Objective: To explore the molecular mechanisms by which ethionine induces neural tube defects (NTD) through regulating the lysine demethylase 5A (KDM5A)/trimethylated lysine 4 of histone H3 (H3K4me3) axis and modulating the Wnt/β-catenin signaling pathway. Methods: Specific pathogen-free adult male and female C57BL/6 mice (80 females and 35 males) were housed overnight in a 2∶1 female-to-male ratio. Vaginal plugs were examined the following morning to confirm pregnancy. Pregnant mice were randomly divided into two groups: the ethionine experimental group (ethionine group) and the control group

PURPOSE: Surgical site infections (SSIs) represent a leading cause of postoperative morbidity in pediatric neurosurgery, particularly among patients with central nervous system (CNS) tumors. Although global SSI rates range from 1% to 10%, there is limited data from low-resource settings such as Ethiopia. This study aimed to determine the magnitude of SSIs and identify associated risk factors among pediatric CNS tumor patients treated at tertiary referral hospitals in Ethiopia. METHODS: A retrospective cross-sectional study was conducted by reviewing 250 medical records of pediatric CNS tumor p

BACKGROUND: Genomic imprinting is an epigenetic gene regulatory mechanism; disruption of this process during early embryonic development can have major consequences on both fetal and placental development. The periconceptional period and intrauterine life are crucial for determining long-term susceptibility to diseases. Treatments and procedures in assisted reproductive technologies (ART) and adverse in-utero environments may modify the methylation levels of genomic imprinting regions, including insulin-like growth factor 2 (IGF2)/H19, mesoderm-specific transcript (MEST), and paternally expres

From the foregoing description of the histological changes in the leptomeninx it is quite evident that we are dealing with a chronic, stationary, healing form of tuberculous inflammation. This statement is substantiated, in the first place, by the clinical history. The only reasonable interpretation of the symptoms would establish the duration of the process as four months. The imaginable contingency that there existed first a meningeal syphilitic lesion that was dispersed by the iodide of potassium only to be followed by a tuberculous infection is so remote and unlikely that it need not be di

BACKGROUND: Congenital heart disease (CHD) is a type of heart structure defect caused by the abnormal development of the heart and major blood vessels during the embryonic stage. With the continuous innovation of surgical treatment techniques and the improvement of perioperative management, the survival rate of children with CHD has significantly improved. However, accompanying central nervous system development problems have become increasingly prominent. This study investigated the clinical utility of neurite orientation dispersion and density imaging (NODDI) in evaluating preoperative cereb

BACKGROUND: Aberrant right subclavian artery is a common anatomical variant of the embryonic aortic arch, with a prevalence ranging from 0.4 to 2.0%. Although frequently associated with vascular rings or congenital cardiac defects, prenatal assessment primarily relies on the three-vessel and trachea view in ultrasonography. Currently, there is no consensus regarding whether isolated ARSA necessitates invasive diagnostic procedures. This study aimed to evaluate the necessity of routine invasive prenatal diagnosis for fetuses with sonographically isolated ARSA. By conducting a long-term postnata

The basic-helix-loop-helix (bHLH) proteins are a superfamily of DNA-binding transcription factors that regulate numerous biological processes in both invertebrates and vertebrates. One family of bHLH transcriptional repressors is related to the Drosophila hairy and Enhancer-of-split proteins. These repressors contain a tandem arrangement of the bHLH domain and an adjacent sequence known as the Orange domain, so we refer to these proteins as bHLH-Orange or bHLH-O proteins. Phylogenetic analysis reveals the existence of four bHLH-O subfamilies, with distinct, evolutionarily conserved features. A

STUDY QUESTION: What are the effects of experimentally manipulating meiosis signalling by addition of retinoic acid (RA) in cultured human fetal gonads?
SUMMARY ANSWER: RA-treatment accelerated meiotic entry in cultured fetal ovary samples, while addition of RA resulted in a dysgenetic gonadal phenotype in fetal testis cultures.
WHAT IS KNOWN ALREADY: One of the first manifestations of sex differentiation is the initiation of meiosis in fetal ovaries. In contrast, meiotic entry is actively prevented in the fetal testis at this developmental time-point. It has previously been shown that RA-tr

