Development of cochlear spiral ligament fibrocytes of the common marmoset, a nonhuman model animal.

Spiral ligament fibrocytes generate potassium gradients, which hair cells require to convert mechanical sound waves into electrical palsy. Together with the stria vascularis, they regulate endolymph electrolyte homeostasis. Developing spiral ligament fibrocytes and generating endocochlear potential with an appropriate endolymph ion composition are essential for hearing. Understanding spiral ligament fibrocyte development is useful for studying age-related and genetic hearing loss, as well as for regenerative therapy and cochlear immunology. Despite interspecies differences, most studies of coc

Vascular, cardiac, and renal lesions attributed to primary systemic hypertension in western pygmy marmosets (Cebuella pygmaea).

In a retrospective study of a western pygmy marmoset (Cebuella pygmaea) colony, postmortem examination of 1/8 juvenile and 29/47 adult animals identified vascular, cardiac, and renal lesions consistent with systemic hypertension. This included frequent renal arteriolar hypertrophy, hyaline and proliferative arteriolosclerosis, fibrinoid necrosis of arterioles, glomerulosclerosis, and nephrosclerosis. Affected animals ranged from 0.6 to 12 years of age (mean 6 years) and had an observed male predominance. Genealogical relatedness was evident in several breeding pairs and spanned multiple genera

Urogenital Lesions in Nonhuman Primates at 2 National Primate Research Centers.

Given their genetic and anatomic similarities to humans, nonhuman primates (NHPs) may serve as animal models for urogenital diseases of humans. The purpose of this study was to examine the frequency of spontaneous urogenital lesions occurring over a 30-year period at the Yerkes and Southwest National Primate Research Centers and to compare and contrast lesions occurring in Old World versus New World primates. Lesions occurring in the chimpanzee (Pan troglodytes), baboon (Papio spp.), rhesus macaque (Macaca mulatta), cynomolgus macaque (Macaca fascicularis), pig-tailed macaque (Macaca nemestrin

Neural plasticity following lesions of the primate occipital lobe: The marmoset as an animal model for studies of blindsight.

For nearly a century it has been observed that some residual visually guided behavior can persist after damage to the primary visual cortex (V1) in primates. The age at which damage to V1 occurs leads to different outcomes, with V1 lesions in infancy allowing better preservation of visual faculties in comparison with those incurred in adulthood. While adult V1 lesions may still allow retention of some limited visual abilities, these are subconscious-a characteristic that has led to this form of residual vision being referred to as blindsight. The neural basis of blindsight has been of great in

Mealworm feeders as environmental enrichment for common marmosets.

The impact of a foraging enrichment device, the "mealworm feeder," on the behavior of the common marmoset was examined. In 3-h weekly exposures to the wormfeeder device, behavioral observations were conducted to compare the rates of feeder use, use of other enrichment devices, stereotyped behavior, and inactivity, to those of control sessions in which the enrichment device was not provided. Significantly decreased rates of pacing and time spent sitting still were observed in association with placement of the mealworm feeder. Feeder use declined over a period of 3 h, even if the feeders' conten

Effects of lesions of different parts of the septo-hippocampal system in primates on learning and retention of information acquired before or after surgery.

Data from a large series of experiments on marmosets with lesions of the septal/diagonal band area (DB), fornix or CA1 area of the hippocampus are analysed in terms of retention of information learned before surgery, acquisition of new information and retention of information acquired after surgery. It is shown that although all three lesions impair acquisition of a specific type of new information, lesions of CA1 result in a severe retrograde amnesia but no forgetting of that type of information adequately acquired after surgery, whereas lesions of the DB do not cause retrograde amnesia but d

Pathology and immunohistochemistry of callitrichid hepatitis, an emerging disease of captive New World primates caused by lymphocytic choriomeningitis virus.

Callitrichid hepatitis is an arenavirus infection that recently emerged as a highly fatal disease of New World primates in the Callitrichidae family. As we previously reported, these primates develop hepatitis after contact with mice that are infected with variants of LCMV (LVMCCH), recently determined to have 86% identity with GC-P gene of the Armstrong and Western strains of LCMV. Here, we describe the histopathological lesions and tissue localization of viral antigens in confirmed cases of callitrichid hepatitis from recent outbreaks in two U.S. zoos. The liver in marmosets and tamarins wit

CRTC1 knockdown in the marmoset visual cortex induces neuronal IEG overexpression, HFOs, and neurodegeneration.

CRTC1 is highly expressed in the brain and functions as a coactivator of CREB, regulating transcription of genes essential for neuronal function and plasticity. To elucidate its role in the primate cortex, we performed CRTC1 knockdown using short hairpin RNA (shRNA) in the visual cortex (V1) of common marmosets (Callithrix jacchus). Unexpectedly, the knockdown caused widespread cFOS induction beyond the injection site. Concurrently, reduced NeuN expression, appearance of cell death markers, increased glial markers, and glial scarring were observed around the lesion. Longitudinal electrocortico

Exploring efficient and effective mammalian models for Alzheimer's disease.

The aim of this study was to explore and discuss efficient and effective mammalian models for Alzheimer's disease (AD). In this study, efficient AD models are characterized by a small body size, a short lifespan, and rapid development of the main pathology including amyloid plaque formation. Effective AD models are expected to exhibit not only the main pathology, but also co-pathology associated with other neurodegenerative diseases (e.g., Lewy body dementia), systemic disturbances such as disrupted central-peripheral homeostasis, and sleep-circadian failures. This reflects recent findings ind

Sarcocystid pathogens in captive monkeys from southern Brazil.

