A pan-serotype human monoclonal antibody protects against pneumococcal infection by targeting multiple choline binding domain proteins.

Streptococcus pneumoniae remains a global health threat, particularly to young children, the elderly, and immunocompromised individuals. Pneumococcal vaccines targeting the bacterial capsule polysaccharide do not protect against all 100+ pneumococcal serotypes, contributing to non-vaccine serotype infections and antibiotic resistance. To address these limitations, we isolated human monoclonal antibodies (mAbs) targeting pneumococcal surface proteins and identified a first-in-class mAb, derived from a patient with prior pneumococcal infection, namely mAb 5995-40. mAb 5995-40 bound multiple pneu

Infant Mouse Model for the Study of Shedding and Transmission during Streptococcus pneumoniae Monoinfection.

One of the least understood aspects of the bacterium Streptococcus pneumoniae (pneumococcus) is its transmission from host to host, the critical first step in both the carrier state and the disease state. To date, transmission models have depended on influenza A virus coinfection, which greatly enhances pneumococcal shedding to levels that allow acquisition by a new host. Here, we describe an infant mouse model that can be utilized to study pneumococcal colonization, shedding, and transmission during bacterial monoinfection. Using this model, we demonstrated that the level of bacterial sheddin

Expanded prevalence of group 4 capsule expression and its genetic requirements in a broad array of Shigella strains.

UNLABELLED: The expression of a group 4 capsule (G4C) has been identified in Shigella sonnei and Shigella flexneri serotype 6 and demonstrated to play a significant role in pathogenic processes in S. sonnei. However, it has not been examined phenotypically in S. flexneri 6. Despite the impact of G4C, no systematic analysis of its expression or the presence of the genomic determinants has been performed across a large collection of S. sonnei and S. flexneri serotypes 2a and 6 or any of the other >46 Shigella strains and serotypes that cause disease. Our examination of the genomes of 1,219 geogr

Investigation of the essential role of capsular polysaccharide in serotype 1 Streptococcus pneumoniae.

Streptococcus pneumoniae remains a major global cause of morbidity and mortality, responsible for an estimated half million deaths annually despite widespread implementation of pneumococcal conjugate vaccines. Notably, invasive pneumococcal disease (IPD) caused by serotype 1 persists and disproportionately affects vulnerable populations in low-resource settings, despite inclusion of this serotype in available vaccines. Serotype 1 exhibits atypical biology characterized by short nasopharyngeal carriage and high invasive potential. This phenotype has been linked to capsule-mediated evasion of ho

Capsular Typing and Molecular Characterization of Streptococcus agalactiae Strains Isolated From Bovine Mastitis in Iran.

Streptococcus agalactiae infections may cause clinical or subclinical mastitis in dairy cows by invading the mammary gland. This research included the isolation of 29 strains of S. agalactiae from 425 milk samples obtained from cows affected by clinical mastitis in Chaharmahal and Bakhtiari province, Iran. The antimicrobial sensitivity of S. agalactiae strains was determined using 16 antibiotics from seven different classes. The epidemiological spread of S. agalactiae was determined by identifying the serotypes of isolates using multiplex PCR. In addition, the presence of antibiotic-resistance

Discovery of anti-mucoviscous activity of rifampicin and its potential as a candidate antivirulence agent against hypervirulent Klebsiella pneumoniae.

A recent increase in the incidence of hypervirulent Klebsiella pneumoniae (hvKP) infections, especially those caused by a sublineage of clonal group CG23 (CG23-I), is raising serious health concerns worldwide. The high virulence of hvKP is, at least in part, attributed to the overproduction of capsular polysaccharide (CPS), which is triggered by a positive regulator of capsular polysaccharide synthesis (cps) genes, named rmpA (regulator of mucoid phenotype A). Although extensive research has been conducted on the mechanisms of hvKP virulence, no study has focused on the development of antiviru

Hyaluronic acid derived from other streptococci supports Streptococcus pneumoniae in vitro biofilm formation.

