We have used a preparation of soluble human insulin receptor ectodomain and a novel photoreactive, biotinylated derivative of insulin (4-azidosalicyloyl(B1-biocytinyl-B2-lysine)-insulin) to identify a new hormone contact site within the extracellular domain of the insulin receptor. The ectodomain was photoaffinity-labeled and digested to completion with trypsin, and the resulting tryptic fragment was purified by either HPLC or by streptavidin-affinity chromatography. Neuron-specific enolase and phosphoglycerate mutase with specific activities of 106 and 215 U/mg, respectively, have been purified from human brain. Hydrophobic chromatography for enolase and blue Sepharose affinity chromatography for phosphoglycerate mutase were used as the last steps of purification. A heterobifunctional complex with fully preserved enolase and phosphoglycerate mutase activities was synthesized with the use of a bifunctional reagent, N-succinimidyl 3-(2-pyridyldithio)propionate. Omentum tissue has potent angiogenic activity, so transplantation is often effective in treatment of moyamoya disease. BACKGROUND: About 50% of populations in developed countries have bile supersaturated with cholesterol, which is a major risk factor for cholesterol gallstone formation. Despite the prevalence of supersaturated bile, only about 10% of these populations develop gallstones. Human choriogonadotropin (HCG) of high immunoactivity was isolated from hydatidiform mole by acid and salt precipitation, immunoaffinity and DEAE-Sephadex chromatography. Polyacrylamide gel electrophoresis, immunodiffusion, immunoelectrophoresis and N-terminal amino acid analyses showed that the product obtained is essentially homogeneous. Molar HCG was found to resemble HCG from urine of normal pregnant women in amino acid composition bu to differ from it in having a lower content of mannose and N-acetylglucosamine.
