A periapical Actinomyces colony was studied by immunoelectron microscopy with a specific antiserum against Actinomyces israelii. Positive as well as some negative micro-organisms were seen within the colony. Reacting antigens were localized in the cytoplasm and the cell wall of the bacteria, which suggests that more than one antigen is involved in the reactions with the present antiserum. The ultrastructural results confirmed the applicability of the peroxidase method with Actinomyces antiserum in paraffin sections for diagnostic purposes.

The present studies assessed the relationship between murine splenic B-lymphocyte colony-forming cells and the number of mature B lymphocytes in the spleen following administration of either cyclophosphamide or azathioprine. Cyclophosphamide (200 mg/kg) reduced the number of splenic B lymphocytes by a mean of 77%. However, a greater reduction in colony-forming cells was noted. Five days after cyclophosphamide administration, the number of colonies per spleen was reduced by more than 90%. Furthermore, the recovery of colony-forming activity was delayed in comparison with restoration of the B-ce

Cells in T cell colonies grown in vitro from normal human peripheral blood leukocytes can kill a variety of different target cells against which the blood donor has not knowingly been previously sensitized, i.e., the colony cells exhibit spontaneous cytotoxicity. Tumor target cells are preferentially killed. The ability to kill two different targets segregates independently in different colonies, which implies that the effector cells are specificity restricted. A majority (perhaps all) of the effector cells possess receptors for sheep erythrocytes and do not possess receptors for the Fc end of

Thiamine deficiency encephalopathy occurred in a specific-pathogen-free cat colony where the diet consisted exclusively of commercially canned diet. This product was described by the manufacturer as a complete diet for cats. The clinical diagnosis was confirmed by the demonstration of bilaterally symmetrical hemorrhage and microvascular fibrinoid necrosis in the brain and low erythrocyte transketolase levels. Treatment of clinically ill cats with vitamin B complex was followed by recovery within 24 hours.

First, lymphocyte transplantation into total body-irradiated rats is discussed. The effect on spleen colony formation caused by the transplantation of untreated lymphocytes, as well as of lymphocytes previously incubated with PHA, or with PHA plus L-asparaginase, or with lymphokines, was studied. Then the effect of the urinary colony-stimulating factor in vitro, and the in vitro feeder-layer activity of leucocytes on colony formation of human and mice bone marrow cells in haematological diseases is dealth with. The injection of rat lymphocytes previously incubated for 24 hours with PHA resulte

Lethally whole-body irradiated mice upon receiving a relatively small quanity of marrow cells show a distinct sex difference in ability to support colony formation in spleen and cell proliferation in femoral marrow. The difference should be defined with reference to the organ in question. Female mice manifest more efficient splenic haematopoiesis while among the males myeloid haematopoiesis, as measured by 59Fe and 125IUdR incorporation, seems to be greater. Endotoxin treatment and hypertransfusion exert profound influences, interacting with the sex-hormonal status of the animal.

Colony stimulating activity (CSA) was estimated in daily urine samples and weekly plasma specimens of a 4-year-old girl with familial cyclic neutropenia. Nadirs of neutrophils and peaks of monocytes were found in regular cycles of 28 days. A close correlation was observed between nadirs of neutrophils and peaks of blood monocytes and colony stimulating activity in the urine. The role of colony stimulating factor as regulatory factor in granulopoiesis in cyclic neutropenia is discussed.

A child with repeated infections was immunologically normal but was found to have neutropenia with periodic elevations of the absolute mature polymorphonuclear count at 21-day intervals. Immediately following the PMN rise, bone marrow morphology and in vitro cultures demonstrated a maturation arrest at the myelocyte stage with an increase in proliferative capacity. His cycle was not altered by infusions of normal plasma or by injections of epinephrine or typhoid vaccine. Infusion of 10 ml/kg of "stimulated" plasma from donors reactive to TV, obtained 60 minutes following immunization, resulted

The in vitro susceptibility of murine myeloid colony-forming cells to the antiproliferative activities of three ansamycin antibiotics was determined. These cells were found to be 10- to 40-fold more susceptible than the corresponding human ones.

A series of febrile illnesses, some severe, among hospital personnel was traced to an outbreak of lymphocytic choriomeningitis in a hospital research colony of Syrian hamsters. By rapid virus isolation and identification techniques, a definitive diagnosis was made 5 days after specimens were received. This outbreak emphasizes the dangers of working with experimental hamsters and tumor cell lines passaged in them.

Intraperitoneal injection of complete Freund's adjuvant which caused an extensive cellular response in the reticuloendothelial system greatly increased the severity of infection which resulted from footpad inoculation of ectromelia virus in mice. The mortality was greatest during the proliferative phase of the cellular response, indicated by the colony-forming cell response. Exposure of colony-forming cells to virus in vitro showed that those from normal bone marrow were much less susceptible to virus than those from the bone marrow of adjuvant-treated mice and more particularly those from the

The effects of phytohemagglutinin (PHA) were studied in irradiated mice to see if a definite myeloproliferative effect could be demonstrated in vivo. The data obtained suggested the following conclusions. PHA treatment of the bone marrow donor only, causes a consistent but slight reduction in transplantable spleen colony-forming unit (CFU) content of the bone marrow 24 hr after the last PHA injection, but no change was found in the proportion of the various colony types. PHA treatment of the irradiated recipient of normal bone marrow causes no change in the number of spleen colonies. However,
