Cochliomyia hominivorax and Lucilia cuprina are major pests of livestock. Their larvae infest warm-blooded vertebrates and feed on host's tissues, resulting in severe industry losses. As they are serious pests, considerable effort has been made to develop genomic resources and functional tools aiming to improve their management and control. Here, we report a significant addition to the pool of genome manipulation tools through the establishment of efficient CRISPR/Cas9 protocols for the generation of directed and inheritable modifications in the genome of these flies. American trypanosomiasis, or Chagas disease, is a growing public health problem in Panama, and further forest degradation due to human population growth is expected to worsen the situation. Most people infected with the parasite Trypanosoma cruzi are silently ill, and their life expectancy is severely compromised, which contributes to further deterioration of living conditions in endemic regions. Here, we review the outcomes of nearly 100 years of ecological and epidemiological investigation about Chagas disease in Panama, in an attempt to highlight progress, identify needs, and re-orient futu

The objectives of this study were to evaluate activity, rumination time, and their association with 3 kinds of pasture flies for organic dairy cows (n=57) fed 3 grain supplementation strategies during the grazing season from May to September 2013. Cows were assigned to 1 of 3 replicate supplementation groups: (1) no corn-grain supplementation (100% pasture, PAS, n=19); (2) low corn-grain (2.72kg/cow per day, LG, n=19); and (3) high corn-grain (5.44kg/cow per day, HG, n=19). Cows calved during 2 seasons (fall and spring) at the University of Minnesota West Central Research and Outreach Center,

BACKGROUND: Visceral leishmaniasis (VL) is a disease caused by two known vector-borne parasite species (Leishmania donovani, L. infantum), transmitted to man by phlebotomine sand flies (species: Phlebotomus and Lutzomyia), resulting in ≈50,000 human fatalities annually, ≈67% occurring on the Indian subcontinent. Indoor residual spraying is the current method of sand fly control in India, but alternative means of vector control, such as the treatment of livestock with systemic insecticide-based drugs, are being evaluated. We describe an individual-based, stochastic, life-stage-structured model

Expansion of mosquito-borne pathogens into more temperate regions of the world necessitates tools such as mathematical models for understanding the factors that contribute to the introduction and emergence of a disease in populations naïve to the disease. Often, these models are not developed and analyzed until after a pathogen is detected in a population. In this study, we develop a spatially explicit stochastic model parameterized with publicly available U.S. Census data for studying the potential for disease spread in Urbanized Areas of the United States. To illustrate the utility of the mo

Green synthesis technology is one of the rapid, reliable and best routes for the synthesis of silver nanoparticles (AgNPs). There are bioactive compounds with enormous potential in Azadirachta indica (Neem). The extraordinary mosquitoes warrant nanotechnology to integrate with novel molecules. This will be sustainable technology for future. Here, we synthesized AgNPs using aqueous extracts of leaves and bark of Az. indica (Neem). We tested AgNPs as larvicides, pupicides and adulticides against the malaria vector Anopheles stephensi and filariasis vector Culex quinquefasciatus. The results were

The Government of Tanzania introduced indoor residual spraying (IRS) in Muleba district in north-western Tanzania after frequent malaria epidemics. Malaria parasitological baseline and two cross-sectional follow-up surveys were conducted in villages under the IRS program and those not under IRS to assess the impact of IRS intervention. After two rounds of IRS intervention there was a significant reduction of malaria parasitological indices in both two villages. In IRS villages overall, parasitemia prevalence was reduced by 67.2%, splenomegaly was reduced by 75.8%, whereas anemia was reduced by

As a geographically isolated island chain with no native mosquitoes, Hawaii is a model for examining the mechanisms behind insect vector invasions and their subsequent interactions with each other and with human populations. The yellow fever mosquito, Aedes aegypti, and the Asian tiger mosquito, Ae. albopictus, have been responsible for epidemics of dengue in Hawaii. As one of the world's earliest locations to be invaded by both species, Hawaii's history is particularly relevant because both species are currently invading new areas worldwide and are implicated in outbreaks of emergent or reeme

The Camargue is an extensive wetland in the southeast of France, which is highly influenced by human activities. Large ponds, marshes and irrigated fields provide abundant potential breeding sites for mosquitoes. mosquitoes, which are important in terms of the nuisance they cause to people and animals, the limitations they impose on tourism and their potential threat to human health. Several of the mosquito species present are potential vectors of malaria and West Nile virus. Therefore, the population dynamics of these species were monitored over an entire breeding season during March-October

