Distribution and properties of excitatory and inhibitory junction potentials in circular muscle of the guinea-pig stomach.

In the presence of guanethidine (10(-6)-5 X 10(-6) M), transmural nerve stimulation evoked an excitatory junction potential (e.j.p.) in the fundus region and an inhibitory junction potential (i.j.p.) in the antrum region of the circular muscle of the guinea-pig stomach. The e.j.p. was blocked by atropine (over 2 X 10(-7) M) while the i.j.p. was blocked by apamin (10(-7) M) but not by adrenergic or cholinergic receptor antagonists. Therefore the i.j.p. may be non-adrenergic and non-cholinergic in nature. In the presence of atropine, nerve stimulation evoked the non-adrenergic, non-cholinergic i

Involvement of neurotransmitters in the action of apelin-13 on passive avoidance learning in mice.

The widespread distribution of apelin-13 and apelin receptors in the brain suggests an important function of this neuropeptide in the brain that has not been explored extensively so far. In the present work, apelin-13 was found to facilitate the consolidation of passive avoidance learning in mice. In order to assess the possible involvement of transmitters in this action, the animals were pretreated with the following receptor blockers in doses which themselves did not influence the behavioral paradigm: phenoxybenzamine (a nonselective α-adrenergic receptor antagonist), propranolol (a β-adrene

Crosstalk through cholinergic and aryl hydrocarbon receptor pathways in the anti-inflammatory actions of microbial metabolites.

The intestinal epithelium maintains immune and metabolic homeostasis at the host-microbiota interface, and its disruption contributes to inflammatory bowel disease (IBD). Microbial metabolites such as short-chain fatty acids (SCFAs) and indole derivatives are known to regulate epithelial and immune functions, but their roles in cholinergic signaling and aryl hydrocarbon receptor (AhR) pathways remain incompletely understood. Caco-2 intestinal epithelial cells were stimulated with lipopolysaccharide (LPS; 10 µg/mL) and co-treated with SCFAs (acetate, propionate, butyrate) or indole derivatives

Influence of the Cholinergic System on the Pathogenesis of Glioblastoma: Impact of the Neutrophil Granulocytes.

Glioblastoma (GBM) is the most common malignant primary brain tumor in adults. Since numerous studies highlight the significance of cholinergic system components in tumor development, acetylcholine (ACh) and the differential activation of its receptors could play a crucial role in GBM progression. The aim of this study was to test this hypothesis by assessing the relevance of the cholinergic system in GBM cells and their microenvironment. We analyzed bulk RNA-seq expression data using the TIMER2.0 web server, focusing on the impact of patient survival in relation to muscarinic receptors (CHRM)

Passive transfer of Sjogren's syndrome IgG produces the pathophysiology of overactive bladder.

OBJECTIVE: The presence, in patients with primary and secondary Sjogren's syndrome (SS), of autoantibodies that acutely inhibit M(3) muscarinic receptor (M3R)-mediated bladder contractions is difficult to reconcile with the fact that symptoms of detrusor overactivity and other features of cholinergic hyperresponsiveness occur in this disease. This study was undertaken to examine the in vivo effects of these autoantibodies on bladder function by examining bladder responsiveness and compliance following passive transfer of patient IgG to mice. METHODS: Contractile responses of isolated bladder

Subanaesthetic ketamine treatment alters prefrontal cortex connectivity with thalamus and ascending subcortical systems.

BACKGROUND: Acute treatment with subanaesthetic doses of NMDA receptor antagonists, such as ketamine, provides a translational model with relevance to many of the symptoms of schizophrenia. Previous studies have focused specifically on the prefrontal cortex (PFC) because this region is implicated in many of the functional deficits associated with this disorder and shows reduced activity (hypofrontality) in schizophrenia patients. Chronic NMDA antagonist treatment in rodents can also induce hypofrontality, although paradoxically acute NMDA receptor antagonist administration induces metabolic hy

Intracellular calcium release mediated by noradrenaline and acetylcholine in mammalian pineal cells.

