stability, and function. Whether BMPs have similar roles at vertebrate NMJs is currently unknown. To examine this question, we generated doxycycline-inducible, muscle-specific BMP4 null mice, refer

Effects of Focal Muscle Vibration on Static and Dynamic Balance in Patients with Parkinson's Disease: Preliminary Results of a Retrospective Study.

Background and Objectives: Postural instability is a key feature of Parkinson's disease (PD), contributing to disability and increased risk of falls. Pharmacological treatments are important, but it is necessary to integrate them with rehabilitation programs that provide benefits for gait and balance. Focal muscle vibration (fMV) has been proposed as a proprioceptive-oriented intervention to enhance postural control, but evidence in PD remains heterogeneous. This observational, retrospective, and controlled pilot study aimed to evaluate whether the integration of fMV into a standardized rehabi

Peripheral anatomy of the dolphin ear and associated nervous structures: Insights from macroscopic dissection, DICE-µCT, histology, and confocal microscopy.

The peripheral auditory system of dolphins comprises specialised bony, fatty, vascular, and neural structures adapted for underwater hearing and diving physiology. These include the external ear canal, acoustic fat bodies, sinuses, and associated neurovascular networks, which together support sound conduction, protection, and possibly sensory functions. Despite advances in gross anatomical description, the detailed integration of these tissues, particularly the innervation, neurovascular organisation, and their functional implications, remains poorly understood. Previous studies have described

Mapping peripheral misreaching in a large cohort of stroke patients.

Peripheral reaching to objects outside the direct line of sight supports motor flexibility. Deficits in this ability are the hallmark of optic ataxia, yet impairments may exist along a continuum and warrant investigation in larger, unselected samples. This study examined peripheral misreaching and its neuroanatomical correlates in stroke patients. Peripheral reaching was assessed using a standardized reach-to-grasp task in 86 ischemic stroke patients and 22 mean age-matched controls. Group-level analyses assessed effects of target location and reaching hand, and case-control comparisons quanti

A Non-surgical Multimodal Approach to Adolescent Idiopathic Scoliosis (Lenke 5C) Using an Intensive Two-Week CLEAR Institute Protocol: A Report of Two Cases.

Adolescent idiopathic scoliosis (AIS) is a progressive three-dimensional spinal deformity often managed with physiotherapeutic scoliosis-specific exercises (PSSE), bracing, or spinal fusion. Emerging evidence suggests that multimodal, high-frequency non-surgical rehabilitation may produce measurable improvements in spinal alignment through combined biomechanical loading and neuromuscular re-education. This case series evaluates radiographic and functional outcomes in two AIS subjects (Lenke 5C) undergoing a two-week intensive CLEAR protocol followed by ScoliBrace® implementation. Two female ad

Current status and future prospects of brain-computer interfaces in the field of neurological disease rehabilitation.

Neurological disorders represent a significant category of diseases that profoundly affect human health, accounting for the second leading cause of global mortality. This group of conditions includes stroke, multiple sclerosis (MS), amyotrophic lateral sclerosis (ALS), spinal cord injury, Parkinson's disease, and cerebral palsy, among others. These disorders are highly susceptible to sequelae and profoundly impact individuals' daily lives. In this context, Brain-Computer Interface (BCI) technology has demonstrated considerable potential in the domain of neurorehabilitation, although numerous c

Post-inhibitory rebound excitation drives extensor activity following spinal cord stimulation.

BACKGROUND: Neural circuits throughout the CNS can exhibit rebound excitation following prolonged periods of inhibition. However, the potential to control this phenomenon and harness it for clinical applications remains largely unexplored. OBJECTIVE: We investigate rebound excitatory responses evoked by spinal cord stimulation (SCS) that can generate functional motor output, providing a testable model of circuit-level rebound excitation relevant to neuromodulation across the CNS. METHODS: Brief (5 s) electrical stimulation trains were delivered to the lumbar spinal cord in adult cats. We rec
