[Embryonal alcohol syndrome ].

The permanent alcohol abuse of a pregnant woman is of high risk for the embryo especially during the first weeks of pregnancy. It is prerequisite to the embryonic alcohol syndrome that the ethylalcohol passes the placenta. Subsequently hypotrophic infants with multiple malformations of minor and major quality are born. Part of the malformations are characteristic, part of them are unspecific for the disease. They altogether represent the entity of the embryonic alcohol syndrome. Occurrence and intensity of the disease depend on the daily quantity of pure alcohol, especially during the first mo

Therapeutic Effects of Catechins in Less Common Neurological and Neurodegenerative Disorders.

In recent years, neurological and neurodegenerative disorders research has focused on altered molecular mechanisms in search of potential pharmacological targets, e.g., imbalances in mechanisms of response to oxidative stress, inflammation, apoptosis, autophagy, proliferation, differentiation, migration, and neuronal plasticity, which occur in less common neurological and neurodegenerative pathologies (Huntington disease, multiple sclerosis, fetal alcohol spectrum disorders, and Down syndrome). Here, we assess the effects of different catechins (particularly of epigalocatechin-3-gallate, EGCG)…

A Pilot Randomised Controlled Trial of Sublingual Melatonin for Sleep Onset Insomnia in Children With Foetal Alcohol Spectrum Disorder (FASD).

AIM: Children with foetal alcohol spectrum disorder (FASD) present with significant sleep disturbance. This pilot trial aimed to examine the feasibility of conducting a larger randomised controlled trial (RCT) in this population and the potential effectiveness of melatonin to treat onset insomnia. METHODS: Children with sleep onset delay and a designation of 'at risk' of FASD or a diagnosis of FASD were recruited to a 10-week, double-blind, placebo-controlled, crossover study of melatonin. Sleep outcomes were assessed using actigraphy. The child sleep habit questionnaire (CSHQ) and measures o

Comparison of adaptive behavior in children with heavy prenatal alcohol exposure or attention-deficit/hyperactivity disorder.

BACKGROUND: Adaptive behavior, the ability to respond successfully to everyday demands, may be especially sensitive to the effects of heavy prenatal alcohol exposure. Similar adaptive dysfunction is common in other developmental disorders including attention-deficit/hyperactivity disorder (ADHD). ADHD is frequently present in alcohol-exposed children and this overlap in clinical presentation makes identification of alcohol-exposed children difficult. Direct comparison of children with prenatal alcohol exposure and ADHD may yield distinct patterns of cognitive and behavioral performance and add

[Early diagnosis important in hearing loss in children].

Owing to the identification of pre-, peri- and post-natal risk factors for the development of hearing loss or impairment, diagnosis is now possible at 10-14 months of age and rehabilitative measures can be implemented. It is vital that hearing impairment is discovered at an early stage, as language development occurs predominantly before the age of three. Several methods have been evolved for testing hearing in infants.

Alcohol exposure in utero and functional deficits in children during the first four years of life.

The behavioural effects of varying amounts of alcohol exposure in utero were studied using a longitudinal prospective design. An unselected sample of 1529 pregnant women was interviewed in the fifth month of pregnancy regarding beverage consumption, diet, drug use, and socioeconomic background; a cohort of approximately 500 infants was selected at delivery for follow-up. Lengthy individual behavioural and anthropometric examinations were made of the children between birth and four years of age. Alcohol-related behavioural effects are still statistically significant at four years, as they were

Gypenosides protected the neural stem cells in the subventricular zone of neonatal rats that were prenatally exposed to ethanol.

Fetal alcohol spectrum disorder (FASD) can cause severe mental retardation in children who are prenatally exposed to ethanol. The effects of prenatal and early postnatal ethanol exposure on adult hippocampal neurogenesis have been investigated; however, the effects of prenatal ethanol exposure on the subventricular zone (SVZ) have not. Gypenosides (GPs) have been reported to have neuroprotective effects in addition to other bioactivities. The effects of GPs on neural stem cells (NSCs) in the FASD model are unknown. Here, we test the effect of prenatal ethanol exposure on the neonatal SVZ, and

Pre- and postnatal alcohol exposure delays, in female but not in male rats, the extinction of an auditory fear conditioned memory and increases alcohol consumption.

Repeated exposure to alcohol increases retrieval of fear-conditioned memories, which facilitates, among other factors, the emergence of post-traumatic stress disorder (PTSD). Individuals with PTSD are more likely to develop alcohol and substance abuse related disorders. We assessed if prenatal and early postnatal alcohol exposure (PAE) increased the susceptibility to retain aversive memories and if this was associated with subsequent heightened alcohol consumption. Pregnant Sprague-Dawley rats were exposed for 22 hr/day, throughout pregnancy and until postnatal Day 7 to a single bottle of sucr

Alcohol-related harm to others in New Zealand: evidence of the burden and gaps in knowledge.

