As a renewable energy source, cellulosic ethanol has excellent prospects that can help break our dependence on fossil fuels. Simultaneous saccharification and fermentation (SSF)is the most appealing strategy for producing cellulosic ethanol. However, for the SSF process, the disparity between the optimal temperatures for cellulases (50 °C) and yeasts (30 °C) hinders the efficacy of SSF. Herein, the composite material of 2D titanium carbon nanosheets (Ti3C2Tx) and graphene oxide (GO), having excellent photothermal conversion efficiency, was used as the carrier for cellulase immobilization. This

Enzyme immobilization is an efficient and cost-effective approach to recovering, stabilizing, and enhancing enzyme catalytic properties. It is a challenge, however, for coimmobilized multiple enzymes to perform consecutive reactions without being inactivated under similar conditions. Here, we present a facile enzyme immobilization platform using β-lactoglobulin amyloid fibril hydrogels. Two different hydrogels, loading either RuBisCO alone (hereby termed AFR*) or seven enzymes related to the Calvin Cycle (hereby termed AF7E hydrogel), show immobilization efficiency of over ∼95% while simultane

Ethanol is environmentally friendly, not hazardous, and can be produced from renewable sources. In this study, ethanol was applied as a cosolvent for the tyrosinase-catalyzed synthesis of catechol compounds from phenolic compounds, which are abundant in fruit pomace. To investigate the cosolvent function of ethanol on tyrosinase, six types of tyrosinase from Priestia megaterium and water-insoluble phenolic substrates (i.e., phloretin and resveratrol) were used. Biocatalytic activity and stability of tyrosinase were maintained in the ethanol-water system up to 80 % (v/v) ethanol with an optimum

Furanosteroids are known to exhibit inhibitory activity against phosphatidylinositol-3-kinase and are expected to serve as a basis for the development of therapeutic drugs for various diseases. In this study, a novel protocol is presented for preparation of the furanosteroid A-ring moiety. More specifically, the lipase-catalyzed kinetic resolution of racemic 1-(3-bromofuran-2-yl)-2-chloroethanol with β-substituted (Z)-acrylates and the subsequent intramolecular Diels-Alder reaction of the generated enantiomerically enriched esters were performed to obtain multi-functionalized fused cyclohexene

The conversion of CO2 into fuels and chemicals via biological routes has gained significant attention due to its high efficiency and mild reaction conditions. However, biological CO2 fixation faces several challenges, including the low solubility of CO2, the poor stability of free enzymes and cells under harsh conditions, and the high cost of cofactors such as NAD(P)H. Metal-organic frameworks (MOFs), with their unique characteristics including high surface area, tunable pore structure, and ease for functionalization, have emerged as promising candidates to address these limitations. When inte

An in vivo self-assembly strategy was developed to overcome the limitation of traditional enzyme immobilization methods. An engineered E. coli system was employed to immobilize carbonyl reductase (LcCR) onto intracellular polyhydroxyalkanoate (PHA) microspheres, which was realized by fusion expression of LcCR with covalent PhaC or non-covalent PhaP. The fusion of LcCR and PHA surface binding proteins (Phac, PhaP) was optimized by adjusting the fusion direction (C/N end) and replacing peptide linkers (GSA, (G4S)3, (G4S)4). The expression form for fusion of N-terminal of LcCR with three series c

Thiamine diphosphate (ThDP)-dependent enzymes are well known biocatalysts for CC bond-forming reactions. While this enzyme class is mainly investigated for the formation of acyloins of secondary alcohols, recent studies have expanded its scope to utilize ketones as electrophiles in asymmetric carboligation reactions for the formation of tertiary alcohols. Chiral tertiary alcohols are ubiquitous motifs in natural products and important building blocks for the synthesis of bioactive compounds. ThDP-dependent enzymes are emerging as one of the most promising classes of biocatalysts for synthesiz

Azaspiro[2.y]alkanes are increasingly valuable scaffolds in pharmaceutical drug discovery; however, an asymmetric catalytic method for their synthesis remains unknown. Here, we present a stereodivergent carbene transferase platform for the cyclopropanation of unsaturated exocyclic N-heterocycles to provide structurally diverse and pharmaceutically relevant azaspiro[2.y]alkanes in high yield (21->99% yield) and excellent diastereoselectivity and enantioselectivity (diastereomeric ratio (dr) values from 52.5:47.5 to >99.5:0.5; enantiomeric ratio (er) values from 51:49 to >99.5:0.5). Enzymes represent renewable biological resources, and their employment as industrial biocatalysts not only enhances production efficiency but also mitigates environmental pollution associated with excessive organic solvent usage. (R)-N-Cyclopropylpiperidin-3-amine serves as a crucial pharmaceutical intermediate, traditionally produced via chiral resolution. Imine reductases (IREDs) present an atom-efficient alternative by directly catalyzing the reductive amination of prochiral ketones with amines to yield chiral alkylated amines. Through site-directed mutagenesis of IR-G36-M5, we developed th

