Lysine acetyl salicylic acid in acute renal pain.

The effect of repeated doses of 1.8 g lysine acetyl salicylic acid (LAS) i.v. on severe pain secondary to acute renal colic (ARC) was studied in 45 consecutive patients. Clinically acceptable analgesia was obtained in 65% of the cases. No additional pain relief was achieved with the combination of pethidine 100 mg i.v. + metoclopramide 10 mg, i.m. (narcotics). Pain relief occurred within five minutes in one third of the patients while in the rest within 30 minutes. Significant reduction of systolic blood pressure (mean +/- S.D.) 23.8 +/- 19.5, pulse rate (mean +/- S.D.) 19.5 +/- 10.1 and vomit

[Lysine acetyl salicylate. Clinical contribution and preliminary data].

In the general study of antipyretic products for children prone to febrile convulsions, the authors describe the results obtained administering, the rectal route, three different doses of lysine acetylsalicylate (20-30-40 mg/kg/b.w.) to 53 children with fever. The lower dose was often inadequate in these patients, who need a quick reduction of body temperature below 38 degrees C. The two higher doses were useful both in clinical and in statistical terms, without relevant differences between groups. A statistically significant inverse correlation between blood levels of salicylates and temperat

Species differences in hypocalcemia induced by acetyl-salicylic acid.

The study was performed to explore species differences in hypocalcemia induced by acetylsalicylic acid (ASA). ASA decreased plasma and serum calcium (Ca) level at the dose of more than 200 mg/kg s.c. and p.o. in mouse, rat and guinea pig, and a correlation between serum Ca and salicylic (SA) levels was recognized in these species. However, hypocalcemia was not observed in rabbit and dog by the oral administration of ASA 400 mg/kg though high serum levels were found. Species differences were recognized in ASA-induced hypocalcemia. It was suggested that the differences were probably due to the d

N-Acetyl-beta-glucosaminidase activity in hydatidiform mole.

The N-acetyl-beta-glucosaminidase activity in hydatidiform mole is two-fold higher than that in full-term placenta. Qualitatively, the enzymes from the two tissues are similar with respect to KM values and pH optima. Both enzymes also contain a new isoenzyme form detectable by polyacrylamide gel electrophoresis. However, the molar enzyme is more susceptible to heat denaturation, presumably due to the presence of a higher level of the heat-labile isoenzyme form A in this tissue. Data are also presented incicating that the placenta is not the source of the N-acetyl-beta-glucosaminidase activity

N-acetyl-beta-hexosaminidase: role in the degradation of glycosaminoglycans.

Extracts of cultured normal human skin fibroblasts released radioactivity from a (14)C-labeled heptasaccharide prepared by addition of [(14)C]N-acetylgalactosamine to the nonreducing terminus of a hexasaccharide derived from chondroitin 4-sulfate whereas fibroblast extracts from patients with Tay-Sachs and Sandhoff-Jatzkewitz diseases did not. The results suggest that N-acetyl-beta-hexosaminidase A is responsible for degradation of the oligosaccharide substrate.

Molecular basis for preferential protective efficacy of antibodies directed to the poorly acetylated form of staphylococcal poly-N-acetyl-beta-(1-6)-glucosamine.

Poly-N-acetyl-glucosamine (PNAG) is a staphylococcal surface polysaccharide influencing biofilm formation that is also under investigation for its vaccine potential. Antibodies that bind to PNAG with either low (<15%) or high (>90%) levels of acetate are superior at opsonic and protective activity compared with antibodies that bind to PNAG with only high levels (>70%) of acetate. PNAG is synthesized by four proteins encoded within the intercellular adhesin (ica) locus icaADBC. In Staphylococcus epidermidis, icaB encodes a deacetylase needed for the surface retention of PNAG and optimal biofilm

Amino acid solutions for premature neonates during the first week of life: the role of N-acetyl-L-cysteine and N-acetyl-L-tyrosine.

Tyrosine and cyst(e)ine are amino acids that are thought to be essential for preterm neonates. These amino acids have low stability (cyst(e)ine) or low solubility (tyrosine) and are therefore usually present only in small amounts in amino acid solutions. Acetylation improves the stability and solubility of amino acids, facilitating a higher concentration in the solution. We compared three commercially available amino acid solutions, Aminovenös-N-päd 10%, Vaminolact 6.5%, and Primène 10%, administered to 20 low-birth-weight neonates on total parenteral nutrition from postnatal day 2 onward. Ami…

LncRNA HITT stabilizes SIRT6 to restrain H3/H4 acetylation and suppress colorectal cancer growth.

