The role of alkaloids in Ipomoea carnea toxicosis: a study in rats.

Ipomoea carnea promotes in livestock a toxicosis histologically characterized by vacuolated cells in different organs. The toxic principles of I. carnea are the alkaloids swainsonine and calystegines B1, B2, B3 and Cl. However, it has not been determined whether the effects observed in rats treated with this plant are only due to swainsonine or if the calystegines have some additive toxic effect. Thus, the aim of the present study was to evaluate in rats the toxic effects of the L carnea aqueous fraction (AF) and of its different alkaloids when administered individually at the same concentrati…

Evidence for the monophyletic evolution of benzylisoquinoline alkaloid biosynthesis in angiosperms.

Benzylisoquinoline alkaloids (BIAs) consist of more than 2500 diverse structures largely restricted to the order Ranunculales and the eumagnoliids. However, BIAs also occur in the Rutaceae, Lauraceae, Cornaceae and Nelumbonaceae, and sporadically throughout the order Piperales. Several of these alkaloids function in the defense of plants against herbivores and pathogens--thus the capacity for BIA biosynthesis is expected to play an important role in the reproductive fitness of certain plants. Biochemical and molecular phylogenetic approaches were used to investigate the evolution of BIA biosyn…

Alkaloid delta agonist BW373U86 increases hypoxic tolerance.

Activation of delta opioid receptors increases survival time during acute, lethal hypoxia in mice. delta Agonists therefore present a promising avenue for therapeutic application to reduce the morbidity and mortality associated with clinical hypoxia in settings such as drowning, head injury apnea, and complicated childbirths. However, most delta agonists now available are peptides, and may have limited clinical utility. In the present study, we evaluate the neuroprotective ability of an alkaloid delta agonist, BW373U86. Alkaloid compounds, due to increased stability and increased systemic dist

Uterine rupture in association with alkaloidal ("crack") cocaine abuse.

The use of alkaloidal ("crack") cocaine in pregnancy can result in systemic and focal vasoconstriction and abnormal uterine contractions forceful enough to cause the complete rupture of a gravid uterus along a previous vertical cesarean section scar. A 43-year-old woman was admitted to the emergency department with acute abdominal pain at 34 weeks' gestation. She had had a low vertical cesarean section 3 years previously for breech presentation with acute fetal distress at 28 weeks' gestation. The patient had a history of heavy "crack" abuse and admitted to inhaling "crack" throughout the day

Cerebrovascular complications of the use of the "crack" form of alkaloidal cocaine.

BACKGROUND AND METHODS: The use of cocaine, especially one of its alkaloidal forms ("crack"), has been increasingly associated with cerebrovascular disease. To clarify the clinical, radiologic, and pathological features of the events associated with cocaine use, we identified 28 patients at four medical centers who had stroke temporally related to the use of alkaloidal cocaine (during or within 72 hours of use). RESULTS: The 28 patients had the following types of cerebrovascular event: cerebral infarction (n = 18 [2 hemorrhagic; 1 fatal]) in the areas supplied by the middle cerebral artery (n

Discriminative stimulus properties of (+)cathine, an alkaloid of the khat plant.

The effects of the psychostimulant (+)cathine (norpseudoephedrine) were examined in a two-choice, food-motivated, drug-discrimination paradigm. Rats were able to discriminate cathine from vehicle and this effect was dose- and time-dependent. Prior administration of cathine resulted in a diminished response (tolerance) to subsequent cathine and this effect developed and dissipated rapidly. Thus, different dose-response curves were generated depending upon whether cathine or vehicle was administered the day before testing. The development of tolerance also shortened cathine's time course of acti

Antiamoebic activity of chonemorphine, a steroidal alkaloid, in experimental models.

Chonemorphine, a steroidal alkaloid isolated from Chonemorpha fragrans Moon (Apocyanaceae) was identified as an antimoebic principle during the course of a screening programme for novel antiparasitic agents from plant sources. At a dosage of 100 mg/kg x 4 chonemorphine led to a 100% cure of experimental hepatic infection in golden hamsters and cleared 90% of the intestinal infection in weanling Wistar rats at 200 mg/kg (x4) dosages. The discovery of chonemorphine as an antiamoebic agent is an addition to the few known plant amoebicides such as emetine and conessine.

