Residual alcohol use disorder symptoms after treatment predict long-term drinking outcomes in seniors with DSM-5 alcohol use disorder.

BACKGROUND: Risk of relapse within the first months after alcohol use disorder (AUD) interventions is substantial among older adults. For this vulnerable group, little information exists on how this risk is associated with residual DSM-5 AUD symptoms after treatment. AIMS: To investigate among older adults who received short-term treatment for DSM-5 AUD (1) the prediction of drinking behaviors and quality of life 12 months after treatment initiation by 6-month DSM-5 AUD symptoms, AUD severity, and AUD remission, and (2) whether these DSM-5 AUD indicators provide prognostic information beyond

KLB is associated with alcohol drinking, and its gene product β-Klotho is necessary for FGF21 regulation of alcohol preference.

Excessive alcohol consumption is a major public health problem worldwide. Although drinking habits are known to be inherited, few genes have been identified that are robustly linked to alcohol drinking. We conducted a genome-wide association metaanalysis and replication study among >105,000 individuals of European ancestry and identified β-Klotho (KLB) as a locus associated with alcohol consumption (rs11940694; P = 9.2 × 10-12). β-Klotho is an obligate coreceptor for the hormone FGF21, which is secreted from the liver and implicated in macronutrient preference in humans. We show that brain-spe…

Effects of CRF1-receptor and opioid-receptor antagonists on dependence-induced increases in alcohol drinking by alcohol-preferring (P) rats.

BACKGROUND: Selective breeding of rats over generations and induction of alcohol dependence via chronic vapor inhalation both enhance alcohol consumption in animal models. The purpose of this study was to determine whether dependence-induced increases in alcohol consumption by P rats is sensitive to naltrexone, a general opioid receptor antagonist (but with highest affinity at the mu-opioid receptor at low doses), and the recently characterized small molecule CRF(1)-receptor antagonist MPZP (N,N-bis(2-methoxyethyl)-3-(4-methoxy-2-methylphenyl)-2,5-dimethyl-pyrazolo[1,5-a]pyrimidin-7-amine). Polygenic risk score for problematic alcohol use predicts heavy drinking and alcohol use disorder symptoms in young adulthood after accounting for adolescent alcohol use and parental alcohol use disorder.

BACKGROUND: Genome-wide association studies (GWAS) have identified hundreds of loci associated with alcohol-related traits. GWAS permit the calculation of polygenic risk scores (PRS), which aggregate genetic risk for a trait across the genome. To evaluate the usefulness of a PRS for problematic alcohol use (PAU)-which subsumes alcohol use disorder (AUD) and alcohol-related problems-we tested whether this PRS predicted heavy drinking and alcohol problems after accounting for family history of AUD and prior drinking history, robust and established predictors of PAU. METHODS: Participants (N=665

Alcohol and educational inequalities: Hazardous drinking prevalence and all-cause mortality by hazardous drinking group in people aged 50 and older in Europe.

BACKGROUND: We examined educational inequalities in hazardous drinking prevalence among individuals aged 50 or more in 14 European countries, and explored educational inequalities in mortality in hazardous drinkers in European regions. UNLABELLED: Methods: We analyzed data from waves 4, 5 and 6 of the Survey of Health Ageing and Retirement in Europe (SHARE). We estimated age-standardized hazardous drinking prevalence, and prevalence ratios (PR) of hazardous drinking by country and educational level using Poisson regression models with robust variance. We estimated the relative index of inequa

Subgroups of Young Sexual Minority Women Based on Drinking Locations and Companions and Links With Alcohol Consequences, Drinking Motives, and LGBTQ-Related Constructs.

OBJECTIVE: Sexual minority women (SMW; e.g., lesbians, bisexual women) are at increased risk for alcohol use disorders and related problems compared with heterosexual women. However, little is known about the social context in which drinking occurs in this high-risk population. This study used latent class analysis to identify subgroups of SMW based on drinking locations and companions and examined whether class membership was associated with consequences, drinking motives, and LGBTQ-related constructs (e.g., outness, discrimination). METHOD: A sample of 670 SMW reported on alcohol use, drink

The way one thinks affects the way one drinks: subjective evaluations of alcohol consequences predict subsequent change in drinking behavior.

Heavy alcohol use and related consequences are common during the college years and are associated with deleterious outcomes for both the students themselves and the college community. Some college students make self-initiated changes to their drinking to avoid such outcomes, but little is known about how such adjustments occur, or characteristics that are associated with making these adjustments. Based on Social Learning Theory (SLT), one cognitive factor that may predict within-person changes in drinking is the subjective evaluations of alcohol consequences (i.e., the extent to which conseque

Higher drinking frequency corresponds to lower synaptic density in people with alcohol use disorder.

BACKGROUNDChronic alcohol use leads to synaptic dysfunction in preclinical studies. However, whether in vivo synaptic density deficits are found in people with alcohol use disorder (AUD) remains unclear.METHODSThirty-two people with AUD (n = 17 women; n = 15 men) and 29 control participants (n = 17 women; n = 12 men)completed 1 PET brain imaging scan with the radiotracer [11C]UCB-J, which binds to SV2A, a marker of synaptic density. The levels of synaptic density were quantified by estimating the nondisplaceable binding potential (BPND) across 4 regions of interest: frontal cortex, striatum, h

When alcohol fills the gap: Basic psychological need satisfaction and frustration predict drinking patterns.

