Direct Access to Iron Carbenes from Aldehyde, Ketone, and Formamide Feedstocks.

Metallocarbenes are powerful intermediates for constructing complex molecular architectures, yet their generation typically relies on hazardous diazo or ylide precursors. We report photocatalytic and electrochemical strategies for direct carbene formation from unmodified carbonyl compounds using a low-valent iron system. The reaction proceeds through net oxidative addition of Fe(I) into the carbonyl bond, followed by α-protonation and α-elimination to generate a reactive Fe-carbene intermediate. This platform enables cyclopropanation of diverse alkenes─including complex, drug-like, and amino-a

[Aldehyde dehydrogenases as therapeutic targets for oxidative stress management in pathological processes].

Cell damage due to lipid aldehydes accumulation has been associated with different chronic diseases such as atherosclerosis, myocardial infarction, kidney disease, and some types of cancer. Therefore, there is a general interest in developing new strategies to deal with aldehyde accumulation. Aldehyde dehydrogenases (ALDH) are a group of enzymes that catalyze the oxidation of lipid aldehydes to carboxylic acids, avoiding the toxic effects of these molecules. Due to their relevant role in cellular aldehydes detoxification and the pathogenesis and progression of different diseases, various thera

Stereoelectronically Controlled Regioselective Allylation of Aldehyde Under Photoredox/Chromium Catalysis.

The formation of structurally defined α,γ-disubstituted allylmetallic intermediate which could undergo regioselective allylation with carbonyl compounds remains a formidable challenge in synthetic chemistry. Although regioselective in situ formation of α,γ-disubstituted allylchromium intermediate controlled by coordinative directing groups has been achieved, how to achieve high selectivity for reactions involving α,γ-disubstituted allylic chromium intermediates through electronic control has been elusive. In this study, we report a silyl-group-directed strategy that exploits the α-silicon effe

Post-mortem degradation of flunitrazepam by aldehyde oxidase 1 and hemoglobin in blood.

PURPOSE: Flunitrazepam is a widely used hypnotic agent detected in many fatal intoxication and suicide cases. However, its concentration in blood during autopsy differs from that at the time of death; it is either undetected or detected at lower concentrations than expected in post-mortem blood. In this study, we evaluated interactions between flunitrazepam and human blood using liquid chromatography coupled with quadrupole-Orbitrap mass spectrometry (LC-Q-Orbitrap-MS). METHODS: We measured flunitrazepam, 7-aminoflunitrazepam via reduction by aldehyde oxidase 1 (AOX1), and the degradation pro

N -aldehyde modified phosphatidylethanolamines formed by lipid peroxidation inhibit efferocytosis.

BACKGROUND: Impairments in efferocytosis, the phagocytic clearance of apoptotic cells, contribute to the development of chronic inflammation and promote the formation of large necrotic cores within atherosclerotic plaques. A better understanding of the factors that impair efferocytosis could lead to improved interventions. Many chronic inflammatory conditions feature excessive lipid peroxidation, which generates reactive lipid aldehydes that can potentially form N -aldehyde-modified phosphatidylethanolamines (NAMPs). We therefore investigated the extent to which levels of NAMPs are elevated un

Inhibiting Aldehyde Dehydrogenase Function Using WIN 18,446 to Synchronize Spermatogenesis.

While beneficial to the individual, the asynchronous progression of spermatogenesis makes the evaluation of stage-specific or cell-specific function within the testes challenging for researchers. Here, we describe a method using the aldehyde dehydrogenase inhibitor WIN 18,446 to halt all-trans retinoic acid (atRA) synthesis and prevent the commitment of undifferentiated spermatogonia to meiosis. Following treatment with exogenous atRA after WIN 18,446, individual stages of spermatogenesis can be analyzed with high specificity and this method serves as an invaluable tool to study the progressio

Anti-infective therapy of inhibiting Staphylococcus aureus ClpP by protocatechuic aldehyde.