The effects of noradrenergic and cholinergic receptor agonists on intracellular Ca2+ concentration ([Ca2+]i) in single dissociated rat pineal cells were investigated by microfluorimetric measurements in Fura-2 acetoxymethyl ester (Fura-2/AM) loaded cells. Noradrenaline (NA) evoked characteristic biphasic increments of intracellular Ca2+ consisting of one or more leading spikes followed by a plateau, resulting from the release of Ca2+ from intracellular stores and from the influx of Ca2+ from the external medium, respectively. This response was reproduced by the alpha 1-adrenoceptor agonist, ph

Influence of serotonin on the human ureter: an in vitro pharmacological study.

The influence of serotonin (5-hydroxytryptamine, 5-HT) on human ureteral smooth muscle was explored in a series of in vitro experiments. 5-HT evoked a dose-dependent contraction of the ureter. The effect of 5-HT was unaltered by blockade of 5HT3 and 5HT4 receptors and muscarinic cholinergic receptors. The 5HT2 receptor antagonist ketanserin (KT; 10(-5)-10(-4) M) and mixed 5HT1/5HT2 receptor antagonist methysergide (MS; 5 x 10(-5) M) inhibited the effect of 5-HT. Since the antagonist dissociation constant at 5HT2 receptors for both KT and MS is found to be approximately 10(-9) M, it is conclude

Human amylin actions on rat cholinergic basal forebrain neurons: antagonism of beta-amyloid effects.

Human amylin (hAmylin), a 37-amino acid pancreatic peptide, and amyloid beta protein (A beta), a 39-43 amino acid peptide, abundantly deposited in the brains of Alzheimer's patients, induce neurotoxicity in hippocampal and cortical cultures. Although the mechanism of this neurotoxicity is unknown, both peptides are capable of modulating ion channel function that may result in a disruption of cellular homeostasis. In this study, we examined the effects of hAmylin on whole cell currents in chemically identified neurons from the rat basal forebrain and the interactions of hAmylin-induced response

Electrically evoked release of glutamate in rat hippocampal slices: effects of various drugs and fimbria-fornix lesions.

A model of electrically evoked release of glutamate from rat hippocampus was developed and used to detect possible changes induced by lesions of hippocampal afferences. Neuronal glutamate in hippocampal slices was labelled by preincubation with [3H]glutamine. The slices were then superfused with physiological medium in the presence of the glutamate uptake inhibitor L-transpyrrolidine-2,4-dicarboxylic acid (100 microM or 3 microM) and stimulated twice electrically (S1, S2: 240 pulses, 3 Hz, 2 ms, 26-30 mA); various drugs were added before S2. Pharmacologic characteristics of Meige dystonia: differentiation from tardive dyskinesia.

Idiopathic orofacial dyskinesia, also called Brueghel's syndrome, blepharospasm-oromandibular dystonia, and Meige dystonia, is characterized by involuntary facial movements. Since this disorder can be difficult to distinguish from tardive dyskinesia, we have generated a neuropharmacologic profile of Meige dystonia. Symptoms were improved by antagonists of both dopamine and acetylcholine and worsened by the cholinergic agonist physostigmine, consistent with a hypothesis of relative excess in both dopamine and acetylcholine neuronal activities. Since tardive dyskinesia is hypothesized to be char

Involvement of neurotransmitters in the 'anxiolytic-like' action of atrial natriuretic peptide in rats.

Effects of centrally administered rat atrial natriuretic peptide (ANP1-28) in different doses (50, 100, 150, 200, 500 or 1000 ng) were examined in rats with respect to anxiolytic properties in an elevated plus-maze model. In doses of 100, 150 and 200 ng, ANP1-28 abolished the normal preference for the closed arms of the maze, and increased the percentage of time spent on the open arms; this is consistent with an 'anxiolytic-like' effect. Doses of 50, 500 and 1000 ng of rANP1-28 produced no behavioral effects in the elevated plus-maze model. Pretreatment with a dopaminergic blocker, an alpha-ad

Orally administered atropine enhances motor cortex excitability: a transcranial magnetic stimulation study in human subjects.

