Two point mutations convert a catalytically inactive carbonic anhydrase-related protein (CARP) to an active enzyme.

A murine carbonic anhydrase-related protein (CARP) has been expressed in Escherichia coli and purified to near homogeneity. The polypeptide chain consists of 290 amino acid residues and has a calculated molecular mass of 32,950 Da. By introducing two mutations, Arg117 --> His and Glu115 --> Gln, we created a metal-binding center homologous to that in the carbonic anhydrases from the animal kingdom. In contrast to unmodified CARP, this double mutant was isolated as a 1:1 zinc-protein complex. While unmodified CARP is catalytically inactive, the mutant catalyzes CO2 hydration with a significantl

[Transfer of the pronator teres tendon to the extensor carp radialis tendon in wrist flexion deformity in cerebral palsy].

The flexion deformity of the wrist in cerebral palsy is quite common, carrying disability to the effective use of the hands. From 1980 to 1990, 36 transferences of the pronator teres to the extensor carpi radialis were reviewed. The authors describe the operative technique by a unique incision, that may be used in cerebral palsy, and conclude that it is a good method for correction of deformity.

Inner plexiform circuits in the carp retina: effects of cholinergic agonists, GABA, and substance P on the ganglion cells.

Th effects on ganglion cell light responses and spontaneous activity of neurotransmitter candidates, applied by nebulizer spray and iontophoresis, were studied in the isolated carp retina. ACh, GABA, and substance P had strong effects on the ganglion cells; dopamine and the amino acids aspartate, glutamate, and glycine and only weak effects. ACh and substance P exerted their actions even when synaptic transmission was blocked by cobalt chloride, suggesting postsynaptic receptors for those agents on the ganglion cell membrane. The 3 amino acids and dopamine do not appear to act directly on the

Recombinant lactobacillus expressing G protein of spring viremia of carp virus (SVCV) combined with ORF81 protein of koi herpesvirus (KHV): A promising way to induce protective immunity against SVCV and KHV infection in cyprinid fish via oral vaccination.

Spring viremia of carp virus (SVCV) and koi herpesvirus (KHV) are highly contagious and pathogenic to cyprinid fish, causing enormous economic losses in aquaculture. Although DNA vaccines reported in recent years could induce protective immune responses in carps against these viruses via injection, there are a number of consequences and uncertainties related to DNA vaccination. Therefore, more effective and practical method to induce protective immunity such as oral administration would be highly desirable. In this study, we investigated the utilities of a genetically engineered Lactobacillus …

Microcystin-LR disrupts gut metabolic homeostasis through microbiota-metabolite axis remodeling in grass carp.

Microcystin-LR (MC-LR) exerts detrimental effects on gut mucosal integrity and functional homeostasis in aquatic organisms. This study investigated the gut toxicity of MC-LR in grass carp exposed to 5 and 10 μg/L MC-LR through integrated histological, biochemical, molecular, microbiomics, metabolomic, and cellular analyses. MC-LR exposure caused pronounced gut structural damage, characterized by epithelial disorganization, villus shortening, and crypt alteration. MC-LR exposure significantly reduced antioxidant enzyme activities (SOD, CAT, and GSH) and increased MDA levels in the gut, reflecti

Development and Validation of a Viability RT-qPCR Assay for Detecting Infectious Spring Viraemia of Carp Virus (SVCV).

Spring viremia of carp virus (SVCV) is a contagious pathogen associated with significant mortality and economic losses in freshwater aquaculture. Although reverse transcription quantitative PCR (RT-qPCR) enables rapid detection, it does not distinguish between infectious and non-infectious viral particles, which may lead to overestimation of infection risk. In this study, a viability RT-qPCR (vqPCR) assay targeting the glycoprotein (G) gene of SVCV was developed and analytically evaluated. Primers and probes were designed based on 45 full-length G-gene sequences representing genogroups Ia-Id t

Cryoprotection of Tremella polysaccharides in silver carp myofibrillar protein: stability improvement against freeze-thaw damage.

BACKGROUND: Myofibrillar proteins of silver carp are susceptible to freezing denaturation, significantly degrading surimi quality. Tremella polysaccharide (TP), a natural edible mushroom polysaccharide, offers potential cryoprotective value and can inhibit this deterioration. The aim of this study was to investigate the cryoprotective effect of TP on the myofibrillar protein of silver carp. RESULTS: Tremella polysaccharide effectively mitigated protein aggregation and freeze-induced denaturation. Specifically, 2.0% TP enhanced freeze-thawed protein solubility by 107.23%, reduced turbidity by

Synergistic Enhancement of Gel Properties in Unwashed Silver Carp Surimi by Hydroxypropyl Starch, Gallic Acid, and Xylitol.