OBJECTIVE: Test whether perceived basic psychological need satisfaction from alcohol interacts with basic psychological need frustration to predict hazardous/harmful drinking consistent with self-determination theory's need-density hypothesis. METHOD: Study 1 was a cross-sectional study (742 U.S. adults) that showed effects of perceived need satisfaction from alcohol and general need frustration on hazardous/harmful drinking (Alcohol Use Disorders Identification Test). Study 2 was a preregistered 5-day diary study (baseline and four daily surveys; n = 786, 3,358 observations) that assessed ef

Dorsomedial Striatum Calcium Permeable AMPA Receptors in the Development of Aversion-Resistant Alcohol Drinking.

RATIONALE: Alcohol use disorder is defined by drinking alcohol despite knowledge of negative consequences, often referred to as aversion-resistant drinking (ARD). The dorsomedial (DMS) and dorsolateral striatum (DLS) are necessary for goal-directed and habitual action selection, respectively. Leading hypotheses posit that once drug use becomes compulsive, DMS dependence degrades while DLS dependence increases. This shift may be mediated by changes in synaptic weights from glutamatergic inputs. OBJECTIVES: Using a combination of western-blot, micro-injections, and ex-vivo electrophysiology, we

Automated Operant Intermittent Alcohol Drinking In Socially-Housed Mice via The Intellicage Testing System.

Despite known confounds of social isolation on rodent behavior, many investigators rely on singly housing mice to assess alcohol preference in preclinical models of alcohol use disorder (AUD). This protocol describes a social alcohol drinking task that allows for high-throughput, automated assessment of alcohol drinking behaviors in mice without the confounding impacts of stress induced by social isolation. The IntelliCage testing system enables investigators to simultaneously assess operant alcohol drinking behavior in up to 16 same-sex mice, each uniquely identified with a subcutaneously imp…

Alpha1 adrenergic receptors integrate across multiple separable heart rate variability metrics during alcohol drinking.

Alcohol Use Disorder remains a major challenge and is now a leading cause of death in adults. Biomarkers like heart rate variability (HRV) could give important and novel insights into how pharmacotherapies counteract excessive drive for alcohol, including those promoted by adrenergic receptors (AR). Indeed, α1AR inhibition helps heavy drinkers with greater withdrawal-related mood disruption and arousal, while α1ARs more generally are implicated in cognitive flexibility. To uncover how α1ARs might regulate drinking-related arousal, we used cardiovascular telemetry in adult male and female rats.

Associations of alcohol drinking with incident dementia: a prospective study from the UK Biobank.

Relationship between alcohol drinking and incident dementia remained uncertain. This study used UK Biobank cohort data to investigate the association between alcohol drinking and dementia risk, and potential effect modifications by cardiovascular disease (CVD) risk, APOE4 gene, and sex. We excluded infrequent drinkers and participants with baseline dementia or dementia within two years of follow-up. Drinking status was defined as non-drinking, low-moderate and heavy drinking (by weekly alcohol units). Drinking behaviors included drinking with meals and drinking type. Primary outcome was all-ca…

Factors that increase risk for alcohol-induced blackouts in high-intensity drinking young adults.

BACKGROUND: Alcohol-induced blackouts are prospectively associated with negative drinking outcomes. While typically requiring heavy drinking, blackouts are not reported on all heavy drinking events or by all individuals who drink heavily. This study extends previous research by identifying the young adults most likely to experience blackouts assessed prospectively. Hypotheses focused on previously supported (female sex, White race, younger age, family history of alcohol problems, lower subjective response to alcohol, and higher tolerance) and novel predictors (possible traumatic brain injury;

A hidden cost of drinking: Alcohol use and gendered inequalities in unpaid care work.

The impact of alcohol use on families is an important, yet frequently understudied, avenue of research. Unpaid care work, which encompasses childcare, eldercare, cooking, and cleaning, and alcohol use, are both gendered. Men are more likely to consume alcohol, binge drink, and have alcohol use disorder and women do more unpaid care work in families. Given the gendered nature of both, this study aims to assess if alcohol consumption impacts the distribution of unpaid care work in families by gender. Using the Panel Study of Income Dynamics (PSID), a longitudinal dataset, this paper analyzes how

Predictors of high- and low-risk drinking after group treatment for alcohol use disorder.

BACKGROUND: Understanding the factors influencing alcohol use disorder (AUD) treatment outcomes is essential. More knowledge about patient characteristics that predict treatment outcomes can help personalise interventions, improve treatment planning and address the needs of specific subgroups. The frequency of treatment attendance may also affect drinking outcomes after treatment. Despite research efforts, uncertainty remains about how patient factors and treatment attendance influence treatment outcomes. AIMS: To examine how patient factors and treatment attendance predict high- or low-risk

Impact of alcohol strength on attitudes and decisions concerning special occasion drinking during pregnancy.

BACKGROUND: Special occasions are a risk factor for drinking during pregnancy. This study determined the impact of alcohol strength on attitudes around, and perceived harm of, drinking in pregnancy. If perceived harm decreases with lower strength alcohol, this may promote drinking when abstinence is recommended. METHODS: Two online 'special occasion' vignette studies randomised female participants to one of three drink strength conditions (11%, 7.5%, 0% alcohol beverage volume [ABV]). In the study (N = 1128), participants were asked to imagine themselves or someone else choosing to consume th

Sex Differences in Mouse Models of Voluntary Alcohol Drinking and Abstinence-Induced Negative Emotion.

Alcohol Use Disorder (AUD) is a growing problem worldwide, causing an incredible burden on health and the economy. Though AUD impacts people of all backgrounds and demographics, increasing evidence has suggested robust sex differences in alcohol drinking patterns and AUD-induced negative emotionality or hyperkatifeia. Rates of problematic drinking have significantly risen among women, and women face more severe negative emotional consequences in abstinence such as increased risk of comorbidity with an anxiety or mood disorder and more severe symptoms of depression. As such, a bevy of preclinic…
