Barriers to the Pharmacologic Rescue of W1282X CFTR.

W1282X CFTR is the most prevalent CF-causing variant among cystic fibrosis patients of Ashkenazi descent and a mutational defect for which targeted drug therapy is not available. We show that administration of the potentiator VX-770 can augment levels of truncated W1282X CFTR in the plasma membrane, demonstrating that an established gating activator (i.e., "potentiator") also rescues W1282X protein expression and surface localization (i.e., "corrector" function). Additionally, acute in vitro treatments with approved modulators VX-809 or VX-661 result in immediate potentiation of W1282X-depende

PTI-801 (posenacaftor) shares a common mechanism with VX-445 (elexacaftor) to rescue p.Phe508del-CFTR.

The deletion of a phenylalanine at position 508 (p.Phe508del) in the CFTR anion channel is the most prevalent variant in people with Cystic Fibrosis (CF). This variant impairs folding and stability of the CF transmembrane conductance regulator (CFTR) protein, resulting in its defective trafficking and premature degradation. Over the last years, therapeutic accomplishments have been attained in developing small molecules that partially correct p.Phe508del-CFTR defects; however, the mechanism of action (MoA) of these compounds has only started to be uncovered. In this study, we employed biochemi

CFTR modulator therapy: transforming the landscape of clinical care in cystic fibrosis.

Following discovery of the cystic fibrosis transmembrane conductance regulator (CFTR) gene in 1989 and subsequent elucidation of the varied CFTR protein abnormalities that result, a new era of cystic fibrosis management has emerged-one in which scientific principles translated from the bench to the bedside have enabled us to potentially treat the basic defect in the majority of children and adults with cystic fibrosis, with a resultant burgeoning adult cystic fibrosis population. However, the long-term effects of these therapies on the multiple manifestations of cystic fibrosis are still under

Spectrophotometric determination of paracetamol in urine with tetrahydroxycalix[4]arene as a coupling reagent and preconcentration with triton X-114 using cloud point extraction.

In the present paper, conventional spectrophotometry in conjunction with cloud point extraction-preconcentration were investigated as alternative methods for paracetamol (PCT) assay in urine samples. Cloud point extraction (CPE) was employed for the preconcentration of p-aminophenol (PAP) prior to spectrophotometric determination using the non-ionic surfactant Triton X-114 (TX-114) as an extractant. The developed methods were based on acidic hydrolysis of PCT to PAP, which reacted at room temperature with 25,26,27,28-tetrahydroxycalix[4]arene (CAL4) in the presence of an oxidant (KIO(4)) to fo

Mechanistic Insights into the Analgesic Action of Acetaminophen: Its Bioactive Metabolite Induces Peripheral Analgesia in Addition to Central Analgesia.

The molecular target and detailed mechanism of analgesic acetaminophen are still not fully understood despite being commonly used for many years. Through a literature search, the present study aimed to characterize the cyclooxygenase (COX) inhibitory properties of acetaminophen in comparison with nonsteroidal anti-inflammatory drugs (NSAIDs) and to gain novel mechanistic insights into its analgesic action, especially focusing on the peripheral mechanisms. Comparative characterization indicated that acetaminophen is relatively selective for COX-2 but less potent to inhibit COX than NSAIDs. This

Non-invasive identification of brain signatures of acute liver injury.

Background: In many disorders, metabolic and inflammatory derangements that originate in peripheral organs have a deleterious impact on the brain. Brain functional impairment, defined as hepatic encephalopathy, is one of the main diagnostic criteria for acute liver failure (ALF), a severe complication of acute liver injury (ALI). While brain inflammation (neuroinflammation) and metabolic alterations significantly contribute to hepatic encephalopathy, their non-invasive evaluation remains challenging. Methods: To address this limitation, we utilized dual radiotracer [18F]-fluoro-2-deoxy-2-D-glu

Competition between resident rhizosphere bacteria enhances phytopathogen suppression via emergent antagonism.

Resident bacterial interactions can shape the invasion resistance of rhizosphere microbiomes, but whether interactions between weakly antagonistic resident bacteria can generate emergent antagonism against invading pathogens remains poorly understood. Here, we used a systematic pairwise interaction screening to identify Ralstonia pickettii RAL5 and Acinetobacter oleivorans ACI4 bacterial pair, that together provided a strong suppression of the phytopathogenic R. solanacearum Rs1115 strain. Although RAL5 and ACI4 monocultures only weakly inhibited pathogen growth, the RAL5-ACI4 co-cultures stro

Enhanced catalytic transfer hydrogenation of p-nitrophenol using formaldehyde: MnO2-supported Ag nanohybrids with tuned d-band structure.

