Decreased striatal monoaminergic terminals in olivopontocerebellar atrophy and multiple system atrophy demonstrated with positron emission tomography.

We used [11C]dihydrotetrabenazine, a new ligand for the type 2 vesicular monoamine transporter (VMAT2), with positron emission tomography to study striatal monoaminergic presynaptic terminals in 4 patients with multiple system atrophy, 8 with sporadic olivopontocerebellar atrophy, and 9 normal control subjects. Specific binding in the striatum was significantly reduced in the multiple system atrophy patients as compared with the normal control group, with average reductions of 61% in the caudate nucleus (p = 0.002) and 58% in the putamen (p = 0.009). Smaller reductions were found in the sporad…

[Sites of seizure discharges after intracerebroventricular injection of anisodamine and the antagonism by diazepam in rabbits].

Anisodamine is a tropine alkaloid isolated from Scopolia tangutica Maxim. To determine the original sites of anisodamine seizure discharge, permanent electrodes were implanted into different parts of the brain in rabbits and the electrical activities were continuously recorded by monopolar leads. Injection of anisodamine 1.5 mg/kg into the lateral ventricle of conscious rabbits always produced abnormal discharges. The spike discharges appeared first in the amygdala and consisted of rhythmic large surface-positive spikes. Multiple spikes then appeared in the hippocampus, caudate nucleus, midbra…

Rat neural tissue cathepsin D: ultrastructural immunocytochemistry.

The cellular and subcellular localization of cathepsin D, an aspartyl endopeptidase, was investigated in the central and peripheral nervous systems of the rat by light and electron microscopic immunocytochemistry. The reaction of rabbit anti-rat brain cathepsin D within ventral cervical spinal cord, cerebellum, corpus callosum, caudate nucleus, optic nerve, trigeminal ganglion, fifth cranial nerve and sciatic nerve was localized with an indirect immunoperoxidase technique. A number of tissue processing methods were utilized, but only in tissues fixed in paraformaldehyde-lysine-periodate and se…

Recombinant human nerve growth factor is biologically active and labels novel high-affinity binding sites in rat brain.

Iodinated recombinant human nerve growth factor (125I-rhNGF) stimulated neurite formation in PC12 cell cultures with a half-maximal potency of 35-49 pg/ml, compared with 39-52 pg/ml for rhNGF. In quantitative ligand autoradiography, the in vitro equilibrium binding of 125I-rhNGF to brain sections showed a 10-fold regional variation in density and was saturable, reversible, and specifically displaced by up to 74% with rhNGF or murine NGF (muNGF). At equilibrium, 125I-rhNGF bound to these sites with high affinity (Kd 52-85 pM) and low capacity (Bmax less than or equal to 13.2 fmol/mg of protein)

Whole-brain neural connectivity to cholinergic neurons in the nucleus basalis of Meynert.

The cholinergic neurons in the nucleus basalis of Meynert (NBM) are a key structure in cognition, the dysfunction of which is associated with various neurological disorders, especially dementias. However, the whole-brain neural connectivity to cholinergic neurons in the NBM remains to be further and comprehensively researched. Using virus-based, specific, retrograde, and anterograde tracing, we illustrated the monosynaptic inputs and axon projections of NBM cholinergic neurons in choline acetyltransferase (ChAT)-Cre transgenic mice. Our results showed that NBM cholinergic neurons received main

Chronic food restriction increases fos-like immunoreactivity (FLI) induced in rat forebrain by intraventricular amphetamine.

Chronic food restriction enhances behavioral responsiveness to amphetamine and other abused drugs. Because this effect is evident when drugs are administered intracerebroventricularly (i.c.v.) as well as systemically, it would seem to reflect increased sensitivity of a neural substrate rather than a change in drug disposition. In the present study, c-Fos immunohistochemistry was used to evaluate whether the magnitude and pattern of cellular activation induced by i.c.v. amphetamine is altered by a regimen of food restriction previously shown to potentiate amphetamine reward. In the absence of a

Localization and connections of the tail of caudate and caudal putamen in mouse brain.