Anoplophora glabripennis (Motschulsky) (Coleoptera: Cerambycidae) is one of the most serious nonnative invasive forest insects discovered in North America in recent years. A. glabripennis is regulated by federal quarantines in the United States and Canada and is the subject of eradication programs that involve locating, cutting, and chipping all infested trees. Other control methods are needed to aid in eradication and to form an integrated management program in the event eradication fails. We conducted laboratory bioassays to determine the toxicity of two systemic insecticides, azadirachtin a

Franco-Brazilian cooperation in the field of microbiology and tropical diseases dates back to the onset of those disciplines. Physicians were sent over by France, namely Marchoux, Simond and Salimbeni from 1901 to 1905 to study yellow fever, and Emile Brumpt from 1913 to 1914 to teach parasitology. These missions brought in some important results. After confirming that the yellow fever agent was a filterable virus and that Stegomya (Aedes) its only vector, Simond and Marchoux clarified the biology of the mosquito and showed that sexual transmission of the virus could occur. They also set up di

An urgent need exists for new agents to control mosquito vectors of disease. Mosquito larvicides based on the bacteria Bacillus thuringiensis subsp. israelensis (Bti) or B. sphaericus (Bs) are effective in many habitats, but use is limited by their high cost. Moreover, mosquito resistance evolves rapidly to Bs where it is used intensively. The efficacy of these bacteria is due to a binary protein (BsB) in Bs and four proteins (Cry4A, Cry4B, Cry11A, and Cyt1A) in Bti. Here we report the use of cyt1A promoters and a 5' mRNA stabilizing sequence to synthesize high levels of Bs2362 binary toxin in

Insecticide-treated bedding materials (sheets and blankets) could be protective against vectors of malaria and leishmaniasis--especially in complex emergencies, epidemics and natural disasters where people are more likely to sleep in exposed situations. Comparison of cotton top-sheets impregnated with different pyrethroids (permethrin 500 mg/m2, deltamethrin 25 mg/m2 or alphacypermethrin 25 mg/m2) for effectiveness against mosquitoes (Diptera: Culicidae) was undertaken in a refugee camp in Pakistan. Predominant species encountered were Anopheles stephensi Liston, An. pulcherrimus Theobald, An.

A retrospective analysis was carried out on census data collected from house-to-house surveys during 1991-1992 in 4 areas endemic for Andean cutaneous leishmaniasis (uta) in the Department of Lima, Peru. Major changes in mean annual incidence in susceptible persons have taken place in these sites during the last 60 years. In particular, there is strong support for the hypothesis that, from the 1950s to the 1970s, the transmission rate was temporarily suppressed, largely as a by-product of the DDT house spraying campaign against malaria. These results are consistent with (i) anecdotal evidence,

Bacillus thuringiensis crystal (Cry) proteins have been expressed in commercial transgenic crops for nearly 30 years, providing safe and effective control of insect pests and significantly reducing the application of hazardous chemical pesticides. B. thuringiensis crystal proteins have also been shown to target parasitic nematodes, including plant parasitic nematodes. Recently, transgenic soybean crops expressing Cry14Ab have been shown to provide control against the soybean cyst nematode Heterodera glycines, marking the first time a crystal protein is being commercialized in transgenic crops

The integrated pest management (IPM) strategy was developed and used in combination with pesticides and beneficial biological control agents. To further develop IPM efficiency, it is important to evaluate the side effects of pesticides on biological control agents. Aphidius gifuensis is one of the most important aphid natural enemies and has been successfully used to control Myzys persicae and other aphid species. Imidacloprid (IMD) is a popular pesticide used worldwide and is highly toxic to non-target arthropods. Here, we investigated the short-term sublethal toxicity of IMD in Aphidius gifu

Nearly 1.4 billion people in 73 countries worldwide are threatened by lymphatic filariasis, a parasitic infection that leads to a disease commonly known as elephantiasis. Filariasis is vectored by mosquitoes, with special reference to the genus Culex. The main control tool against mosquito larvae is represented by treatments with organophosphates and insect growth regulators, with negative effects on human health and the environment. Recently, green-synthesized nanoparticles have been proposed as highly effective larvicidals against mosquito vectors. In this research, we attempted a reply to t

We are interested in generating a Y-autosome translocation of the Resistance to dieldrin (Rdl) locus in the malaria vector mosquito Anopheles stephensi Liston (Diptera: Culicidae), for use in sterile insect release. To ensure stability of the system, a recombination suppressing inversion can also be induced which encompasses the Rdl locus. As a first step, here we report the cloning of fragments of the Rdl gene from both An. stephensi and An. gambiae Giles using degenerate primers in the polymerase chain reaction. These fragments encode the second membrane-spanning region of the gamma-aminobut