The enantioselective conjugate hydrocyanation of α,β-unsaturated aldehydes remains a long-standing challenge in synthetic chemistry. Here, we report the redesign of 2-deoxy-D-ribose-5-phosphate aldolase (DERA) into an efficient biocatalyst capable of promoting the asymmetric conjugate addition of hydrogen cyanide (generated in situ from trimethylsilyl cyanide) to aromatic enals via an iminium activation pathway. The evolved variant DERA-CN enables the efficient formation of various C4-nitriles with high conversions (up to 99%) and good enantioselectivity (up to 98% e.e.). Control experiments r

Chiral tertiary alcohols are important organic compounds in science as well as in industry. However, their preparation in enantiomerically pure form is still a challenge due to their complex structure and steric hindrances compared with primary and secondary alcohols, so kinetic resolution could be an attractive approach. Lipase A from Candida antarctica (CAL-A) has been shown to catalyze the enantioselective esterification of various tertiary alcohols with excellent enantioselectivity but low activity. Here we report a mutagenesis study by rational design to improve CAL-A activity against ter

A dual lipase system has been developed to convert soybean oil into biodiesel through synergistic catalysis of Thermomyces lanuginosus lipase (TLL) and Yarrowia lipolytica lipase 2 (YLL) in this study. Pichia pastoris recombinant strains expressing lipases were successfully constructed, and the activities of TLL and YLL in the fermentation supernatant reached 23,142.71 ± 280.54 U/mL and 895.44 ± 27.31 U/mL, respectively. Immediately thereafter, free lipase was used to catalyze the preparation of biodiesel from soybean oil. After optimizing reaction conditions, 80 U/g oil TLL and 20 U/g oil YLL

Enzymes are renowned for their catalytic efficiency and selectivity, but many classical transformations in organic synthesis have no biocatalytic counterpart. Aldolases are prodigious C-C bond-forming enzymes, but their reactivity has only been extended past activated carbonyl electrophiles in special cases. To probe the mechanistic origins of this limitation, we use a pair of aldolases whose activity is dependent on pyridoxal phosphate. Our results reveal how aldolases are limited by kinetically favourable proton transfer with solvent, which undermines aldol addition into ketones. We show how

Cytochrome P450s belong to a family of heme-binding monooxygenases, which catalyze regio- and stereospecific functionalisation of C-H, C-C, and C-N bonds, including heteroatom oxidation, oxidative C-C bond cleavages, and nitrene transfer. P450s are considered useful biocatalysts for the production of pharmaceutical products, fine chemicals, and bioremediating agents. Despite having tremendous biotechnological potential, being heme-monooxygenases, P450s require either autologous or heterologous redox partner(s) to perform chemical transformations. Randomly distributed P450s throughout a bacteri

Using the ATP-independent transacylase CapW required for the biosynthesis of capuramycin-type antibiotics, we developed a biocatalytic approach for the synthesis of 43 analogues via a one-step aminolysis reaction from a methyl ester precursor as an acyl donor and various nonnative amines as acyl acceptors. Further examination of the donor substrate scope for CapW revealed that this enzyme can also catalyze a direct transamidation reaction using the major capuramycin congener as a semisynthetic precursor. Biological activity tests revealed that a few of the new capuramycin analogues have signif

The facultative intracellular pathogen Legionella pneumophila, the causative agent of Legionnaires disease, infects and replicates within human alveolar macrophages. L. pneumophila delivers almost 300 effector proteins into the besieged host cell that alter signaling cascades and create conditions that favor intracellular bacterial survival. In order for the effectors to accomplish their intracellular mission, their activity needs to be specifically directed toward the correct host cell protein or target organelle. Here, we show that the L. pneumophila effector GobX possesses E3 ubiquitin liga

Iodothyronine deiodinases (IDs) are mammalian selenoenzymes that catalyze the conversion of thyroxine (T4) to 3,5,3'-triiodothyronine (T3) and 3,3',5'-triiodothyronine (rT3) by the outer- and inner-ring deiodination pathways, respectively. These enzymes also catalyze further deiodination of T3 and rT3 to produce a variety of di- and monoiodo derivatives. In this paper, the deiodinase activity of a series of peri-substituted naphthalenes having different amino groups is described. These compounds remove iodine selectively from the inner-ring of T4 and T3 to produce rT3 and 3,3'-diiodothyronine