The increasing antibiotic resistance and widespread pathogenicity of Staphylococcus aureus (S. aureus) has resulted in severe infections in both human and animals, necessitating the urgent development of novel therapeutic agents. Targeting virulence factors has emerged as a promising approach to combat infections caused by drug-resistant bacteria. Protocatechuic aldehyde (PCA) was identified as an inhibitor of S. aureus ClpP through an enzyme activity inhibition assay. Treatment with PCA at concentrations of 64 and 128 μg/mL significantly reduced the proteolytic and hemolytic activities of S.

Aldehyde metabolism governs resilience of mucociliary clearance to air pollution exposure.

Air pollution is a serious environmental threat to public health; however, the molecular basis underlying its detrimental effects on respiratory fitness remains poorly understood. Here, we showed that exposure to particulate matter ≤ 2.5 μm (PM2.5), a substantial fraction of air pollutants, induced the generation of reactive aldehyde species in the airway. We identified aldehyde dehydrogenase 1A1 (ALDH1A1), which was selectively expressed in airway epithelium, as an enzyme responsible for detoxifying these reactive aldehyde species. Loss of ALDH1A1 function resulted in the accumulation of alde

Aldehydic load as an objective imaging biomarker of mild traumatic brain injury.

Mild traumatic brain injury (mTBI) is neurological impairment induced by biomechanical forces without structural brain damage, currently without an objective diagnostic tool. Downstream injury stems from oxidative damage leading to the production of neurotoxic aldehydes. A collagen-based 3D corticomimetic in vitro model of concussion was developed, confirming aldehyde production following impact. Total aldehyde levels were mapped in vivo following mTBI using a novel CEST-MRI contrast agent, ProxyNA3, in a new model of closed-head, awake, single-impact concussion in aged and young mice with ald

Aldehyde-induced DNA-protein crosslinks- DNA damage, repair and mutagenesis.

Aldehyde exposure has been shown to lead to the formation of DNA damage comprising of DNA-protein crosslinks (DPCs), base adducts and interstrand or intrastrand crosslinks. DPCs have recently drawn more attention because of recent advances in detection and quantification of these adducts. DPCs are highly deleterious to genome stability and have been shown to block replication forks, leading to wide-spread mutagenesis. Cellular mechanisms to prevent DPC-induced damage include excision repair pathways, homologous recombination, and specialized proteases involved in cleaving the covalently bound

Contribution of aldehyde oxidase to methotrexate-induced hepatotoxicity: in vitro and pharmacoepidemiological approaches.

BACKGROUND: Methotrexate (MTX) is partially metabolized by aldehyde oxidase (AOX) in the liver and its clinical impact remains unclear. In this study, we aimed to demonstrate how AOX contributes to MTX-induced hepatotoxicity in vitro and clarify the relationship between concomitant AOX inhibitor use and MTX-associated liver injury development using the U.S. Food and Drug Administration Adverse Event Reporting System (FAERS). METHODS: We assessed intracellular MTX accumulation and cytotoxicity using HepG2 cells. We used the FAERS database to detect reporting odds ratio (ROR)-based MTX-related

Endothelial Cell Protein Targeting by Myeloperoxidase-Derived 2-Chlorofatty Aldehyde.

Neutrophils are important cellular mediators of injury and repair in diseases including ischemic heart disease, atherosclerosis, and sepsis. Myeloperoxidase-derived (MPO)-oxidants released from neutrophils are potential mediators of endothelial injury in disease. MPO-derived HOCl attacks plasmalogen phospholipid to liberate 2-chlorofatty aldehyde (2-ClFALD). Both 2-ClFALD and its oxidation product, 2-chlorofatty acid (2-ClFA), are electrophilic lipids, and both probably react with proteins through several mechanisms. In the present study, we investigate protein modification specifically by 2-C

Effects of intracardiac delivery of aldehyde dehydrogenase 2 gene in myocardial salvage.

Intrinsic activity of aldehyde dehydrogenase (ALDH)2, a cardiac mitochondrial enzyme, is vital in detoxifying 4-hydroxy-2-nonenal (4HNE) like cellular reactive carbonyl species (RCS) and thereby conferring cardiac protection against pathological stress. It was also known that a single point mutation (E487K) in ALDH2 (prevalent in East Asians) known as ALDH2*2 reduces its activity intrinsically and was associated with increased cardiovascular diseases. We and others have shown that ALDH2 activity is reduced in several pathologies in WT animals as well. Thus, exogenous augmentation of ALDH2 acti

Activation of Aldehyde Dehydrogenase 2 Ameliorates Glucolipotoxicity of Pancreatic Beta Cells.