Little is known concerning the time-course and structural dynamics of reactivation of compensatory myogenesis in denervated muscle, its initiating cellular mechanisms, and the relationship between this process and the progression of postdenervation atrophy. The purpose of this study was to investigate the interrelations between temporal and spatial patterns of the myogenic response in denervated muscle and progressive atrophy of muscle fibers. Another objective was to study whether reactivation of myogenesis correlates with destabilization of the differentiated state and death of denervated mu

STUDY QUESTION: Are there age-related differences in gene expression during the germinal vesicle (GV) to metaphase II (MII) stage transition in euploid human oocytes?
SUMMARY ANSWER: A decrease in mitochondrial-related transcripts from GV to MII oocytes was observed, with a much greater reduction in MII oocytes with advanced age.
WHAT IS KNOWN ALREADY: Early embryonic development is dependent on maternal transcripts accumulated and stored within the oocyte during oogenesis. Transcriptional activity of the oocyte, which dictates its ultimate developmental potential, may be influenced by age a

WNT2B is canonically characterized as a secreted WNT-family ligand, which is transported to the extracellular space via the endoplasmic reticulum (ER)-Golgi pathway and binds to cell surface FZDs (frizzled class receptors) to trigger downstream signaling cascades. Here, we identify a previously unrecognized non-secretory intracellular function of WNT2B in impairing endosomal trafficking to inhibit macroautophagy/autophagy, as well as a non-canonical LC3B-II-dependent autophagic secretion mechanism for WNT2B. Specifically, the non-secretory intracellular pool of WNT2B via its conserved middle d

We report 20 cases of a distinct, previously unrecognized renal neoplasm, anaplastic sarcoma of the kidney with polyphenotypic features. The tumors were identified by re-reviewing tumors with unusual anaplastic features from the National Wilms Tumor Study Pathology Center, the International Society of Pediatric Oncology and the United Kingdom Children's Cancer Study Group trials. Patients ranged in age from 10 months to 41 years (median age 5 y, mean age 12 y) and females predominated (1.5:1). Twelve tumors presented in the right kidney, and 5 in the left (laterality was unknown in 3 cases). T

STUDY QUESTION: Can third-generation sequencing (TGS)-based polar body (PB) analysis serve as a viable alternative to conventional trophectoderm (TE) biopsy for preimplantation genetic testing for aneuploidy (PGT-A)?
SUMMARY ANSWER: This study demonstrates the feasibility of using TGS and PB biopsy for clinical PGT-A, particularly in advanced maternal age (AMA) patients.
WHAT IS KNOWN ALREADY: TE biopsy, the current standard approach for PGT-A, is limited by embryonic mosaicism. Mosaicism potentially leads to false-positive aneuploidy diagnoses, resulting in the discard of genetically normal

The aim of this study was to characterize the specific role of the various vascular endothelial growth factor (VEGF) isoforms in different aspects of blood vessel formation: vessel outgrowth, arterial and venous differentiation, and vascular remodeling and patterning. Although the role of VEGF in the early stages of vascular assembly has been studied extensively, its role in the maturation stage, involving vascular remodeling and patterning, as well as in the establishment of arteries and veins, remains enigmatic. The three major VEGF isoforms are known to differ in their solubility (VEGF120 i

Both in routine diagnostics and detailed, highly specialized workups, major advances have been observed in many areas of ultrasound due to an increase in expertise and improved technology in recent years. This is particularly true in the case of fetal neurosonography 1 2 3 4. Malformations of the CNS together with fetal heart defects are among the most common congenital anomalies. From the embryonic phase to the late third trimester, the CNS undergoes extensive development and maturation processes. The diagnosis of CNS anomalies is therefore primarily dependent on the time at which the examina

OBJECTIVES: Maxillary transverse deficiency is a common malocclusion frequently observed in orthodontic clinics. Miniscrew-assisted rapid palatal expansion (MARPE) not only produces greater skeletal expansion but also offers advantages such as simple miniscrew implantation without flap elevation, enhanced patient comfort, and an expanded age range and indications for palatal expansion. However, the fixed connection between the expander and the miniscrews makes the expander difficult to remove, significantly hindering its clinical application. This study aims to employ a non-common path of inse

Bone diseases such as osteoporosis and osteoarthritis have become important human health problems, requiring a deeper understanding of the pathogenesis of related diseases and the development of more effective treatments. Bone organoids are three-dimensional tissue masses that are useful for drug screening, regenerative medicine, and disease modeling because they may mimic the structure and physiological activities of organs. Here, we describe various potential methods for culturing bone-related organoids from different stem cells, detailing the construction processes and highlighting the main
