Aflatoxin B1 alters central and systemic tryptophan and tyrosine metabolism: influence of immunomodulatory drugs.

Semi-chronic exposure of ICR male Mice to Aflatoxin B1 (AFB1) in non-toxic doses results in elevated lung tryptophan (TRP) levels without change in serotonin (5-HT) or 5-hydroxyindole-3-acetic acid (5-HIAA) levels. This change is organ specific in that TRP levels are not altered in spleen, duodenum, heart or central nervous system (CNS). Acute (48 hour) flunixin treatment decreases lung TRP levels and reverses the AFB1 mediated increase in lung TRP levels. On the other hand, flunixin treatment decreases CNS TRP levels in control mice but not in AFB1 treated mice. Aflatoxin B1 treated mice have

Inactivation of aflatoxin B1 in corn meal, copra meal and peanuts by chlorine gas treatment.

More than 75% degradation of aflatoxin B1 (AFB1) was achieved after treatment of AFB1-spiked corn meal, spiked copra meal (the residue of the kiln-dried coconut kernels after mechanical expulsion of oil) and peanuts artificially infected with Aspergillus parasiticus, with 11, 16 and 35 mg chlorine gas per g meal or peanuts, respectively. At these chlorine gas treatment levels, extension of the exposure period of the corn meal and copra meal beyond 2.5 hr, and the peanuts beyond 1 day, did not increase the percentage degradation of AFB1. The mutagenicity of chlorine-treated copra meal and peanu

Food additives and plant components control growth and aflatoxin production by toxigenic aspergilli: a review.

Growth and aflatoxin production by toxigenic aspergilli are partially or completely inhibited by the undissociated form of acetic, benzoic, citric, lactic, propionic and sorbic acids. Salts such as sodium chloride, potassium chloride and sodium nitrate, at low concentrations, can enhance aflatoxin production. At higher concentrations they become inhibitory, but marked inhibition requires amounts of the salts greater than are commonly used in foods. Phenolic antioxidants, sometimes added to foods to prevent oxidative deterioration, also are inhibitory to toxigenic aspergilli. Other inhibitory a

Effect of protein calorie malnutrition and cell replication on aflatoxin B1-induced hepatocarcinogenesis in rats.

Malnourished and well-fed neonatal Holtzman rats 10 days of age were exposed to 3 doses of aflatoxin B1 [(AFB1) CAS: 1162-65-8] at intervals of 96 hours to study the combined effect of malnutrition and cell replication in AFB1-induced hepato-carcinogenesis. The neonatal model made use of the fact that cell replication persists in the liver for 3 weeks of postnatal life. Malnutrition during suckling was induced by adopting the techniques of Widdowson and McCance of increasing the litter size to 16. Following AFB1 administration, the malnourished animals were rehabilitated on a high-protein pell

Carcinogenicity of aflatoxin B1 in rhesus monkeys: two additional cases of primary liver cancer.

Three of 42 (7%) monkeys given aflatoxin B1 (AFB1) for longer than 2 years have developed primary malignant neoplasms of the liver. Liver biopsies performed at intervals during aflatoxin administration revealed that neoplasia was preceded by pathologic lesions of the liver, including toxic hepatitis, proliferation of pseudotubules, and hyperplastic nodules. Serum alpha-fetoprotein levels, monitored in one of the monkeys by radioimmunoassay, paralleled tumor growth and recurrence of the hepatocellular carcinoma. Normal serum alpha-fetoprotein levels were noted for a monkey with hemangioendothel

Induction of mutations in DNA-repair deficient bacteria by a liver microsomal metabolite of aflatoxin B1.

Certain strains of Salmonella typhimurium and Escherichia coli, particularly those which are very sensitive to u.v. light, are killed when incubated with rat liver mixed function oxidases and aflatoxin B(1). UvrA or recA strains of E. coli are more susceptible than the wild-type strain, while the double mutant uvrA recA is the most sensitive strain yet tested. The aflatoxin B(1) metabolite is also able to induce reverse mutations in 2 histidine auxotrophic strains of S. typhimurium, one strain of which is reverted specifically by frame shift mutagens and the other by agents inducing base pair

Integrative network toxicology and computational modeling uncover the mechanism of aflatoxin B1-induced nephrotoxicity.

Aflatoxin B1 (AFB1) is a highly toxic mycotoxin that causes severe renal injury, yet the molecular basis of its nephrotoxicity remains poorly defined. Here, we integrated multi-omics data with computational modeling to elucidate the mechanisms of AFB1-induced kidney damage. Three human renal transplant biopsy microarray datasets (GSE1563, GSE30718, and GSE61739), comprising 121 injury samples and 84 control samples and representing distinct forms of transplant-associated renal injury, including acute rejection and ischemic kidney injury, were analyzed using differential expression and weighted

Ratiometric Fluorescence Sensing of Aflatoxin B1 Using a Cationic Perylene Probe and Rhodamine B.

As one of the most toxic and prevalent mycotoxins, aflatoxin B1 (AFB1) poses a persistent threat to food safety and public health due to its widespread contamination of cereal commodities and its well-documented carcinogenic, genotoxic, and hepatotoxic effects. Although fluorescence sensing provides a rapid approach to AFB1 screening, most existing fluorescence strategies rely on antibodies or aptamers, which are costly, laborious, and susceptible to environmental degradation; moreover, single-emission readouts remain vulnerable to variations in probe concentration, excitation conditions, and

Fluorescence resonance energy transfer-based aptasensor for rapid detection of aflatoxin M1 in milk.