The catalytic reduction of p-nitrophenol (4-NP) to p-aminophenol (4-AP) is important in fine chemical synthesis, especially in the pharmaceutical, dye, and agrochemical industries. Sustainable methods are increasingly prioritized in these fields. Catalytic transfer hydrogenation (CTH) using formaldehyde (HCHO) as a hydrogen donor presents a safer and more environmentally friendly alternative due to its low-toxicity by-products and ease of handling. In this study, we developed manganese dioxide (MnO2)-supported silver (Ag) nanoparticles (NPs) as an efficient catalyst for the CTH of 4-NP using H

Iron single-atom nanozyme with dual enzyme-like activities for detection of environmental pollutants and colorimetric sensing array to identification of phenolic species.

Organophosphorus pesticides (OPs) and phenolic compounds are widely residual in the environment, and it is necessary to construct simple and effective detection methods. Herein, iron single-atom (Fe-N/C) nanozyme with double mimetic enzyme catalytic properties was designed and prepared through a controlled pyrolysis strategy of Fe-ZIF-8 precursors. The double enzyme-like activities of Fe-N/C were discussed in depth. The oxidase-like (OXD) activity refers to Fe-N/C acting as an oxidase mimetic to generate singlet oxygen (1O2) with the assistance of O2, thereby enabling the oxidation of 3,3',5,5

Effect of the long-term use of a NOAEL dose of acetaminophen (paracetamol) on hepatic, renal, and neural tissues of aged albino rats.

BACKGROUND: Paracetamol is one of the most popular drugs; it is used daily by many people especially the elderly, without a limitation on the length of the period allowed for continuous use. Harms from long-term use are less clear, particularly in extrahepatic regions. AIM: This study aimed to investigate whether using paracetamol at a non-observable adverse effect level dose, known not to cause toxic effects, for a long period can induce toxicity in aged male albino rats. METHODS: A daily dose of 500 mg per kg body weight of paracetamol was given to adult male albino rats for 12 weeks. Duri

Green Synthesis of Silver Nanoparticles Using the Leaf Extract of the Medicinal Plant, Uvaria narum and Its Antibacterial, Antiangiogenic, Anticancer and Catalytic Properties.

Silver nanoparticles (AgNPs) made by green synthesis offer a variety of biochemical properties and are an excellent alternative to traditional medications due to their low cost. In the current study, we synthesised AgNPs from the leaf extract of the medicinal plant Uvaria narum, commonly called narumpanal. The nanoparticles were characterised by ultraviolet-visible (UV-Vis) spectroscopy, Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM) and transmission electron microscopy (TEM). SEM analysis showed AgNPs are highly crystalline and spherical with an average dia

Association between acetaminophen use and vitiligo in US women and men.

BACKGROUND/OBJECTIVES: Exposure to chemical phenols, which can act as tyrosine analogues and result in anti-melanocyte autoimmunity, has been associated with vitiligo. Acetaminophen (N-acetyl-p-aminophenol) is an over-the-counter analgesic of phenolic origin. The risk of vitiligo with systemic exposure to acetaminophen has not yet been evaluated. METHODS: We examined the risk of vitiligo with regular use acetaminophen in women, the Nurses' Health Study (NHS) and in men, the Health Professionals Follow-up Study (HPFS). Regular acetaminophen use was asked biennially from 1990 in NHS and from 19

3-D porous cellulose nanofibril aerogels with a controllable copper nanoparticle loading as a highly efficient non-noble-metal catalyst for 4-nitrophenol reduction.

Nitrophenols(NPs) are highly toxic compounds that occur in various industrial effluents. Herein, we investigated Cu nanoparticle-loaded cellulose nanofibril (CNF/PEI-Cu) aerogels as a catalyst for degrading 4-nitrophenol (4NP) in the wastewater. Non-noble metal based low-cost catalyst material and easily scalable preparation method make CNF/PEI-Cu aerogel as an appropriate catalyst for practical application in 4NP wastewater treatment. Our strategy to improve the loading amount of homogeneously distributed Cu nanoparticles was to functionalize a CNF aerogel using polyethylene imine (PEI), whic

A critical review over the removal of paracetamol (acetaminophen) from synthetic waters and real wastewaters by direct, hybrid catalytic, and sequential ozonation processes.

Paracetamol (PCT) or acetaminophen is a widely prescribed drug to treat fever and mild to moderate pain. The PCT uptake by animals and humans is not complete, being excreted through their urine to contaminate the aquatic/natural environments. Trace amounts of this drug have been found in sewage sludge, hospital wastewaters, wastewater plant treatments, surface waters, and even drinking water. PCT denatures proteins and oxidize lipids in cells with damage of their genetic code. Its toxicity over macrophytes, protozoan, algae, bacteria, and fishes has been reported. Ozonation methods have been p
