Oesophagus carcinoma amino acids control.

Serum amino acids level examination of the oesophagus carcinoma patients admitted to I Surgical Clinic of Teaching Hospital in Olomouc from 1/1/1990 to 31/7/1991 were carried out by the authors. The standard values were found with the group of patients with resectionable tumor or with a tumor in non-advanced stage on the one hand and on the other hand there were some lower values with the patients with a tumor in an advanced stage. In this way two groups of the sick were examined--the first one with benign oesophagus stenosis accompanied by nutritional disorder, the second with resectionable l

Differential Impact of Dietary Branched Chain and Aromatic Amino Acids on Chronic Kidney Disease Progression in Rats.

The metabolism of dietary proteins generates waste products that are excreted by the kidney, in particular nitrogen-containing urea, uric acid, ammonia, creatinine, and other metabolites such as phosphates, sulfates, and protons. Kidney adaptation includes an increase in renal plasma flow (RPF) and glomerular filtration rate (GFR) and represents a burden for diseased kidneys increasing the progression rate of CKD. The present study aimed at identifying potential differences between amino acid (AA) groups constituting dietary proteins regarding their impact on RPF, GFR, and CKD progression. We

Divergent amino acid and sphingolipid metabolism in patients with inherited neuro-retinal disease.

OBJECTIVES: The non-essential amino acids serine, glycine, and alanine, as well as diverse sphingolipid species, are implicated in inherited neuro-retinal disorders and are metabolically linked by serine palmitoyltransferase (SPT), a key enzyme in membrane lipid biogenesis. To gain insight into the pathophysiological mechanisms linking these pathways to neuro-retinal diseases we compared patients diagnosed with two metabolically intertwined diseases: macular telangiectasia type II (MacTel), hereditary sensory autonomic neuropathy type 1 (HSAN1), or both. METHODS: We performed targeted metabol

Decreased purine synthesis during amino acid starvation of human lymphoblasts.

Normal human lymphoblasts starved for each of several essential, but not essential, amino acids had decreased DNA and RNA synthesis but no change in free intracellular purine nucleotides. The rates of purine nucleotide synthesis via the de novo and salvage pathways were measured by incorporating [14C]formate and [14C]hypoxanthine labels, respectively, into lymphoblasts starved for an amino acid or treated with a protein synthesis inhibitor. After 3 h of starvation, purine synthesis via the de novo pathway decreased 90% and via the salvage pathway decreased 60%. Cycloheximide and puromycin each

Silicon Quantum Dots Enhance the Nutritional Quality and Digestive Bioaccessibility of Cherry Radish (Raphanus sativus L.) via Rhizosphere Modulation.

A 50-day greenhouse pot experiment was conducted to evaluate the effects of soil-applied silicon quantum dots (Si QDs) and SiO2 nanoparticles (SiO2 NPs) at 50 mg kg-1 on cherry radish (Raphanus sativus L.), using unamended soil and ionic Si as comparative controls. Si QDs exhibited higher Si bioavailability and assimilation efficiency, increasing edible taproot biomass by 90.7% relative to the unamended control. Mechanistically, Si QDs enhanced taproot ammonium accumulation (23.1%) by stimulating urease activity and enriching beneficial rhizobacteria (Ramlibacter, Ensifer, Thauera). This promo

Vegan proteins may reduce risk of cancer, obesity, and cardiovascular disease by promoting increased glucagon activity.

Amino acids modulate the secretion of both insulin and glucagon; the composition of dietary protein therefore has the potential to influence the balance of glucagon and insulin activity. Soy protein, as well as many other vegan proteins, are higher in non-essential amino acids than most animal-derived food proteins, and as a result should preferentially favor glucagon production. Acting on hepatocytes, glucagon promotes (and insulin inhibits) cAMP-dependent mechanisms that down-regulate lipogenic enzymes and cholesterol synthesis, while up-regulating hepatic LDL receptors and production of the

Infusion of enteral vs parenteral nutrients using high-concentration branch-chain amino acids: effect on wound healing in the postoperative rat.

