Application of in vitro lipolysis for the development of oral self-emulsified delivery system of nimodipine.

The objective of the current study was to optimize for the first time the formulation variables of self-emulsified drug delivery system (SEDDS) based on drug solubilization during lipolysis under a biorelevant condition of digestion such as lipase activity, temperature, pH, fed-fasting state, etc. Nimodipine (ND), a BCS class II, was used as a model drug to prepare the SEDDS. Various oils, surfactants, and cosurfactants were screened for their solubilization potential of ND. Area of self-emulsification was identified using various ternary phase diagrams. Box-Behnken design was employed to inve

Lack of seipin in neurons results in anxiety- and depression-like behaviors via down regulation of PPARγ.

The Seipin gene was originally found to be responsible for type 2 congenital lipodystrophy and involved in lipid droplet formation. Seipin is highly expressed in the central nervous system as well. Seipin mutations have been identified in motor neuron diseases such as Silver syndrome and spastic paraplegia. In this study, we generated neuron-specific seipin knockout mice (seipin-nKO) to investigate the influence of seipin deficiency on locomotion and affective behaviors. In comparison with control mice, 8-week-old male seipin-nKO mice, but not female mice, displayed anxiety- and depression-lik

Molecular aspects of adipoepithelial transdifferentiation in mouse mammary gland.

The circular, reversible conversion of the mammary gland during pregnancy and involution is a paradigm of physiological tissue plasticity. The two most prominent cell types in mammary gland, adipocytes and epithelial cells, interact in an orchestrated way to coordinate this process. Previously, we showed that this conversion is at least partly achieved by reciprocal transdifferentiation between mammary adipocytes and lobulo-alveolar epithelial cells. Here, we aim to shed more light on the regulators of mammary transdifferentiation. Using immunohistochemistry with cell type-specific lipid dropl

Long-chain polyunsaturated fatty acid transport across human placental choriocarcinoma (BeWo) cells.

Long-chain polyunsaturated fatty acids (LCPUFAs) such as docosahexaenoic acid (DHA) and arachidonic acid (AA) are essential for proper development of fetal brain and retina. These LCPUFAs are selectively enriched in the fetal circulation compared with the maternal circulation. In the current study we investigated the transfer of LCPUFAs and a non-essential fatty acid (oleic acid, OA) in a transwell monolayer system of placental choriocarcinoma (BeWo) cells. We show that incubation with OA results in increased triglyceride accumulation and lipid droplet formation compared with that of DHA. The

Comparison of hypolipidemic activity of synthetic gallic acid-linoleic acid ester with mixture of gallic acid and linoleic acid, gallic acid, and linoleic acid on high-fat diet induced obesity in C57BL/6 Cr Slc mice.

Hyperlipidemia is the major risk factors of heart disease such as atherosclerosis, stroke, and death. In the present study, we studied the effect of gallic acid (GA), linoleic acid (LA), mixture of GA and LA (MGL), and chemically synthesized gallic acid-linoleic acid ester (octadeca-9,12-dienyl-3,4,5-trihydroxybenzoate, GLE) on the ability to ameliorate hyperlipidemia in C57BL/6 mice fed a high-fat diet (HFD). GLE, GA, LA, and MGL were mixed with HFD and the composition of the test compounds were 1% of the diet for 7 weeks. After 7 weeks, the average body weight of ND and GLE groups was lower

GLP-1R agonist semaglutide protects human forebrain organoids from cholesterol-induced neurotoxicity.

INTRODUCTION: Dysregulated cholesterol metabolism represents a critical metabolic stressor in the central nervous system, contributing to neuronal injury across multiple neurological conditions, including Alzheimer's disease (AD). While glucagon-like peptide-1 receptor (GLP-1R) agonists show neuroprotective potential, their capacity to directly mitigate cholesterol-induced metabolic stress in human neural cells remains to be established. METHODS: Here, we modeled metabolic stress using human induced pluripotent stem cell-derived forebrain organoids exposed to cholesterol overload. The protect

Intermittent time-restricted feeding improves physical performance and modulates mitochondrial morphology in a muscle fiber type dependent manner in middle-aged mice.

