The m.3244G>A mutation in mtDNA is another cause of progressive external ophthalmoplegia.

We sequenced all mitochondrial tRNA genes in a 61-year-old man with chronic progressive external ophthalmoplegia and mitochondrial myopathy but without mtDNA rearrangements, and identified a heteroplasmic m.3244G>A mutation in the tRNA(Leu(UUR)) gene. This mutation had been previously associated with the MELAS phenotype, but not described in any detail. The mutation load in muscle was 84% and COX-negative fibers harbored greater levels of mutant genomes than COX-positive fibers. The m.3244G>A mutation affects a highly conserved nucleotide in the dihydrouridine loop and has been associated with

STABILITY 2: A protocol for a randomised trial in orthopaedic surgery to determine whether anterior cruciate ligament (ACL) reconstruction using a patellar or quadriceps tendon autograft with or without lateral extra-articular tenodesis is more effective at reducing the incidence of recurrent instability in young, active athletes.

INTRODUCTION: Anterior cruciate ligament reconstruction (ACLR) is complicated by high failure rates in young, active individuals, which is associated with worse outcomes and higher rates of osteoarthritis (OA). Failure of ACLR limits activity, reduces quality of life and substantially increases socio-economic costs. Thus, strategies to reduce ACLR failure are imperative. In the STABILITY 1 trial, we showed that adding a lateral extra-articular tenodesis (LET) to ACLR with a hamstring tendon (HT) graft provided greater stability than HT alone (Relative Risk Reduction (RRR)=0.38; 95% CI 0.21 to

Effects of Rocuronium on Ischemia-reperfusion Injury and Intracellular ATP Consumption: A Randomized Controlled Trial.

BACKGROUND: Muscle relaxants reduce "total" oxygen consumption, and 1.2 mg/kg of rocuronium has been shown to attenuate ischemia-reperfusion … CONCLUSIONS: We were unable to confirm that 0.9 mg/kg of rocuronium attenuates IRI or postoperative quadriceps muscle weakness. While the present results suggest that rocuronium reduced intracellular ATP consumption, no clinically beneficial effects were observed.

Kinesio taping in knee osteoarthritis: mechanisms, clinical evidence, and rehabilitation implementations.

Kinesio taping is widely used as a non-pharmacological adjunct in knee osteoarthritis (OA), although its mechanisms and clinical value remain debated. This narrative review synthesizes current evidence on kinesio taping in knee OA, addressing mechanisms and effects on pain, function, muscle activation, proprioception, and range of motion (ROM). PubMed/MEDLINE, Scopus, Web of Science, Directory of Open Access Journals (DOAJ), and Google Scholar were searched through 19 May 2026 using MeSH terms "Osteoarthritis, Knee" and "Athletic Tape" with related keywords. Eligible sources included systemati

Quantification of a serum titin fragment reflects dystrophin restoration in mdx mice and disease severity in patients with dystrophinopathies.

Increased dystrophin expression in muscle has been accepted as a surrogate endpoint for accelerated approval of gene therapies within Duchenne Muscular Dystrophy (DMD). Previous studies identified an N-terminal titin fragment in urine as a biomarker suitable for monitoring disease progression and therapeutic interventions. Plasma levels of a titin fragment comprising aa 2,229-2,352 were found to negatively correlate with dystrophin expression in mdx mice treated with AAV9-CK8-μDys5 micro-dystrophin gene therapy. In this study, we sought to investigate which titin fragments can be detected in s

Walking speed and incline impact on mediolateral-stability control.

BACKGROUND: Controlling and stabilizing the body are crucial for walking on an inclined path. Healthy individuals adjust their gait to improve stability by reducing stride length and increasing cadence. Incline influences anteroposterior stability; however, its effects on mediolateral stability are inconclusive. RESEARCH QUESTION: What are the effects of walking inclination and speed on mediolateral stability using foot placement quantified by explained variance (EV)? METHODS: Fifteen healthy young adults walked on a treadmill under four speed (moderate 0.83 m/s, fast 1.0 m/s) and grade (0 a

Quantitative muscle magnetic resonance imaging as a biomarker for inclusion body myositis in clinical trials: exploring the in vivo effects of arimoclomol.

OBJECTIVES: To investigate the intramuscular effects of arimoclomol using quantitative magnetic resonance imaging (qMRI) of the thighs in a subset of inclusion body myositis (IBM) participants from a multicentre, randomised, double-blind, placebo-controlled trial, and to further evaluate the utility of qMRI assessments as outcome measures. METHODS: Eighteen participants (10 placebo, 8 arimoclomol-treated) were recruited to undergo an MRI at baseline, 12 and 20 months. Spearman correlations between baseline clinical measures and qMRI measurements [fat fraction (FF), remaining muscle area (RMA)

UEFA Women's Elite Club Injury Study: a prospective study on 1527 injuries over four consecutive seasons 2018/2019 to 2021/2022 reveals thigh muscle injuries to be most common and ACL injuries most burdensome.