Some toxic properties of a carcinogenic pyrrolizidine alkaloid, petasitenine.

Some toxic properties of petasitenine, a hepatocarcinogenic pyrrolizidine alkaloid, were confirmed in 3 parts of studies. Electron microscopic observation of rat liver on the stage of acute toxicity following administration of a toxic dose of petasitenine disclosed the distinctive changes i.e., nucleolar segregation, degradation of endoplasmic reticulum such as formation of concentric whorls in the liver cells. Biochemical test for the mitochondrial function in the isolated mitochondria from rat liver cells revealed on inhibitory effects of petasitenine in the respiratory system, indicating th

The recovery of alkaloids after their location on thin layer chromatograms.

A method was developed for the recovery of 29 alkaloids from TLC plates after they had been complexed with either potassium iodoplatinate (for tertiary amines) or carbon disulphide (for primary and secondary amines). A comparison was made between the percentage recoveries of these alkaloids from 5 different types of TLC plate. In addition, the stability of these alkaloids on TLC plates over a 7 day period was studied. Assays were, where possible, carried out by UV spectrophotometry but gas chromatography was used for alkaloids which possessed low UV absorbtivity values. All the alkaloids extra…

Cisplatin-synergistic monoterpene indole alkaloids with diverse scaffolds from Rauvolfia vomitoria.

Twelve previously unreported monoterpene indole alkaloids (MIAs), rauvotines A - L (1-12), and thirteen known analogs (13-25), were obtained from Rauvolfia vomitoria. These MIAs were categorized as ajmaline (1,2,13), suaveoline (3, 4, 14, 15), peraksine (5, 6, 16, 17), yohimbine oxindole (7-9, 18), and yohimbine (10-12, 19-25) frameworks, respectively. Structurally, 3 and 4 constituted the first suaveoline-type MIAs characterized by a trans-fused 1H-pyrano[3,4-c]pyridine ring system. 7 represented the first natural yohimbine oxindole MIA with a 7R configuration. Compounds 10-12 were identified…

Biosynthetic mechanism and ecological function of chirality in Solanum steroidal alkaloids.

Chirality fundamentally influences the bioactivity and ecological roles of plant natural products. Steroidal glycoalkaloids (SGAs) in tomato and wild relatives exhibit this stereochemical variation. Forward genetics identifies C26 hydroxylase GAME8 as the key enzyme controlling SGA chirality. Cultivated tomato harbors two identical GAME8 copies yielding S-configured intermediates, whereas Solanum pennellii homologs GAME8a and GAME8b dominantly produce R- and S-configured products, respectively. Site-saturation mutagenesis and molecular docking reveal four residues sufficient to switch this chi…

Pyridine alkaloids possess important biological activities and are widely used in fields such as medicine and pesticides. This paper comprehensively reviews the research progress in the chemical synthesis and biosynthesis of pyridine alkaloids. In terms of chemical synthesis, there are diverse synthesis methods for arylpyridine compounds. For example, 2,4,6-triarylpyridine can be synthesized by using iron-organic framework materials or other reagents. The 3-ethylsulfone pyridine compounds with aryltriazole structures can be synthesized through specific reactions. And 2- arylpyridine can also b…

Photo-affinity and Metabolic Labeling Probes Based on the Opioid Alkaloids.

The opioids are powerful analgesics yet possess contingencies that can lead to opioid-use disorder. Chemical probes derived from the opioid alkaloids can provide deeper insight into the molecular interactions in a cellular context. Here, we designed and developed photo-click morphine (PCM-2) as a photo-affinity probe based on morphine and dialkynyl-acetyl morphine (DAAM) as a metabolic acetate reporter based on heroin. Application of these probes to SH-SY5Y, HEK293T, and U2OS cells revealed that PCM-2 and DAAM primarily localize to the lysosome amongst other locations inside the cell by confoc…
