Ethanol-Induced Neurodegeneration and Glial Activation in the Developing Brain.

Ethanol induces neurodegeneration in the developing brain, which may partially explain the long-lasting adverse effects of prenatal ethanol exposure in fetal alcohol spectrum disorders (FASD). While animal models of FASD show that ethanol-induced neurodegeneration is associated with glial activation, the relationship between glial activation and neurodegeneration has not been clarified. This review focuses on the roles of activated microglia and astrocytes in neurodegeneration triggered by ethanol in rodents during the early postnatal period (equivalent to the third trimester of human pregnanc…

Involvement of sphingolipids in ethanol neurotoxicity in the developing brain.

Ethanol-induced neuronal death during a sensitive period of brain development is considered one of the significant causes of fetal alcohol spectrum disorders (FASD). In rodent models, ethanol triggers robust apoptotic neurodegeneration during a period of active synaptogenesis that occurs around the first two postnatal weeks, equivalent to the third trimester in human fetuses. The ethanol-induced apoptosis is mitochondria-dependent, involving Bax and caspase-3 activation. Such apoptotic pathways are often mediated by sphingolipids, a class of bioactive lipids ubiquitously present in eukaryotic …

Maternal drug therapy: effect on fetal and neonatal growth and neurobehavior.

Fetal and neonatal growth retardation may result in long term neurological and mental morbidity. There are multiple causes for intrauterine growth retardation but one etiology proposed is maternal drugs. In many instances it is difficult to determine if the growth retardation is due to the drugs or it is due to the disease process for which the mother receives the drugs. Weight and FOC appear to be more adversely effected than length. Combination drug therapy, for example anticonvulsant drugs, results in a greater incidence in teratology, growth retardation and lower IQ than monotherapy. Neuro

Paternal lineage effects of prenatal ethanol exposure: Offspring neuroanatomy and behavior in a multigenerational mouse model of FASD.

BACKGROUND: Fetal alcohol spectrum disorders (FASD) encompass a range of phenotypes in offspring exposed to ethanol via maternal consumption during pregnancy. In a series of studies, our laboratory identified deleterious effects of prenatal ethanol exposure (PrEE) in our FASD mouse model. The first filial generation (F1) of PrEE offspring exhibited abnormal neocortical gene expression, ectopic intraneocortical connectivity, altered neuroanatomy, and disrupted behavior. Our results suggest that PrEE can induce transgenerational transmission of the abnormal phenotypes, potentially via epigenetic…

Feasibility study of a family-focused intervention to improve outcomes for children with FASD.

BACKGROUND: Growing evidence shows that children with fetal alcohol spectrum disorder (FASD) can benefit from interventions, and specifically interventions focused on improving self-regulation. However, novel ways of improving outcomes for children with FASD need further investigation so that programs target not only the individual child but also the family context, which includes the parent-child relationship. AIMS: The current study aimed to evaluate the feasibility of an adapted version of the Parents under Pressure (PuP) program that addresses self-regulatory processes, through improving

Male germline transmits fetal alcohol epigenetic marks for multiple generations: a review.

Alcohol exposure during fetal and early postnatal development can lead to an increased incidence of later life adult-onset diseases. Examples include central nervous system dysfunction, depression, anxiety, hyperactivity, and an inability to deal with stressful situations, increased infection and cancer. Direct effects of alcohol leading to developmental abnormalities often involve epigenetic modifications of genes that regulate cellular functions. Epigenetic marks carried over from the parents are known to undergo molecular programming events that happen early in embryonic development by a wa

Elevated fatty acid ethyl esters in meconium of sheep fetuses exposed in utero to ethanol--a new animal model.

Specific fatty acid ethyl esters (FAEE) in meconium of newborns have been shown to correlate with maternal ethanol exposure. An animal model is needed to assess the validity of this biomarker. We hypothesized that the pregnant/fetal sheep is a feasible animal model for validating FAEE as a biomarker of prenatal ethanol exposure. Nine pregnant ewes were treated during the third trimester with different i.v. ethanol doses. The control group consisted of 14 pregnant ewes exposed to similar volumes of saline. On gestational d 133, the fetuses were delivered and meconium samples removed. FAEEs were

A stereo-photogrammetric method to measure the facial dysmorphology of children in the diagnosis of fetal alcohol syndrome.