OBJECTIVE: Injuries in women's football (soccer) have scarcely been investigated, and no study has been conducted in the highest competitive level involving club teams from different countries. Our aim was to investigate the time-loss injury epidemiology and characteristics among women's elite football players over four seasons. METHODS: 596 players from 15 elite women's teams in Europe were studied prospectively during the 2018/2019 to 2021/2022 seasons (44 team seasons). Medical staff recorded individual player exposure and time-loss injuries. Injury incidence was calculated as the number o

Identification of distinct immune signatures in inclusion body myositis by peripheral blood immunophenotyping using machine learning models.

OBJECTIVE: Inclusion body myositis (IBM) is a progressive late-onset muscle disease characterised by preferential weakness of quadriceps femoris and finger flexors, with elusive causes involving immune, degenerative, genetic and age-related factors. Overlapping with normal muscle ageing makes diagnosis and prognosis problematic. METHODS: We characterised peripheral blood leucocytes in 81 IBM patients and 45 healthy controls using flow cytometry. Using a random forest classifier, we identified immune changes in IBM compared to HC. K-means clustering and the random forest one-versus-rest model

Effect of body position and external ankle load on the pendulum test in adults.

BACKGROUND: The pendulum test is commonly used to measure passive stiffness about the knee in healthy and clinical populations. While most studies used an upright position, some studies explored different body positions, particularly with children. Additionally, including external ankle load increases the passive motion of the lower leg in children with Down syndrome. Does combining body position and ankle loading affect joint kinematics and muscle activity of the knee extensors during the pendulum test? METHODS: Twenty young adults participated in this study. Three body positions were includ

Excessive anterior tibial translation in the contralateral uninjured limb is significantly associated with ramp lesion in anterior cruciate ligament injury.

PURPOSE: This study aimed to evaluate the excessive anterior tibial translation (ATT) and muscle strength of patients with ramp lesions. We hypothesised that the higher ATT, lower hamstring-to-quadriceps (HQ) ratio, and higher flexion peak torque influenced by semimembranosus would be associated with ramp lesions. METHODS: One hundred and twenty-one patients who underwent anterior cruciate ligament (ACL) reconstruction were retrospectively evaluated. Clinical evaluation included ATT of the contralateral uninjured limb measured using a KT-1000 arthrometer, the knee flexor and extensor muscle s

Cut-off points of knee extension strength allometrically adjusted to identify sarcopenia risk in older adults: A cross-sectional study.

PURPOSE: Handgrip strength used to identify sarcopenia is not representative of overall strength, especially as greater muscle volume and consequent autonomy-mobility are concentrated in the lower limbs. While absolute strength or relative to body mass is used to define sarcopenia, this relationship is not always linear. The aim of this study was to establish allometrically adjusted cut-off points of lower limb (quadriceps) muscle strength to identify sarcopenia risk in older adults. METHODS: ninety-four physically independent individuals over the age of 60 were measured by DXA and sarcopenia

Neuromuscular function following prolonged intense self-paced exercise in hot climatic conditions.

Muscle weakness following constant load exercise under heat stress has been associated with hyperthermia-induced central fatigue. However, evidence of central fatigue influencing intense self-paced exercise in the heat is lacking. The purpose of this investigation was to evaluate force production capacity and central nervous system drive in skeletal muscle pre- and post-cycle ergometer exercise in hot and cool conditions. Nine trained male cyclists performed a 20-s maximal voluntary isometric contraction (MVC) prior to (control) and following a 40-km time trial in hot (35°C) and cool (20°C) co

Balance dysfunction in adults with haemophilia.

The main focus of lower limb physical performance assessment in people with haemophilia (PWH) has usually been on function, muscle strength and joint flexibility. The impact of haemophilic arthropathy on balance and falls risk is relatively under-explored. The aim of this study was to evaluate balance and related performance in PWH compared with age and gender matched healthy controls. It involved a comprehensive suite of clinical and laboratory measures of static and dynamic balance, mobility, strength, physical activity and falls efficacy completed in 20 PWH (mean age 39.4, 100% male) and 20

Random Electromyostimulation Promotes Osteogenesis and the Mechanical Properties of Rat Bones.

Exercise is often recommended as a promising non-pharmacologic countermeasure to prevent osteoporosis. However, elderly osteoporotic patients generally have physical fitness difficulties preventing them from performing effective and sustainable exercise. Electromyostimulation should be one effective modality for non-pharmacological prevention of osteoporosis without any voluntary physical movements. However, successful stimulation patterns remain controversial. As suggested by our previous in vitro studies, randomized timing of stimulation could be a candidate to maximize the osteogenic effect

What are causes and treatment strategies for patellar-tendinopathy in female runners?

Patellar-tendinopathy (PT) is a common overuse injury in long distance runners, especially in women. Until today, no definite combinations o … mprised 24 subjects (CO=12, PT=12). Biomechanical evaluations indicate eccentric overloading of the quadriceps muscle group (knee extensors), increased pronation velocity as well as a lack of joint coordination as major etiological factors in the development of PT. We assume that eccentric strengthening of the knee extensors, as well as reduction of pronation veloc …