A novel aptasensor for the detection of aflatoxin M1 (AFM1) was developed based on the fluorescence resonance energy transfer (FRET) effect between gold nanoparticles (AuNPs) and FAM-labeled cDNA (FAM-cDNA). Additionally, magnetic nanoparticles (Fe3O4 NPs) were attached with AuNPs to construct a Fe3O4@AuNPs system that was easy to separate by the external magnetic field. In this aptasensor, FAM-labeled cDNA and Fe3O4@AuNPs acted as donor and acceptor, respectively. In the presence of target AFM1, the aptamer of this aptasensor specifically recognized with AFM1, resulting in a turn-on fluoresce

Prenatal Aflatoxin B1 Exposure: A Review of Pathogenesis and Impact on Fetal Skeletal Development and Ossification.

Prenatal exposure to aflatoxin B1 (AFB1) poses a significant risk to fetal development and is associated with reduced birth weight in humans. Experimental studies consistently show that AFB1 induces fetal abnormalities, with skeletal malformations and ossification defects being the most common. However, the specific impact of AFB1 on fetal osteogenesis remains unclear. Given this knowledge gap, this study aimed to review the existing literature concerning the pathogenesis of AFB1 and its potential influence on bone development. The primary mechanisms implicated in AFB1's impact on bone include

Genome-Guided Discovery of Enterobacter Multicopper Oxidases Enabling Enzymatic Detoxification of Aflatoxin B1.

Aflatoxin B1 is a potent Group I carcinogen that contaminates food and feed, necessitating efficient detoxification strategies. This study isolated a novel bacterial strain, Enterobacter sp. HNGD-822 from soil, which efficiently degrades AFB1. Genome analysis identified two novel multicopper oxidase (laccase) genes, EbMCO1 and EbMCO2. The recombinant enzymes were heterologously expressed, exhibiting optimal activity at 50 °C and pH 7.0, with strict Cu2+ dependence. Within 12 h, EbMCO1 degraded over 99% of AFB1, while EbMCO2 also showed high activity under the same conditions. Both enzymes prim

Bacillus velezensis-derived laccase and superoxide dismutase detoxify aflatoxin B1, zearalenone, and alternariol.

Aflatoxin B1(AFB1), zearalenone(ZEN), and alternariol(AOH) are major mycotoxins that threaten food safety, yet enzymes capable of detoxifying these structurally diverse toxins remain limited. In this study, two enzymes from Bacillus velezensis HNGD-JQ06, laccase BVLac and superoxide dismutase BV2SOD, were identified as broad-spectrum detoxification candidates. BVLac degraded 93.33% of AFB1 at 60 °C and pH 7.0, 94.10% of ZEN at 60 °C and pH 8.0, and 96.87% of AOH at 50 °C and pH 8.0. BV2SOD degraded 92.59% of AFB1 and 91.99% of ZEN at 50 °C and pH 7.0, and 98.94% of AOH at 60 °C and pH 8.0. UHP

Aflatoxin B1 Exposure and Hepatocellular Carcinoma in South America: A Multinational Cross-Sectional Analysis.

Aflatoxin B1 (AFB1), a dietary mycotoxin classified as a Group 1 carcinogen and a risk factor for hepatocellular carcinoma (HCC), has seen limited biomarker-based evidence linking it to HCC in South America despite favorable regional contamination conditions. Utilizing a newly validated isotope-dilution HPLC-MS/MS method to quantify AFB1-Lysine (AFB1-Lys) adducts, we conducted a cross-sectional study involving 92 HCC patients and 70 healthy controls across six South American nations. The primary case-control analysis, focusing on 64 HCC patients and 70 controls from Argentina and Colombia, rev

Aflatoxin Mixture-Driven Intrahepatic Cholangiocarcinoma in Rats Involving G1/S Checkpoint Dysregulation.

Aflatoxins (AFs) are potent hepatotropic mycotoxins-AFB1 being the best-characterized-yet their ability to induce intrahepatic cholangiocarcinoma (iCCA) remains underexplored. Male Wistar rats received vehicle (controls; n = 5) or an AFB1-dominant AF mixture (AFB1 39.46 μg/mL; AFB2 1.13 μg/mL; AFG1 17.44 μg/mL; AFG2 0.59 μg/mL- n = 10) by daily gavage for 90 days, at a dose equivalent to 400 μg AFB1 per kg of diet. After 12 months, twelve iCCA tumors were resected and analyzed by histology (H&E) and tissue-microarray-based immunohistochemistry (Cytokeratin-19, Hep Par-1, p53, Cyclin D1, Rb, β-…

Exposure of pregnant and lactating parental mice to aflatoxin B1 promotes hepatotoxicity in offspring mice.

Aflatoxin B1 (AFB1) taints feeds stuffs, endangering livestock's health and resulting in the liver and breast damage. At the same time, while breastfeeding, AFB1 crosses the mammary glands and enters the milk, harming the offspring. This study investigated the liver damaging effects of maternal AFB1 exposure during pregnancy and lactation in offspring mice. The livers of 8-day-old offspring mice were obtained from female mice who were administered AFB1 (2 mg/kg) 1 week prior to and 1 week following birth. The results showed that AFB1 increased the levels of malondialdehyde (MDA), alanine amino

Aflatoxin exposure during pregnancy or infancy and its effect on infant health: a narrative review.

Pregnant women are exposed to various contaminants through foods, with environmental toxicants and aflatoxin (AF) being among the major food contaminants. Therefore, this review was conducted for a better perspective on the AF exposure during pregnancy or infancy, highlighting how exposure through the mother (via placenta and breast milk) and directly through infant foods ultimately affects infant health. The literature suggests that AF exposure during pregnancy may lead to maternal anaemia, premature delivery, pregnancy loss or decreased number of live births. AF crosses through the placenta