Histone post-translational modifications are intimately linked to human diseases, including cancers. While diverse regulatory mechanisms have been described, how long non-coding RNAs (lncRNAs) control histone acetylation remains incompletely understood. Here, integrative bioinformatics analysis implicates the lncRNA HITT (HIF-1α inhibitor at the translation level) in histone modification-related pathways. Mechanistically, HITT selectively suppresses H3/H4 acetylation in colorectal cancer cells. Using siRNA-based loss-of-function screening and pharmacological inhibition, we identify the sirtuin…

ER-Nε-lysine acetylation machinery in neurological disorders: A context-dependent regulator of proteostasis.

Despite extensive characterization of cytosolic and nuclear acetylation, the functional relevance of luminal ER-Nε-lysine acetylation in the nervous system remains poorly understood. This modification is mediated by a coordinated metabolic-secretory axis involving acetyl-CoA availability and the ER-resident acetyltransferases ATase-1 (NAT8B) and ATase-2 (NAT8), which depend on acetyl-CoA import into the ER via the transporter SLC33A1 (AT-1). Upstream, the citrate transporters SLC25A1 and SLC13A5 contribute to shaping the cytosolic acetyl-CoA pool. Emerging evidence identifies this pathway as a…

Postnatal ketone deficiency impairs adult oocyte quality via enhancing histone acetylation in malnourished mice.

Early postnatal life is important for reproductive development, but the effects of early-life nutrition on female reproductive capacity rema … elopment. Transcriptomic and epigenomic analyses showed that Hmgcs2 deficiency disrupted histone H3 acetylation homeostasis in oocytes. These Ace-H3 changes were enriched in genes related to oxidative stress and apoptosis and were accompanied by transcriptional alterations. β-HB supplementation partially restored the altered Ace-H3 patterns. Together, our fin …

KAT5/TIP60 orchestrates H4K16 and H2A.Z acetylation to drive the gonocyte-to-spermatogonia transition.

Spermatogenesis begins with the gonocyte-to-spermatogonia transition (GST) during the perinatal window, yet how chromatin regulation orchestrates this developmental transition remains elusive. Transcriptomic analysis identified histone acetylation as a significantly enriched pathway during the GST, and immunofluorescence revealed a progressive increase in H4K16 acetylation alongside high levels of H2A.Z acetylation. Conditional deletion of the acetyltransferase Kat5/Tip60 drastically reduced both marks, leading to GST failure characterized by impaired prospermatogonial mitotic resumption, migr…

GCN5L1-Mediated Lysine Acetylation Regulates Mitochondrial Bioenergetics and Redox Homeostasis in the Aged Heart.

Precise control of mitochondrial electron transport is essential to maintain mitochondrial coupling and efficiency in ATP production. Furthermore, disruptions to ETC complex function can drive increased oxidant production, resulting in oxidative damage to the mitochondrion and bioenergetic inefficiency. This is highly relevant in the aging heart, as increased cardiac oxidative stress and mitochondrial dysfunction are hallmarks of age-related cardiovascular disease. Lysine acetylation has recently been characterized as a novel regulator of mitochondrial metabolic and bioenergetic function in th…

K49 acetylation of ACSL4 antagonizes its ubiquitination degradation and promotes tumorigenesis in hepatocellular carcinoma.

Lysine acetylation affects hepatocellular carcinoma (HCC) malignancy through multiple pathways. Evidence from our laboratory and other groups indicates that long chain acyl CoA synthetase 4 (ACSL4) is a crucial oncoprotein in HCC. However, the precise mechanisms underlying the post-translational regulation of ACSL4 acetylation remains unknown. Here, we report a previously unknown mechanism of ACSL4 regulation involving acetylation at the lysine 49 (K49) site by the acetyltransferase MOF. ACSL4 acetylation hindered its degradation via the ubiquitin-proteasome pathway. Using mass spectrometry an…

Profile of lysine acetylation in Eimeria tenella and its potential implications for anticoccidial research.

BACKGROUND: Coccidiosis has resulted in substantial economic losses in the poultry industry. The prevalence and severity of drug resistance to anticoccidial agents have exacerbated the challenges associated with controlling coccidiosis. Consequently, exploring novel drug targets is crucial for the prevention and treatment of coccidiosis. METHODS: In this study, we performed acetylation modification proteomics on the sporozoites and merozoites of Eimeria tenella during the asexual stage. Moreover, the anticoccidial efficacy of histone deacetylase (HDAC) inhibitors was evaluated both in vitro a…

Before import: N-terminal acetylation at the interface of plastid protein targeting and proteostasis.

Most chloroplast proteins are synthesized in the cytosol as precursor proteins carrying N-terminal transit peptides that mediate post-translational import into the organelle. During this transient cytosolic phase, precursor proteins are exposed to the cellular N-terminal processing machinery, including methionine aminopeptidases and N-terminal acetyltransferases (NATs). Recent proteomic studies have provided direct evidence that plastid precursor proteins undergo N-terminal acetylation prior to import, establishing a previously unrecognized link between co-translational processing and plastid …