Sarcopenia, defined as the age-related decline in skeletal muscle mass and function, markedly reduces physical performance, threatens functional independence, and diminishes quality of life in older adults. Although the clinical manifestations of sarcopenia typically emerge later in life, underlying molecular alterations, particularly within the mitochondrial network, occur well before symptom onset. Growing evidence indicates that dietary interventions, including caloric restriction as well as changes in meal timing, composition, and overall intake, play a critical role in attenuating age-ass

Structural-state-dependent effects of RVA-treated waxy starch on multiscale evolution and freeze-thaw resistance of myofibrillar protein emulsion gels: a perspective from processing to storage.

Physically modulating the structural state of waxy starch offers a clean-label strategy to improve the freeze-thaw (FT) tolerance of high-fat myofibrillar protein (MP) emulsion gels. In this study, waxy corn starches with distinct structural states, including native crystalline starch (NCS), partially disordered starches with gelatinization degrees of 45% (PDS45) and 70% (PDS70), and amorphous starch (AS), were prepared by thermal-shear treatment. These starches were incorporated into MP emulsions to elucidate their roles during emulsification, thermal gelation, and FT storage. During emulsifi

Loss of the cardiac-enriched mitochondrial transporter SLC25A34 promotes heart failure by attenuating fatty acid oxidation.

Heart failure is characterized by profound structural and metabolic remodeling. Members of the solute carrier family 25 (SLC25), as inner mitochondrial membrane proteins, play critical roles in mitochondrial dysfunction during heart failure; however, the role of cardiac-enriched SLC25 family members remains incompletely understood. In this study, we identify solute carrier family 25 member 34 (SLC25A34) as a previously unrecognized mitochondrial regulator of cardiac energy metabolism in heart failure. SLC25A34 is highly enriched in the heart and predominantly expressed in adult cardiomyocytes.

Antifungal activity and biochemical characteristics of natamycin against Fusarium graminearum.

Fusarium head blight (FHB) caused by Fusarium graminearum is a destructive fungal disease of wheat. Natamycin is a natural polyene macrolide antifungal compound exhibiting broad-spectrum antifungal activity, but its efficacy and mechanism against F. graminearum remain unclear. In this study, the sensitivity of 47 F. graminearum strains from different regions of China to natamycin was determined. The results showed that natamycin had a good inhibitory effect with the EC50 value ranging from 0.93 to 4.04 μg/mL and a mean EC50 value of 2.20 ± 0.59 μg/mL. No resistance association was found betwee

CPT1B K321 crotonylation contributes to cardiac dysfunction in endotoxic shock.

Lysine crotonylation (Kcr) is a novel posttranslational modification that has been proven to have evolutionary conservation. However, the role of protein Kcr in the pathogenesis of endotoxic shock-induced secondary cardiomyopathy is elusive. Here a classic rat model of endotoxic shock was induced by intraperitoneal lipopolysaccharide (LPS) injection. Crotonylproteomic analysis was subsequently performed on myocardial tissues to profile the Kcr modifications that occurred during endotoxic shock, and we found that the Kcr of carnitine palmitoyltransferase 1B (CPT1B) at the K321 site was signific

Fisetin alleviates osteoporosis by promoting osteogenesis and suppressing adipogenesis via the Wnt/β-catenin signaling pathway in mice.

Osteoporosis (OP) is characterized by an imbalance in which bone marrow mesenchymal stem cells (BMSCs) preferentially differentiate into adipocytes rather than osteoblasts. The natural polyhydroxyflavone fisetin functions as an antioxidant and multi-target signaling regulator with potential to modulate stem cell fate. However, its role and underlying mechanisms in OP remain unclear. This study investigated whether fisetin ameliorates OP by regulating osteogenic and adipogenic differentiation using integrated in vitro and in vivo models. The effects of fisetin on BMSC differentiation were asses
