Bioinformatics analysis identifies IL-23A and JAK2 as regulatory factors in sepsis immune response via the JAK/STAT3 pathway.

INTRODUCTION: Sepsis is a life-threatening organ dysfunction caused by dysregulated host responses to infection. Identifying key genes associated with sepsis and exploring their interactions with immune cells are crucial for advancing diagnostic and therapeutic strategies. METHODOLOGY: In order to explore the genetic underpinnings, five datasets-GSE28750, GSE57065, GSE64457, GSE65682, and GSE95233-were analyzed using the "Limma" package in R to identify differentially expressed genes (DEGs). Functional enrichment analysis was conducted using Gene Ontology (GO) and Kyoto Encyclopedia of Genes

Network-based prediction and real-world patient data observation identify doxycycline as a repurposable drug in Alzheimer's disease.

Alzheimer's disease (AD), a leading global cause of dementia, disability, and mortality, represents a critical unmet need for effective therapeutic interventions. Drug repurposing offers an expedited pathway to address this challenge compared to traditional drug development. Here, we leveraged network-based prediction and real-world patient data validation, a comprehensive strategy to identify unanticipated therapeutic applications for existing medications. Traumatic brain injury (TBI), a major risk factor for earlier and more severe AD, exhibits heterogenous clinical outcomes influenced by ge

QSAR, molecular dynamics, and biological evaluation of novel myeloperoxidase inhibitors via ligand-based pharmacophore modeling as potential anticancer agents.

Myeloperoxidase (MPO) has shown promise as a therapeutic target due to its critical role in inflammatory mechanisms and cancer progression. Despite extensive research on MPO inhibitors, the lack of integrated computational-experimental workflows constrains the efficient identification and validation of biologically relevant candidates. Hence, in this study, a ligand-based pharmacophore model of MPO inhibitors was developed to identify crucial molecular features required for inhibition. A quantitative structure-activity relationship (QSAR) model was built using the Genetic Function Approximatio

Artificial intelligence in epilepsy diagnosis: Clinical readiness, failure modes, and standards for Implementation.

Epilepsy diagnosis is not a single classification task but a sequence of clinically consequential decisions: whether a paroxysmal event is epileptic, whether the patient meets the definition of epilepsy, how seizures and epilepsy should be classified, and what etiology or epileptogenic network explains the disorder. Artificial intelligence (AI) can support these decisions through natural language processing, conversational systems, computer vision, wearable sensors, electroencephalography (EEG), magnetic resonance imaging (MRI), genetics, and multimodal models. The evidence is uneven. Automate

SPFuseRanker: A Multi-Importance Score Fusion Framework for Core Microbiome Identification in Metagenomic Data.

Clinical metagenomic data are typically high-dimensional, sparse, and zero-inflated, and are often characterized by limited sample sizes and measurement noise. In addition, different feature-importance methods may produce inconsistent taxon rankings, which limits the stability of single-method feature selection and complicates the identification of candidate core microbiome members. To address this issue, we propose SPFuseRanker, a score-fusion-based ranking framework for integrating multiple microbial importance measures in metagenomic data. The method constructs a consensus score vector by c

Cognitive changes associated with chemotherapy in breast cancer: an assessment of social cognition and executive functions in Peruvian patients.

BACKGROUND: Cognitive impairment related to chemotherapy-commonly referred to as "chemo brain"-is a well-documented phenomenon among breast cancer patients. These impairments affect memory, attention, executive function, and social cognition, yet remain understudied in low- and middle-income countries. In Peru, where populations present a high proportion of Amerindian ancestry and distinct sociocultural factors, evidence is scarce. METHODS: We conducted a longitudinal study of 143 Peruvian women aged 28-64 years, newly diagnosed with early-stage breast cancer and naïve to chemotherapy, treate

Structural determinants of ligand response specificity in the mast cell activating GPCR, MRGPRX2.

The mast cell-specific G-protein-coupled receptor (GPCR) MRGPRX2 (Mas-Related G Protein-coupled Receptor X2) has roles in itch and pain, and it mediates clinically relevant allergy-like responses to a diverse assortment of drugs. The varied responses of individuals to MRGPRX2 agonists, leading to drug hypersensitivity reactions in some cases, suggests the presence of consequential variants in the population. However, genetic associations with drug responses are poorly understood. We used heterologously-expressed MRGPRX2 to investigate the effect of 18 naturally occurring non-synonymous single

A novel chimpanzee-derived adenovirus, a recombinant of Mastadenovirus exoticum and Mastadenovirus blackbeardi members, reveals lineage-specific recombination hotspots in simian adenoviruses.

Adenoviruses are genetically diverse, non-enveloped double-stranded DNA viruses that infect a wide range of vertebrate hosts, including humans and non-human primates. Simian adenoviruses are of particular evolutionary relevance because of their close relationship with human adenoviruses, the cross-species exchanges documented between the two, and the recombination events that influence their genomic diversity. However, genomic information from simian adenoviruses circulating in the Americas remains limited, and their recombination patterns have not been systematically characterized. Here, we d

Multi-platform analysis of the tumor immune microenvironment associated with breast cancer subtypes.