Starting total parenteral nutrition (TPN) the day after acute surgical stress has beneficial effects on body weight, nitrogen balance, and colonic anastomosis bursting pressure in normally nourished rats. In view of the reported favorable utilization of high-concentration branch-chain amino acids (BCAA) following severe stress, we compared enteral (TEN) to parenteral (TPN) nutrient infusions containing increased BCAA starting the day of operation. Twenty-four male Sprague-Dawley rats, in two groups paired by weight under IP pentobarbital anesthesia underwent jugular vein catheter (CVP) inserti

The Metabolic-Immune Axis: Amino Acids and Immune Cell Dynamics in Mammary Gland Development and Remodeling.

The mammary gland undergoes tightly coordinated cycles of growth, differentiation and regression, which require precise communication between epithelial and stromal cells. Immune cells, whether tissue-resident or recruited, are well-established regulators of mammary development, which is orchestrated by hormones, cytokines, growth factors, microorganisms and nutritional signals. As essential building blocks for protein synthesis, amino acids modulate systemic metabolic homeostasis and influence the development, differentiation, and functional capacity of immune cells. However, the relationship

Peculiarities of one-carbon metabolism in the strict carnivorous cat and the role in feline hepatic lipidosis.

Research in various species has indicated that diets deficient in labile methyl groups (methionine, choline, betaine, folate) produce fatty liver and links to steatosis and metabolic syndrome, but also provides evidence of the importance of labile methyl group balance to maintain normal liver function. Cats, being obligate carnivores, rely on nutrients in animal tissues and have, due to evolutionary pressure, developed several physiological and metabolic adaptations, including a number of peculiarities in protein and fat metabolism. This has led to specific and unique nutritional requirements.

Photoreactivity of amino acids: tryptophan-induced photochemical events via reactive oxygen species generation.

Tryptophan (Trp), an aromatic amino acid, is a constituent of peptides/proteins and is also a precursor of serotonin, kynurenine derivatives, and nicotinamide adenine dinucleotides. There have been a number of reports on photochemical reactions involving peptides/proteins which contain Trp that showed significant photodegradation, dimerization, and photoionization. The photochemical properties of Trp have not been fully elucidated, and this would provide novel insight into the handling of Trp-containing peptides/proteins. Consequently, we have been trying to evaluate the photochemical properti

Cerebrospinal amino acid profiling in a large cohort of neuropediatric patients with epilepsy.

This study addresses challenges in cerebrospinal fluid (CSF) amino acid profiling in pediatric neurological disorders by establishing age-continuous reference intervals. It examines amino acid variations in epilepsy of different etiologies and evaluates the effects of antiseizure medications (ASMs), resolving inconsistencies in previous research. We retrospectively analyzed 410 CSF samples from pediatric patients (201 with epilepsy). Reference intervals were established using linear regression, adjusting for age and sex as confounders. Statistical analysis of age-normalized data included tests

A colorimetric strategy for quantifying amino acids using E. coli auxotrophs displaying gold-binding proteins.

We present a cell-based colorimetric assay for amino acid quantification using Escherichia coli auxotrophs engineered to grow exclusively in the presence of their corresponding target amino acids and to display gold-binding proteins (GBPs) on their surface. Upon the addition of gold nanoparticles (AuNPs), the surface-expressed GBPs mediate AuNP aggregation, resulting in a distinct color shift from pink to blue. This approach enables the detection of ten essential amino acids through characteristic colorimetric changes and allows for quantitative analysis via absorbance measurements with high s

Protein nutrition in the alligator.