Chronic hyperglycemia and hyperlipidemia hamper beta cell function, leading to glucolipotoxicity. Mitochondrial aldehyde dehydrogenase 2 (ALDH2) detoxifies reactive aldehydes, such as methylglyoxal (MG) and 4-hydroxynonenal (4-HNE), derived from glucose and lipids, respectively. We aimed to investigate whether ALDH2 activators ameliorated beta cell dysfunction and apoptosis induced by glucolipotoxicity, and its potential mechanisms of action. Glucose-stimulated insulin secretion (GSIS) in MIN6 cells and insulin secretion from isolated islets in perifusion experiments were measured. The intrace

Aldehyde to Ketone Homologation Enabled by Improved Access to Thioalkyl Phosphonium Salts.

Phosphines were previously unusable as Pummerer-type nucleophiles due to competing redox chemistry with sulfoxides. Here we circumvent this problem to achieve a formal phosphine Pummerer reaction that offers thioalkyl phosphonium salts that, in turn, give rise to diverse vinyl sulfides via Wittig olefinations. Thirty vinyl sulfides are thus prepared from (alkylthioalkyl)triphenyl phosphonium salts and aldehydes. The hydrolysis of these vinyl sulfides offers an efficient and versatile two-step one-carbon homologation of aldehydes to ketones.

Sequential Post-Polymerization Modification of Aldehyde Polymers to Ketone and Oxime Polymers.

A new sequential post-polymerization modification route has been developed for the synthesis of multifunctional polymers from a simple aldehyde polymer. In the first modification step, a template polymer derived from the radical polymerization of 4-vinyl benzaldehyde undergoes Rh-catalyzed hydroacylation with alkenes to furnish a group of ketone polymers. In the second modification step, Schiff base formation with alkoxy ammonium salts introduces a second group-an oxime functionality. Both the steps are highly efficient, introducing evenly distributed dual functionalities at the same position.

Effect of the drug cyclophosphamide on the activity of porcine kidney betaine aldehyde dehydrogenase.

The enzyme betaine aldehyde dehydrogenase (BADH EC 1.2.1.8) catalyzes the synthesis of glycine betaine (GB), an osmolyte and osmoprotectant. Also, it participates in several metabolic pathways in humans. All BADHs known have cysteine in the active site involved in the aldehyde binding, whereas the porcine kidney enzyme (pkBADH) also has a neighborhood cysteine, both sensitive to oxidation. The antineoplastic and immuno-suppressant pre-drug cyclophosphamide (CTX), and its bioactivation products, have two highly oxidating chlorine atoms. This work aimed to analyze the effect of CTX in the activi

Aldehyde-driven transcriptional stress triggers an anorexic DNA damage response.

Endogenous DNA damage can perturb transcription, triggering a multifaceted cellular response that repairs the damage, degrades RNA polymerase II and shuts down global transcription1-4. This response is absent in the human disease Cockayne syndrome, which is caused by loss of the Cockayne syndrome A (CSA) or CSB proteins5-7. However, the source of endogenous DNA damage and how this leads to the prominent degenerative features of this disease remain unknown. Here we find that endogenous formaldehyde impedes transcription, with marked physiological consequences. Mice deficient in formaldehyde cle

Determining the impact of flavored e-liquids on aldehyde production during Vaping.

1,2-Propylene glycol and glycerin, principal components of e-liquids, can thermally degrade to form acetaldehyde, acrolein, and formaldehyde when heated in electronic nicotine delivery systems (ENDS). Recently the flavors in e-liquids were suggested to be the major source of these aldehydes. We used the same 10 ENDS devices to test 5 e-liquid formulations (four flavored & one corresponding non-flavored) and measured device mass loss and levels of acetaldehyde, acrolein, and formaldehyde (30 replicate measurements per formulation). Despite finding reasonable variability in measurements of devic