AIM: To identify and summarise existing New Zealand data quantifying any aspects of harm experienced from the drinking of others. METHODS: Surveys, research data, and administrative databases were identified through literature searching, examination of websites of relevant agencies, and direct enquiry among those working in research, government agencies and relevant NGOs. Accessible data were analysed, and published or collated data were summarised. RESULTS: The prevalence of self-reported harm from others' drinking was higher than harm from own drinking (18% vs 12% in the past year) and was

Minocycline protects developing brain against ethanol-induced damage.

Fetal alcohol spectrum disorders (FASD) are caused by ethanol exposure during the pregnancy and is the leading cause of mental retardation. Ethanol exposure during the development results in the loss of neurons in the developing brain, which may underlie many neurobehavioral deficits associated with FASD. It is important to understand the mechanisms underlying ethanol-induced neuronal loss and develop appropriate therapeutic strategies. One of the potential mechanisms involves neuroimmune activation. Using a third trimester equivalent mouse model of ethanol exposure, we demonstrated that ethan…

Amphetamine sensitization and cross-sensitization with acute restraint stress: impact of prenatal alcohol exposure in male and female rats.

RATIONALE: Individuals with fetal alcohol spectrum disorder (FASD) are at increased risk for substance use disorders (SUD). In typically developing individuals, susceptibility to SUD is associated with alterations in dopamine and hypothalamic-pituitary-adrenal (HPA) systems, and their interactions. Prenatal alcohol exposure (PAE) alters dopamine and HPA systems, yet effects of PAE on dopamine-HPA interactions are unknown. Amphetamine-stress cross-sensitization paradigms were utilized to investigate sensitivity of dopamine and stress (HPA) systems, and their interactions following PAE. METHODS

Metabolic biomarkers of prenatal alcohol exposure in human embryonic stem cell-derived neural lineages.

BACKGROUND: Fetal alcohol spectrum disorders (FASD) are a leading cause of neurodevelopmental disability. The mechanisms underlying FASD are incompletely understood, and biomarkers to identify those at risk are lacking. Here, we perform metabolomic analysis of embryoid bodies and neural lineages derived from human embryonic stem (hES) cells to identify the neural secretome produced in response to ethanol (EtOH) exposure. METHODS: WA01 and WA09 hES cells were differentiated into embryoid bodies, neural progenitors, or neurons. Cells along this progression were cultured for 4 days with 0, 0.1, …

Exposure of rats to a high but not low dose of ethanol during early postnatal life increases the rate of loss of optic nerve axons and decreases the rate of myelination.

Visual system abnormalities are commonly encountered in the fetal alcohol syndrome although the level of exposure at which they become manifest is uncertain. In this study we have examined the effects of either low (ETLD) or high dose (ETHD) ethanol, given between postnatal days 4-9, on the axons of the rat optic nerve. Rats were exposed to ethanol vapour in a special chamber for a period of 3 h per day during the treatment period. The blood alcohol concentration in the ETLD animals averaged approximately 171 mg/dl and in the ETHD animals approximately 430 mg/dl at the end of the treatment on

Hippocampal nitric oxide synthase in the fetal guinea pig: effects of chronic prenatal ethanol exposure.

The effects of chronic maternal administration of ethanol on nitric oxide synthase (NOS) activity and the numbers of NOS containing neurons, and CA1 and CA3 pyramidal neurons in the hippocampus of the near term fetal guinea pig at gestational day (GD) 62 were investigated. Pregnant guinea pigs received oral administration of 4 g ethanol/kg maternal body weight (n = 5), isocaloric sucrose/pair feeding (n = 5) or water (n = 5), or no treatment (NT; n = 5) from GD 2 to GD 61. NOS activity in the 25,000 x g supernatant of hippocampal homogenate was determined using a radiometric assay. The numbers

Metabolic and mitotic changes associated with the fetal alcohol syndrome.

In the USA, fetal alcohol syndrome (FAS) is the leading known cause of mental retardation. FAS is estimated to affect 4000 infants yearly in the USA with an additional 7000 children suffering various forms of fetal alcohol effects in the absence of the full syndrome. A comparable incidence would be expected in other industrialized countries, but essentially no data are available from either developing or third world countries. An understanding of the biochemical causes of FAS has been slow to develop, but progress has been made toward a molecular causation theory of FAS. This paper summarizes

Ethanol alters hormone production in cultured human placental trophoblasts.

Maternal alcohol abuse during pregnancy can lead to abnormalities in fetal development, including the fetal alcohol syndrome (FAS). Although intrauterine growth retardation is a hallmark of FAS, the pathophysiology is not fully understood. A contributing factor may be altered placental function, which could affect fetal growth and development. As a major endocrine organ during pregnancy, changes in the production of placental hormones could affect pregnancy and possibly fetal development. In this study, the effect of continued exposure to ethanol on placental hormone production was examined us
