Hyperstimulation of pituitary function using daily injections of 50,500 or 5000 ng LH-RH agonist for 3 weeks or hypostimulation by immunoneutralization of endogenous LH-RH led to disrupted oestrous cycles and profound changes in ovarian morphology. Despite this, the number of pituitary and ovarian LH-RH receptors remained within the range found in normally cyclic animals. Bone resorption, stimulated by injection of parathyroid extract, was measured in vivo in microphthalmic mice as the rate of release of 3H from bone after incorporation of 3H-proline. Bone resorption in these mice, which inherit osteopetrosis, was less than 10% of the in normal litermates. Autoradiography confirmed the reduction in removal of radioactive bone matrix and in bone growth in microphthalmic mice. Cyclocryotherapy of the monkey eye reliably produces transient increased intraocular pressure followed by prolonged hypotony, during which papilledema occurs. Changes in the distribution of T, B and null cells in the spleen and bone marrow have been studied in inbred mice infected with Trypanosoma brucei. Immunofluorescent staining combined with a 3H-thymidine pulse and autoradiography showed activation of all three cell types. A transient increase in splenic T cells was followed by dramatic increases in B cells (2·5-fold) and in null cells (35-fold). By day 12, after the first peak of parasitaemia, nearly half the spleen cells were dividing. In the bone marrow, very large blast cells appeared within a week of infection.