Oral application of atropine was used to test if a modulation of cholinergic neurotransmission changed motor excitability. Healthy volunteers received either 1 or 2 mg atropine. Paired transcranial magnetic stimulation was used to study intracortical inhibition and intracortical facilitation before, 1 h and 24 h after ingestion of atropine. In addition, the silent period, motor threshold, F wave and motor response amplitudes were measured. The 1 mg dose of atropine induced a loss of intracortical inhibition, the 2 mg dose produced an intracortical disinhibition and enhanced intracortical facil

Nicotine-evoked nitric oxide release in the rat hippocampal slice.

The effects of cholinergic agonists on nitric oxide (NO) release in hippocampal slices from male Sprague-Dawley rats were investigated using electrochemical recording procedures using Nafion and O-phenylenediamine-treated carbon fiber microelectrodes. These microelectrodes are highly selective for NO versus other interferents. Acetylcholine (Ach) with neostigmine, or nicotine was delivered by pressure ejection from pipettes placed within 300 microm of the NO sensors. Both Ach and nicotine produced NO signals ranging from 0.04 to 2.14 microM in the CA1, CA3, and dentate gyrus of the rat hippoca

Histamine-evoked potassium release in the mouse and guinea pig pancreas.

An investigation was made of the effects of histamine on the K+ concentration in the effluent in superfused guinea pig and mouse pancreatic segments. The effect of acetylcholine (ACh) was examined for comparison. Histamine evoked a dose-dependent and transient increase in the K+ concentration in the effluent (K+ release) but is less potent compared to the cholinergic agonist, ACh. At the same doses histamine and ACh evoke a much larger K+ release from mouse superfused pancreatic segments followed in the poststimulus period by a reuptake of K+. However, this reuptake of K+ was not observed in g

A quantitative autoradiographic study of muscarinic cholinergic receptor subtypes in the brains of pyrithiamine-treated rats.

Previous studies describe decreased acetylcholine synthesis in brain as well as neurobehavioral evidence for a central muscarinic cholinergic deficit in pyrithiamine-induced thiamine-deficient rats. In order to further evaluate this possibility, quantitative autoradiographic procedures using [3H]quinuclidinyl benzilate (for total muscarinic binding sites), [3H]pirenzepine (for muscarinic M1 sites) and [3H]AF-DX 384 (for muscarinic M2 sites) were performed at early (presymptomatic) and late (symptomatic) stages of thiamine deficiency induced in rats by administration of the central thiamine ant

Concurrent Use of Anticholinergic and Antidementia Medicines in Older Adults with Dementia: A Systematic Review of Prevalence, Predictors and Clinical Outcomes.

Background: Concurrent use of acetylcholinesterase inhibitors (AChEIs) and medicines with anticholinergic activity is a clinically important medication safety concern in dementia care because these medicines may pharmacologically oppose dementia treatment and increase the risk of avoidable harm. This review systematically synthesised evidence on the prevalence, patterns, predictors and clinical consequences of concurrent use of anticholinergic and anti-dementia drugs (ADDs) in older adults with dementia. Methods: A PRISMA-informed systematic review was conducted employing Ovid MEDLINE, Embase

New Advances in Drug Research for Myopia Control in Adolescents.

BACKGROUND: Myopia is one of the most common eye diseases worldwide, with an increasing incidence observed in recent years. Globally, effective treatments for myopia have been extensively explored. In recent years, research on drugs for the treatment of myopia has become a popular topic in ophthalmology, with some breakthroughs having been achieved. Compared with surgical treatment, drug treatment is easier for people to accept. Although the efficacy of some drugs in delaying the development of myopia has been confirmed, the mechanism and site of action of some drugs are still not completely c

Development and Pilot Testing of an Algorithm-Based Approach to Anticholinergic Deprescribing in Older Patients.

BACKGROUND: Adverse anticholinergic drug reactions are common, yet evidence on how to reduce exposure to anticholinergic activity and reliably measure successful deprescribing is still scant. This study proposes an algorithm-based approach to evaluate and reduce anticholinergic load, and reports the results of its pilot testing. METHODS: Based on published evidence and expert opinion, a list of 85 anticholinergic drugs and 21 algorithms for reducing anticholinergic load, e.g., by recommending alternative drugs with lower risk, were developed. An accompanying test battery was assembled by focu