In diagnosing a child with fetal alcohol syndrome (FAS), anthropometric measurements of the face are conventionally performed by highly trained dysmorphologists using a hand-held ruler. This renders the screening of large populations of children for the facial features characteristic of FAS very time-consuming and costly. This study proposes a new, cost-effective, and non-intrusive method to measure in three dimensions the facial dysmorphology of children using stereo-photogrammetry. The face of each child is photographed in a control frame simultaneously by a pair of high-resolution digital c

Alcohol deters the outgrowth of serotonergic neurons at midgestation.

We have previously demonstrated that treatment of pregnant C57BL mice from gestation days 8 to 14 with alcohol with 20% ethanol-derived calories (EDC) reduced the number of serotonin (5-HT) neurons and retarded their migration in the fetal brains. In the present study, we obtained similar results with the use of 25% EDC and extended our previous findings by demonstrating that besides the alteration of the number of 5-HT neurons, prenatal alcohol exposure also affects their projecting fibers in their early development. Pregnant C57BL mice were divided into an alcohol-exposed (ALC) group given 2

Loss of nicotine-induced effects on locomotor activity in fetal alcohol-exposed rats.

Previous evidence from our laboratory showed that systemic injection of nicotine enhanced attention and memory in control rats, but not fetal alcohol-exposed (FAE) rats. The present study examined the effects of nicotine on two measures of locomotor activity in FAE rats. Subjects were 2-month-old male offspring of Sprague-Dawley rats fed a 35% ethanol-derived caloric diet, a pair-fed sucrose diet, or a chow-fed diet during the last 2 weeks of gestation. The two experiments examined the effects of intraperitoneal injection of saline or nicotine (0.25 or 0.75 mg/kg) on rearing in an operant cham

Fetal protection in Wisconsin's revised child abuse law: right goal, wrong remedy.

Authors examine Wisconsin's recent revision of its child abuse and protection laws to address substance abuse by pregnant women. The new statute enables the state to take the fetus into protective custody. Authors argue that approaching fetal protection using a child abuse model creates a series of symbolic, conceptual, and practical problems of such severity as to undermine its justifiability as a public health measure.

Intragastric intubation: important aspects of the model for administration of ethanol to rat pups during the postnatal period.

One technique for the controlled delivery of ethanol to neonatal rat pups is intragastric intubation. Often, the vehicle used for delivery of ethanol is composed of a nutrient mixture to compensate for decreased suckling or other possible nutritional compromise. This study analyzed the selection of nutrient vehicle, the combination of experimental treatment groups within a litter, and the overall litter size on the growth rate of ethanol-intubated and intubated-control pups, compared with mother-raised control pups. Sprague-Dawley rat pups were raised in litters of 8 or 10, and administered et

Effects of maternal ethanol consumption and buspirone treatment on dopamine and norepinephrine reuptake sites and D1 receptors in offspring.

Previously, it was shown that in utero ethanol exposure results in decreased serotonin (5-HT) and altered concentrations of 5-HT reuptake sites and 5-HT1A receptors in fetal and/or postnatal rats. Because fetal 5-HT is an essential trophic factor, this laboratory previously investigated the hypotheses that the early ethanol-associated 5-HT deficit contributed to subsequent development abnormalities in the serotonergic system and that the effects of the fetal 5-HT deficit could be prevented by maternal treatment with buspirone, a 5-HT1A receptor agonist. The present report determined the effect

Effect of postnatal ethanol exposure on expression of differentiation antigens of murine splenic lymphocytes.

Ethanol is a recognized immunosuppressive agent in the chronic alcoholic. However, the effects of ethanol exposure on the developing immune system have not been extensively investigated. This study evaluated the effects of early postnatal ethanol exposure, via breast milk, on splenic lymphocyte differentiation antigen expression in offspring reared by ethanol-fed mice. Maternal mice were fed a liquid diet containing 20% ethanol-derived calories during pregnancy (E-P), pregnancy and lactation (E-PL), or lactation (E-L). Ad libitum-fed (C) and pair-fed (PF) control groups, fed a control liquid d

Incidence of renal anomalies in children prenatally exposed to ethanol.

OBJECTIVE: Based on a number of studies involving animals as well as human case reports indicating an association between prenatal ethanol exposure and renal malformations, it has been suggested that children with fetal alcohol syndrome (FAS) should be screened for renal anomalies. The purpose of this study was to evaluate a group of children prenatally exposed to alcohol to determine the incidence of renal anomalies and to evaluate the need for such a screening procedure. METHODS: Renal ultrasounds were performed on a total of 84 patients (68 children, 13 adolescents, and three adults). In a