1. Fourteen different protein-containing diets were fed to small alligators. Their rates of digestion and their nutritional values were determined by following changes in free amino acids in the plasma. 2. Fish, chicken and nutria were digested rapidly and all their component essential amino acids disappeared quickly and at the same rate. When given in the dry, fat-free form the amino acids were released and assimilated about 50% faster than when fat was included. 3. None of the isolated proteins tested (casein, gelatin, edestin, gliadin, corn gluten and soy) proved nutritionally adequate and

Halogenated tryptophan derivatives disrupt essential transamination mechanisms in bloodstream form Trypanosoma brucei.

Amino acid metabolism within Trypanosoma brucei, the causative agent of human African trypanosomiasis, is critical for parasite survival and virulence. Of these metabolic processes, the transamination of aromatic amino acids is one of the most important. In this study, a series of halogenated tryptophan analogues were investigated for their anti-parasitic potency. Several of these analogues showed significant trypanocidal activity. Metabolomics analysis of compound-treated parasites revealed key differences occurring within aromatic amino acid metabolism, particularly within the widely reporte

[Dangers of a wrong diet in patients with renal insufficiency in the predialytic state or during chronic hemodialysis].

Patients with chronic renal failure and on chronic intermittent hemodialysis have several metabolic risks such as retention of urea and other products of nitrogen metabolism, catabolism, acidosis, edema, dehydration, hyper- and hypotension, hyperkalemia, renal osteopathy, and renal anemia. Uremic coma is usually avoided by balanced nutritional therapy and treatment with hemodialysis. A dietary regimen containing protein with high amounts of essential amino acids is an important part of treatment. In patients on chronic hemodialysis disorders of protein and amino acid metabolism are caused by e

Impact of different thermal treatments on heavy metals, aroma-active compounds, and protein-related nutritional quality of edible green seaweed Ulva reticulata.

Ulva reticulata, an edible green seaweed, holds promise as a functional food ingredient but is limited by heavy metal contamination and a strong fishy odor. This study evaluated hot water, steam, microwave, and ultrasound processing methods to improve safety, sensory quality, and retention of essential amino acids. Microwave and ultrasound treatments significantly (P < 0.05) reduced heavy metals-arsenic, cadmium, tin, mercury, lead, nickel, and chromium-to within FSSAI limits, with microwave processing achieving up to 88 % reduction. Headspace-GC-MS/MS analysis identified 46 volatile compounds

Probing the Role of Small Molecules and Amino Acids in Protein Adsorption on Gold Nanoparticles: A Thermodynamic and Mechanistic Insight.

A molecular-level understanding of amino acid and small-molecule adsorption onto gold nanosurfaces is essential for designing nanomaterials with tailored biointerfacial properties. Till now, the interactions of gold nanoparticles (GNPs) with complex biomolecules such as peptides, proteins, and nucleic acids have been extensively studied under varying physicochemical conditions (including particle size, surface charge, functionalization, pH, and ionic strength). However, the fundamental mechanistic pathways governing the adsorption of their molecular building blocks─amino acids─onto GNP surface

Serum amino acid changes in rats with thioacetamide-induced liver cirrhosis.

To date, no attempt has been made to study alterations occurring in the amino acid profile in chronic models of thioacetamide-induced liver cirrhosis. In this work, changes in serum amino acids and proteins in rats with thioacetamide-induced liver cirrhosis are reported, together with changes in enzyme activities in the liver and serum. Seventeen female Wistar rats were used. Eight rats were given 300 mg thioacetamide/l in drinking water for 4 months and nine rats were given water ad libitum during the same time-period. Significant increases in glycine, alanine, serine, methionine, glutamate,

Methods to study changing free amino acid pools during embryonic chick development.

During early embryogenesis of the chick, the yolk represents the only source of free amino acids. Leucine and glutamic acid, together with valine, predominate in this nonrenewable nutrient supply; taurine and phosphoethanolamine are present in the lowest concentration. The tyrosine level is already twice as high as phenylalanine. At embryo day 2 (E2), the not-yet fully functioning vitelline blood plexus demonstrates an amino acid profile that rather closely resembles that of the yolk, except for three amino acids. During the transfer process from yolk to plexus, a certain amount of glutamic ac