We investigate the role of hyaluronic acid (HA) on S. pneumoniae in vitro biofilm formation and evaluate gene expressions of virulence and/or biofilm related genes. Biofilms were grown in medium supplied with HA derived from capsule of Streptococcus equi. The biomasses of biofilms were detected by crystal-violet (CV) microtiter plate assay, and the morphology was viewed under scanning electron microscope (SEM). The gene expressions were assessed by relative quantitative RT-PCR. The results showed that the HA support pneumococcal growth in planktonic form and within biofilms. The CV-microtiter

Genome sequence of a Neisseria meningitidis capsule null locus strain from the clonal complex of sequence type 198.

Neisseria meningitidis is a commensal and accidental pathogen exclusively of humans. Although the production of polysaccharide capsules is considered to be essential for meningococcal virulence, there have been reports of constitutively unencapsulated strains causing invasive meningococcal disease (IMD). Here we report the genome sequence of a capsule null locus (cnl) strain of sequence type 198 (ST-198), which is found in half of the reported cases of IMD caused by cnl meningococcal strains.

Capsular expression in Streptococcus pneumoniae negatively affects spontaneous and antibiotic-induced lysis and contributes to antibiotic tolerance.

Penicillin and vancomycin induce a lytic response in Streptococcus pneumoniae that requires the N-acetylmuramyl-l-alanine amidase LytA. We show that clinical isolates of pneumococci of capsular serotypes 1, 4, 6B, and 23F were generally less lytic to penicillin than pneumococci of serotypes 14 and 3. In addition, most 9V isolates were less lytic to vancomycin, compared with isolates of other serotypes. Parent-mutant pairs expressing and not expressing capsular serotypes 2, 4, and 9V were compared for antibiotic-induced lysis. The nonencapsulated variants were considerably more lytic after beta

[Comparative incidence of detection of specific antibodies to Yersinia pestis capsular antigen and lipopolysaccharide in humans immunized with pest vaccine].

Two to 8 years after vaccination, specific antibodies to capsular antigen (F1) and lipopolysaccharide (LPS) are found in the blood sera of but 10 to 20% of subjects 1-4 times epicutaneously immunized with live antipest vaccine. Only regular continuous (for at least 7-15 years) antipest vaccination leads to the production of antibodies to the main antigens of Yersinia pestis, detectable in remote periods in 75.9 +/- 4.5% of vaccinees. It is noteworthy that the number of positive responses and titer of antibodies to LPS is appreciably higher than to F1. Detection of antibodies to Y. pestis is re

Meningitis and epiglottitis: a new immunisation against haemophilus influenzae type b infections.

Since 1985 the number of cases of systemic haemophilus influenzae type b (Hib) disease in children in the Oxford region has been recorded. This study shows that a cumulative incidence of children suffering systematic Hib infections before their fifth birthday is 1:600, 71 per cent of sufferers having meningitis. Because the incidence of haemophilus infections may be increasing in the UK and there is increasing resistance of the organism to antimicrobial therapy, the need for primary prevention through immunisation is becoming increasingly attractive. We have therefore undertaken a safety and i

Carbon source-dependent capsule thickness regulation in Streptococcus pneumoniae.

BACKGROUND: The polysaccharide capsule of Streptococcus pneumoniae plays a major role in virulence, adherence to epithelial cells, and overall survival of the bacterium in the human host. Galactose, mannose, and N-acetylglucosamine (GlcNAc) are likely to be relevant for metabolization in the nasopharynx, while glucose is the primary carbon source in the blood. In this study, we aim to further the understanding of the influence of carbon sources on pneumococcal growth, capsule biosynthesis, and subsequent adherence potential. METHODS: We tested the growth behavior of clinical wild-type and cap

Examining the Distribution and Impact of Single-Nucleotide Polymorphisms in the Capsular Locus of Streptococcus pneumoniae Serotype 19A.