Breast cancer is genetically and histologically heterogenous, and is influenced by a variety of factors, including the tumor microenvironment (TME). The PAM50 subtypes; Luminal A, Luminal B, Normal-like, Basal-like and Her2-enriched, are associated with different tumor phenotypes and overall survival. The quantity and quality of immune cell infiltration in breast tumors play a key role in cancer development and progression and are associated with survival and treatment response. We used multiplex immunohistochemistry, single-cell RNA sequencing, and two deconvolution algorithms to explore the

Digital death and posthumous data: a glocal, relational perspective.

Persistent digital presence now routinely outlives biological death through memorialized profiles, algorithmic "memories," AI-mediated avatars, or biobank reanalysis. This perspective argues that decisions about such posthumous data-including postmortem genomic data-should be grounded in a glocally situated, relational view of personhood and practice, rather than in one-size-fits-all, individual-only rules. A small shared vocabulary is first proposed for use across disciplines: digital afterlife, digital immortality, posthumous lifespan, digital remains, and digital death. Digital death is tre

Development a computer-aided diagnosis system for dental caries detection applying radiographic images.

Delayed diagnosis of certain diseases like dental caries frequently leads to intensified problems due to postponed treatment. Untreated cavities can progress and affect a deeper layers of the tooth, which result in discomfort, infection, and ultimately tooth loss. Presently, manual diagnosis of dental decay in radiographic images by dentists in most dental clinics is prone to errors due to factors such as fatigue and information overload, emphasizing the necessity for automated diagnostic tools. This study develops an intelligent computer-aided diagnosis (CAD) system for accurate and efficient

Assessing chemical exposure risk in breastfeeding infants: An explainable machine learning model for human milk transfer prediction.

Breast milk is essential for infant health, but the transfer of xenobiotic chemicals poses significant risks. Ethical challenges in clinical trials necessitate the use of in vitro predictive models to assess chemical exposure risks in breastfeeding infants. This study introduces an explainable machine learning model to predict the risk of chemical transfer through human milk. Our novel framework integrates ensemble resampling methods with advanced feature selection techniques, addressing data imbalance and enhancing predictive accuracy. Targeted Cerebral Oxygenation Using Dedicated Treatment Versus Usual Care in Extremely Preterm Infants: Protocol for a Multicentre International Phase II Randomised Controlled Trial.

BACKGROUND: Near infrared spectroscopy (NIRS) allows continuous monitoring of cerebral oxygenation and therefore has the potential to be neuroprotective. Recurrent episodes of cerebral hypo-and/or hyperoxia may result in brain injury. The Safe-BoosC-II study reported stable cerebral oxygenation in extremely preterm infants by combining a dedicated treatment guideline with NIRS monitoring using several devices and adult sensors. The ability to maintain stable cerebral oxygenation with a dedicated treatment algorithm using one type of NIRS device with neonatal sensors has not been previously inv

Artificial intelligence-assisted optimization of Eichhornia crassipes extracts and evaluation of their biological activities.

In this research, the extraction conditions for Eichhornia crassipes (Mart.) Solms were optimized using Response Surface Methodology (RSM) and Artificial Neural Network-Genetic Algorithm (ANN-GA) techniques to enhance the biological efficacy of the extracts. The optimization focused on three key variables: extraction temperature, duration, and the ethanol-to-water solvent ratio. Through the ANN-GA model, the optimal parameters were identified as 56.85 °C for temperature, 7.62 h for extraction time, and 23.93% for the ethanol/water proportion. The obtained extracts showed statistically signific

Quantifying Sample Representation in Global Pharmacogenomic Studies of Major Depressive Disorder: A Systematic Review.

Major depressive disorder (MDD) is a substantial public health challenge. Pharmacogenomics (PGx), which identifies genetic variations that predict drug treatment outcomes, may have utility for clinical practice, but adequate representation of all populations is needed. As precision medicine in psychiatry moves towards the use of Artificial Intelligence (AI) and Machine Learning (ML) to predict treatment outcomes using PGx data, representation of diverse populations will be especially important in order to mitigate algorithmic bias and achieve equitable and generalizable findings. This work sou

The Role of Artificial Intelligence in Identifying NF1 Gene Variants and Improving Diagnosis.

Neurofibromatosis type 1 (NF1) is an autosomal dominant disorder caused by mutations in the NF1 gene, typically diagnosed during early childhood and characterized by significant phenotypic heterogeneity. Despite advancements in next-generation sequencing (NGS), the diagnostic process remains challenging due to the gene's complexity, high mutational burden, and frequent identification of variants of uncertain significance (VUS). This review explores the emerging role of artificial intelligence (AI) in enhancing NF1 variant detection, classification, and interpretation. A systematic literature s

Pain Management in Autosomal Dominant Polycystic Kidney Disease: Clinical Challenges and a Stepwise Algorithmic Approach.

Autosomal dominant polycystic kidney disease (ADPKD) is the most common genetic kidney disorder. It is primarily caused by pathogenic variants in the PKD1 or PKD2 genes. This leads to the development of numerous kidney cysts, which can result in kidney enlargement and progression to kidney failure. Pain is a common symptom in ADPKD and can negatively affect quality of life (QOL). This pain is often due to the continuous growth of kidney and liver cysts or associated cystic complications. We present a case of a 29-year-old woman with ADPKD who experienced chronic, refractory right-sided flank p
