Two types of acetylcholine receptors on the soma of primary afferent neurons.

Acetylcholine (ACh) caused two types of depolarizations of the soma membrane of bullfrog primary afferent neurons (dorsal root ganglion cells); the one, a rapid transient depolarization, was nicotinic and the other, a long-lasting one, was muscarinic in nature, respectively. The rapid transient depolarization was due to a simultaneous increase in sodium and potassium conductance, whereas the slow one was caused by a decrease in membrane potassium conductance. These results indicate that the soma of bullfrog primary afferent neuron is endowed with nicotinic and muscarinic receptors.

Comparison of effects of cyanide and acetylcholine on renal hemodynamics and sodium excretion.

The present study compared the effects of acetylcholine to cyanide under the same experimental conditions of renal clearance in anesthetized dogs. Since cyanide is one of the few drugs for which the mechanism of action is known (cytotoxic hypoxia), some insight may be gained into the renal effects of acetylcholine since both produce direct natriuresis and diuresis. Infusion of 0.2 microgram/kg/min of acetylcholine and 12.0 microgram/kg/min of sodium cyanide into the left renal artery resulted in similar effects, i.e., increased fractional excretion of sodium, potassium, calcium, and magnesium.…

A further study of the cardiovascular responses to central administration of acetylcholine in rats.

General characteristics and the nature of cardiovascular responses to acetylcholine (ACh) injected into the lateral cerebral ventricle (LCV), the 4th cerebral ventricle (4th CV) and the cisterna magna (CM) were studied. Acetylcholine, depending on the route of injection, the dose and the peripheral vascular tone, produced both increases and decreases in blood pressure and heart rate. After injection of central anticholinesterase agents (physostigmine, neostigmine, armin), the pressor response to ACh was reversed to a depressor response, with or without a secondary pressor response, accompanied…

A negative inotropic effect of acetylcholine in the presence of several phosphodiesterase inhibitors.

The phosphodiesterase inhibitors papaverine, theophylline and 3-isobutyl-1-methylxanthine (IBMX) reveal a negative inotropic effect of acetylcholine in cat ventricular heart muscle. This effect in unrelated to beta-adrenoceptor stimulation and possibly mediated by the accumulation of cyclic GMP.

Block of acetylcholine-activated ion channels by an uncharged local anaesthetic.

It is now thought that amine local anaesthetic compounds (procaine, lignocaine and related molecules) depress electrical activity in nerve and muscle cells by binding to sites within ion channels and blocking current flow. Such mechanisms have been proposed to account for the effects of these local anaesthetics on both the voltage-dependent sodium current and the postsynaptic actylcholine (ACh)-activated ionic current. Recently, strong evidence for block of ion channels by cationic drug molecules has been obtained by recording current from single ACh-activated channels in the presence of perma

Acute and chronic effects of barbiturates on regional turnover of acetylcholine in brain.

Data concerning both the acute and chronic effects of barbiturates on acetylcholine [ACh] turnover in mouse and rat brain are presented. A single anaesthetic dose of pentobarbital [60 mg/kg, i.p.] to mice increased the endogenous content of ACh and decreased the turnover of ACh in the hippo-campus and cortex. No effect was found in the striatum. At sacrifice after chronic treatment with barbital to rats for about 30 weeks no effect was found on the endogenous ACh content. 3 days after the barbital solution was replaced by water [at the day with maximal number of abstinence convulsions] the amo…

Pathogenesis of the renal injury in choline deficiency: the role of catecholamines and acetylcholine.

A disturbed renal circulation due to an imbalance between vasoconstrictor catecholamines and a vasodilator such as acetylcholine, caused by a decrease in acetylcholine, has been postulated as the basic mechanism of haemorrhagic degeneration of the kidneys in choline deficiency. To explore this hypothesis further a group of male weanling Wistar rats was fed on a choline-deficient diet for 10 days (Group CD). A control group was fed on the same basal diet supplemented with choline (Group CS). Food intake and body weights were registered. The kidneys of choline-supplemented and choline-deficient …

Effects of pyrithiamin and oxythiamin on acetylcholine levels and utilization in rat brain.

Regional cerebral acetylcholine (ACh) levels and utilization rate were assessed in vivo in rats rendered thiamin deficient using the thiamin antagonists pyrithiamin or oxythiamin. ACh levels were significantly reduced in all brain regions of pyrithiamin treated rats and in the medulla-pons and striatum of oxythiamin treated rats compared to controls. ACh utilization was significantly reduced in the midbrain, striatum and hippocampus of pyrithiamin treated rats, but was reduced only in the striatum of oxythiamin treated rats compared to controls. Thus, there are some reductions in ACh levels an…

Identification and estimation of an acetylcholine-like substance in the venom of Dendroaspis jamesoni.

An acetylcholine-like substance was isolated from desiccated Jameson's mamba venom by one-dimensional ascending paper chromatography. The migratory and staining properties of the substance were identical with those of standard acetylcholine. Pharmacological identification on various in vitro and in vivo biological test objects showed that the substance was acetylcholine. It was further confirmed by high-voltage paper electrophoresis. The acetylcholine content was 4.35--4.36 mg/g of desiccated venom as shown by two different biological assay methods, and the index of discrimination was found to…

[Changes in the acetylcholine--cholinesterase system in the dynamics of experimental boltulin poisoning].

A study was made of the effect of botulin, type B, on the acetylcholine-cholinesterase system. High doses of the toxin increased the level of acetylcholine in the peripheral blood nervous system, as well as in the central nervous system. Injection of DLM of the toxin results in elevation of the acetylcholine level in the peripheral nervous system. Cholinesterase activity was practically unchanged.

Mechanism of action of pentagastrin and acetylcholine on the longitudinal muscle of the canine antrum.

1. Electrical and mechanical activities of the longtitudinal muscle of the dog antrum were recorded with the double sucrose-gap technique. 2 … e resting, passive, tension at LO was 570 mg. The active force of isometric contraction produced by acetylcholine increased with strip length up to a maximum, then decreased wtih further increased in length. There were no mechanical responses to pentagastrin. 7. Pentagastrin had two sites of action. On smooth muscle, it increased the frequency of the action pot …

The effect of hemicholinium-3 on choline and acetylcholine levels in a sympathetic ganglion.

Pregangliaaonic stimulation of the cat's superior cervical ganglion in the presence of hemicholinium-3 (HC-3) produced the expected depletion of acetylcholine (ACh) stores, but failed to cause a corresponding reduction in the choline content. These results suggest that either HC-3 possesses an intracellular site of action or that in lower doses it selectively inhibits a specialized choline transport system in cholinergic nerves. At a dose of 2 mg/kg, HC-3 probably blocked ACh synthesis completely in ganglia stimulated at 20 Hz. Under these conditions, there was a rapid depletion of ACh to abou…

Oestrogen-induced sensitization of the uterine artery of the guinea-pig to acetylcholine.

1. It has been shown previously that a dilatation of the uterine artery of the guinea-pig in response to acetylcholine or to stimulation of cholinergic vasodilator nerves occurs only during pregnancy. Experiments have now been performed to determine the hormonal basis of this change in responsiveness.2. Chronic treatment of virgin guinea-pigs with oestradiol, either by intramuscular injection or by subcutaneous implantation, caused the development of a responsiveness of the uterine artery to acetylcholine which was similar in degree to that occurring during pregnancy.3. Simultaneous administra…