Streptococcus pneumoniae serotype 19A prevalence has increased after the implementation of the PCV7 and PCV10 vaccines. In this study, we have provided, with high accuracy, the genetic diversity of the 19A serotype in a cohort of Dutch invasive pneumococcal disease patients and asymptomatic carriers obtained in the period from 2004 to 2016. The whole genomes of the 338 pneumococcal isolates in this cohort were sequenced and their capsule (cps) loci compared to examine their diversity and determine the impact on the production of capsular polysaccharide (CPS) sugar precursors and CPS shedding.

A simple, reliable M'Fadyean stain for visualizing the Bacillus anthracis capsule.

The simple polychrome methylene blue (PMB) staining procedure for blood or tissue smears from dead animals (M'Fadyean reaction) established in 1903 remained accepted as a highly reliable, rapid diagnostic test for anthrax for six decades while that disease was still common in livestock throughout the world. Improvements in disease control led to anthrax becoming rare in industrialized countries and less frequent in developing countries with the result that quality controlled, commercially produced PMB became hard to obtain by the 1980s. Mixed results with alternative methylene blue-based stain

Effects of the capsular polysaccharides of Cryptococcus neoformans on phagocyte migration and inflammatory mediators.

An important virulence factor of the pathogenic fungus Cryptococcus neoformans is its polysaccharide capsule. The capsular polysaccharides glucuronoxylomannan (GXM), galactoxylomannan (GalXM) and the mannoproteins (MPs) display various immunomodulatory effects on the host response, such as the inhibition of phagocytosis, suppression of T-cell mediated immunity, and induction of immunogenic tolerance. Moreover, these capsular polysaccharides are able to interfere with the migration of phagocytes despite adequate stimulation of chemokine production and their concerted action accounts for the mil

The infant rat model adapted to evaluate human sera for protective immunity to group B meningococci.

The infant rat infection model previously developed to evaluate protective ability of passively administered murine antibodies to group B meningococcal (MenB) surface antigens was adapted for human sera. Several challenge doses were tested, aiming at sensitive detection of protection with little interassay variability. Doses of 10(5) and 10(6) colony forming units of strain IH5341 (MenB:15:P1.7,16) injected intraperitoneally gave consistently high levels of bacteremia and meningitis developed in 6 h in 50-100% of the pups. A monoclonal antibody mAb735 to the MenB capsule, injected 1-2 h before

Interaction of pathogenic neisseriae with nonphagocytic cells.

The ability to interact with nonphagocytic cells is a crucial virulence attribute of the meningococcus and the genococcus. Like most bacterial pathogens, Neisseria meningitidis and Neisseria gonorrhoeae initiate infections by colonizing the mucosal epithelium, which serves as the site of entry. After this step, both bacteria cross the intact mucosal barrier. While N. gonorrhoeae is likely to remain in the subepithelial matrix, where it initiates an intense inflammatory reaction, N. meningitidis enters the bloodstream, and eventually the cerebrospinal fluid to cause meningitis. Both pathogens h

A molecular model of artificial glycoprotein with predetermined multiple immunodeterminants for gram-positive and gram-negative encapsulated bacteria.

An artificial molecule was synthesized by covalently linking the oligosaccharide haptens derived frm Streptococcus pneumoniae type 6A and Neisseria meningitidis group C capsular polysaccharides to the non-toxic mutant protein CRM197, serologically related to diphtheria toxin. Immunochemical analysis using polyclonal and monoclonal antibodies showed in the glycoprotein the presence of specific immunodeterminants of the native polysaccharides and of the carrier protein. The immunological activity of this hybrid molecule tested in two animal models gave evidence for anamnestic induction of serum

OxyR positively and directly regulates Vi polysaccharide capsular antigen in Salmonella enterica serovar Typhi.

Salmonella enterica serovar Typhi (S. Typhi) is a human enteropathogen that can overcome oxidative stress and survive in macrophages. OxyR is an important part of the antioxidant defense system. S. Typhi expresses a virulence (Vi) polysaccharide capsular antigen, which provides the bacterium with the ability to avoid host defenses and suppress detection by the innate immune system. This study investigated the effect of OxyR on Vi antigen in S. Typhi. In the oxyR mutant strain, microarray analysis, quantitative real time PCR and β-galactosidase assay confirmed that the viaB operon was positivel