While cost-effective and time-saving in production, unwashed surimi often exhibits inferior gel properties compared to its washed counterpart, thereby limiting its application in high-quality seafood products. To address this challenge, we systematically investigated the effects of hydroxypropyl starch (HPS, 2%), xylitol (6%), gallic acid (GA, 0.15%), and their ternary compound system (TC: 1.71% HPS + 0.14% GA + 6.57% xylitol) on the gel properties of unwashed silver carp surimi. A systematic analysis was conducted to elucidate the structure-property relationships. Microstructure analysis reve

Long non-coding RNA transcripts in crucian carp brain - annotation and expression patterns in anoxia and reoxygenation.

Long non-coding RNAs (lncRNAs) regulate diverse cellular processes, yet their role in anoxia-induced transcriptomic changes in crucian carp (Carassius carassius) - a species that endures months without oxygen at low temperatures - remains unclear. Because existing genome annotations focus on protein-coding genes, we aimed to annotate lncRNAs in crucian carp, assess their potential for involvement in mRNA regulation during anoxia and reoxygenation, and characterize differentially regulated lncRNAs and nearby putative interaction partners (IPs). Using next-generation RNA sequencing of the brain

A Teleost - Specific Sema4E - like Protein Promotes Grass Carp Reovirus Infection by Upregulating HSP70 Expression.

Hemorrhagic disease caused by grass carp reovirus (GCRV) poses a severe threat to the grass carp aquaculture industry. Semaphorin 4E-like (Sema4E-like), a teleost-specific signaling molecule, remains poorly understood in immune regulation. In this study, the grass carp Sema4E-like gene was cloned and characterized, revealing a protein with the canonical "Sema-PSI-Ig" domain architecture. Expression analysis revealed that Sema4E-like was highly expressed in the hepatopancreas and intestine and underwent notable subcellular relocalization following GCRV infection. In vitro experiments demonstrat

Integrated histological, biochemical and molecular analysis of grass carp (Ctenopharyngodon idella) responses to acute hypoxic stress.

Hypoxia critically impacts grass carp (Ctenopharyngodon idella) aquaculture, causing oxidative damage, mortality and economic losses. This study revealed the physiological and genetic response mechanisms of grass carp under hypoxic stress by integrating histology, biochemistry and miRNA-mRNA network analysis. The experimental individuals were confined in sealed containers with pre-aerated water, allowing dissolved oxygen (DO) to decline naturally, and three groups were established: Normoxia (DO: 7.99 ± 0.43 mg∙L-1; control), Hypoxia (DO: 0.88 ± 0.16 mg∙L-1; upward swimming and gulping), and As

Dose-Dependent Toxicity of Pyriproxyfen in Common Carp (Cyprinus carpio): Hematological, Biochemical, Genotoxic, and Molecular Responses.

This study evaluated both the acute and sublethal effects of pyriproxyfen in Cyprinus carpio using an integrated multibiomarker approach. The 96-h LC50 value was calculated as 4.525 mg/L. Following 14 days of sublethal exposure, pyriproxyfen caused clear, dose-dependent changes across hematological, biochemical, genotoxic, and molecular parameters. Hemoglobin and hematocrit levels declined progressively with increasing concentration, pointing to the development of anemia-like conditions. Biochemical analyses revealed a biphasic response pattern: Several parameters initially increased at lower

Dietary Ellagic Acid Protects and Ameliorates Copper Sulfate-Induced Toxicity in Common Carp (Cyprinus carpio).

Copper sulfate (CuSO4) is one of the most widely used chemotherapeutic agents in aquaculture and aquatic veterinary medicine because of its broad-spectrum antiparasitic, antifungal, and antimicrobial activities. However, repeated exposure may induce oxidative stress and impair hematological and immune functions, thereby compromising fish health. This study evaluated the effects of pre-exposure and post-exposure dietary ellagic acid (EA) supplementation on CuSO4-induced hematological, immunological, and oxidative stress alterations in common carp (Cyprinus carpio). A total of 150 fish were rand

A Z-DNA/PKZ-based immunoenhancer confers protective immunity against GCRV infection in grass carp.

Grass carp reovirus (GCRV) is a devastating pathogen that causes hemorrhagic disease in grass carp, resulting in mortality rates of up to 80% in fingerlings and substantial economic losses to the aquaculture industry. The development of a broad-spectrum immunoenhancer to boost intrinsic host defense mechanisms represents an urgent and commercially attractive strategy. In the present study, we constructed a Z-DNA/PKZ-based immunoenhancer by cloning GC-rich Z-DNA sequences and grass carp protein kinase Z (PKZ) into a eukaryotic expression vector, and investigated its potential as an immunoprophy

Grass carp (Ctenopharyngodon idella) WDFY1 activates IFN1 response via TLR3-TRIF-IRF3 axis.

Toll-like receptor 3 (TLR3) is a significant member of the TLR family which plays a vital role in pathogen-associated molecular patterns recognition and activation of innate immunity. In the past few years, TLR3 has been identified in a variety of fish, but its upstream adaptor is rarely reported. In this study, an adaptor protein, called WDFY1 (WD repeat and FYVE domain-containing protein 1), was cloned and identified in grass carp (Ctenopharyngodon idella) (CiWDFY1). Phylogenetic analysis revealed that fish WDFY1s are highly conserved and share a high level of sequence similarity to their ma