Non-human primates (NHP) can serve as intermediate or accidental hosts for sarcocystid pathogens, which vary in pathogenicity and relevance to wildlife and public health. This study aimed to detect DNA from the members of the Sarcocystidae family in tissue samples from 22 captive monkeys that died between 2019 and 2023 in a zoo located in southern Brazil. A total of 84 samples were analyzed using nested PCR, restriction fragment length polymorphism (RFLP), and specific primers to identify Sarcocystis spp., Toxoplasma gondii, Neospora sp., and Hammondia sp. According to our results, the DNA of

Improving Reproductive Success in Captive Marmosets Through Active Female Choice.

The recent upsurge in the use of common marmosets (Callithrix jacchus) as a desirable model for high-priority biomedical research has challenged local and global suppliers struggling to provide sufficient numbers of marmosets for large-scale projects. In laboratories, random male-female pairings are often unsuccessful, with intervals of several months before attempting alternate pairings. Here we address this challenge through a behavioral task that promotes self-directed female selection of potential mates to increase the efficiency of breeding in captive marmosets. We created a partner prefe

The regulation of maternal behaviour in marmosets and tamarins.

This article reviews a long-term investigation into the roles of (1) experience obtained with infants during development and postnatally, and (2) levels of reproductive-steroid hormones during late pregnancy, in the regulation of maternal caregiving motivation and behaviour in the red-bellied tamarin (Saguinus labiatus) and the common marmoset (Callithrix jacchus) (family Callitrichidae, Order Primates). Both observational studies, e.g. post hoc analysis of the relationship between developmental caregiving experience and infant-rearing success, and manipulative studies, e.g. direct analysis of

Transient changes in body weight and behavior during the placentation period in non-human primates and rodents.

During early pregnancy, many women experience physical changes, including nausea and vomiting of pregnancy (NVP), which negatively impact their quality of life. However, the absence of model animals has limited our understanding of such pregnancy-associated physiological changes. Here, we examined pregnancy-associated metabolic and behavioral changes in common marmosets and mice. Marmosets exhibited a transient weight decrease during the period of placental development in approximately 22% of pregnancies. Some marmosets repeatedly showed transient weight loss across multiple pregnancies, sugge

Medial premotor cortex as a potential reservoir of reward expectancy signals influencing the primate frontoparietal network.

Reward expectancy plays an important role in modulating cortical activity associated with motivation, selection, and preparation of subsequent goal-directed movements. However, it is unclear how it is represented in the primate frontoparietal cortex in a form that is separable from activity for motor execution. Here, using common marmosets performing a classical conditioning task with auditory cues, we conducted wide-field one-photon calcium imaging of frontoparietal and auditory cortices while recording orofacial and body movements. Reward expectancy that was separable from licking, eye movem

Human Herpesvirus 1 Associated with Epizootics in Belo Horizonte, Minas Gerais, Brazil.

Human activity in sylvatic environments and resulting contact with wildlife, such as non-human primates (NHPs), can lead to pathogen spillover or spillback. Both NHPs and humans host a variety of herpesviruses. While these viruses typically cause asymptomatic infections in their natural hosts, they can lead to severe disease or even death when they move into novel hosts. In early 2024, deaths of Callithrix penicillata, the black-tufted marmoset, were reported in an urban park in Belo Horizonte, Minas Gerais, Brazil. The epizootic was investigated in collaboration with CETAS/IBAMA and the Zoono

Erroneous predictive coding across brain hierarchies in a non-human primate model of autism spectrum disorder.

In autism spectrum disorder (ASD), atypical sensory experiences are often associated with irregularities in predictive coding, which proposes that the brain creates hierarchical sensory models via a bidirectional process of predictions and prediction errors. However, it remains unclear how these irregularities manifest across different functional hierarchies in the brain. To address this, we study a marmoset model of ASD induced by valproic acid (VPA) treatment. We record high-density electrocorticography (ECoG) during an auditory task with two layers of temporal control, and applied a quantit

Sustained Retinal Defocus Increases the Effect of Induced Myopia on the Retinal Astrocyte Template.

The aim of this article is to describe sustained myopic eye growth's effect on astrocyte cellular distribution and its association with inner retinal layer thicknesses. Astrocyte density and distribution, retinal nerve fiber layer (RNFL), ganglion cell layer, and inner plexiform layer (IPL) thicknesses were assessed using immunochemistry and spectral-domain optical coherence tomography on seventeen common marmoset retinas (Callithrix jacchus): six induced with myopia from 2 to 6 months of age (6-month-old myopes), three induced with myopia from 2 to 12 months of age (12-month-old myopes), five

A Common Marmoset Model of Mother-Infant Intervention for Breastfeeding Disorders in the Presence of Paternal Inhibition and Maternal Neglect.

Parents' psychological stress during the perinatal and neonatal periods continues to increase in an environment of declining birthrates, aging populations, and shrinking family sizes. The increase in child abuse and neglect cases, most likely by inexperienced and insufficiently knowledgeable parents, necessitates education on childcare and intervention techniques in nursing and midwifery training. In particular, attachment formation early in life between mother and infant is crucial. To accurately teach sensitive and comprehensive information on intervention techniques for mother-child attachm

The common marmoset in biomedical research: experimental disease models and veterinary management.

The common marmoset, Callithrix jacchus, is increasingly being used as the preferred nonhuman primate (NHP) model in biomedical research. Marmosets share several physiological and biological similarities with humans, as a Simiiformes species, and their use in research programs advances knowledge in several fields. Their unique characteristics, such as their small size, high fecundity, and rapid growth, offer additional advances in laboratory settings. This article reviews the developments in experimental disease models using marmosets based on our experience at the Central Institute for Experi