The neural circuits of the striatum (caudate and putamen) constitute a crucial component of the extrapyramidal motor system, and dysfunction in these circuits is correlated with significant neurological disorders including Parkinson's disease and Huntington's disease. Many previous studies in rodents revealed the neural connections of the rostral and intermediate parts of the striatum, but relatively fewer studies focused on the caudal striatum, which likely contains both the tail of caudate (CaT) and caudal putamen (PuC). In this study, we investigate the gene markers for the CaT and PuC and

Sensory and motor representations of internalized rhythms in the cerebellum and basal ganglia.

Both the cerebellum and basal ganglia are involved in rhythm processing, but their specific roles remain unclear. During rhythm perception, these areas may be processing purely sensory information, or they may be involved in motor preparation, as periodic stimuli often induce synchronized movements. Previous studies have shown that neurons in the cerebellar dentate nucleus and the caudate nucleus exhibit periodic activity when the animals prepare to respond to the random omission of regularly repeated visual stimuli. To detect stimulus omission, the animals need to learn the stimulus tempo and…

Identification of individual subjects based on neuroanatomical measures obtained 7 years earlier.

We analysed a dataset comprising 118 subjects who were scanned three times (at baseline, 1-year follow-up, and 7-year follow-up) using structural magnetic resonance imaging (MRI) over the course of 7 years. We aimed to examine whether it is possible to identify individual subjects based on a restricted number of neuroanatomical features measured 7 years previously. We used FreeSurfer to compute 15 standard brain measures (total intracranial volume [ICV], total cortical thickness [CT], total cortical surface area [CA], cortical grey matter [CoGM], cerebral white matter [CeWM], cerebellar cortex

Creutzfeldt-Jakob disease: high caudate signal on magnetic resonance imaging.

We have presented an unusual case of CJD in which the magnetic resonance images obtained four months before death revealed high signal intensity in the caudate nuclei bilaterally. These findings correspond to severe involvement of this region at autopsy. This radiologic pattern has not generally been included in the differential diagnosis of CJD.

Subcortical prosopagnosia and anosognosia.

A 59-year-old right-handed woman with acute hemorrhage affecting the head of the left caudate nucleus exhibited prosopagnosia and anosognosia. It is suggested that these symptoms are due to a disconnection syndrome affecting a putative cortico-limbic reticular loop involved in attention. Prosopagnosia can occur with lesions outside the main central visual pathways and is a material-specific motivational neglect to faces: alternatively prosopagnosia in this patient may represent abnormal gating sensory limbic activity since sensory (visual) modulated memory related cholinergic fibers arising fr…

Caudate hemorrhage.

We studied 12 patients with intracerebral hemorrhage in the head of the caudate nucleus. These cases accounted for 7% of a consecutive series of intracerebral hemorrhage. The clinical findings differed from more common varieties of supratentorial hemorrhage. All patients had acute onset of vomiting, headache, stiff neck, decreased level of consciousness, and behavioral changes in a pattern that simulated subarachnoid hemorrhage. Seven patients had gaze paresis and hemiparesis, with or without sensory loss. Two of these seven patients had, in addition, elements of Horner's syndrome. The prognos…

Alcohol interactions with brain opiate receptors.

Ethanol, added in vitro to mouse caudate membranes, inhibited high-affinity binding of 0.2 nM 3H-dihydromorphine (3H-DHM) over an ethanol concentration range of 250-1,000 mM. At lower, physiologically-attainable ethanol concentrations (e.g.: 50 mM), 3H-DHM binding was significantly increased. Over the concentration range of 50-1,000 mM, ethanol inhibited 0.5 nM 3H-[D-Ala2,D-Leu5]enkephalin (3H-ENK) binding to mouse caudate tissue, and no stimulation of 3H-ENK binding was noted at any of these concentrations of ethanol. Ethanol inhibits opiate binding in a pseudo-competitive manner and, therefo

Modulation of glutamatergic functional connectivity by a prototypical antipsychotic: Translational inference from a postsynaptic density immediate-early gene-based network analysis.

BACKGROUND: Although extensively studied, the effect of antipsychotics is not completely understood at a network level. We tested the hypothesis that acute administration of haloperidol would modulate functional connectivity of brain regions relevant to schizophrenia pathophysiology. To assess putative changes in brain network properties and regional interactivity, we studied the expression of Homer1a, an Immediate Early Gene (IEG) demonstrated to be induced by antipsychotic administration and coding for a protein involved in glutamatergic synapses remodeling. METHODS: Sprague-Dawley rats (n
