Omega Fatty Acid and Protein Profiles of Colostrum and Transitional Milk in Mexican Women With and Without Gestational Diabetes: A Cross-Sectional Study.

Background/Objectives: Gestational Diabetes Mellitus (GDM) involves metabolic alterations that may affect breast milk composition. Imbalances in protein and fatty acid (FA) profiles have been reported in mature milk from mothers with GDM. However, evidence for colostrum and transitional milk is limited, despite the key role of ω-3 and ω-6 Polyunsaturated fatty acids (PUFAs) in neonatal neurodevelopment. This study compared ω-3 and ω-6 PUFAs and protein concentrations in colostrum and transitional milk from women with and without GDM. Methods: This cross-sectional study was conducted from Janua

Gestational diabetes mellitus induced variation in the macro-and micro-heterogeneity of N-glycoproteome in human milk from colostrum to mature milk.

Gestational diabetes mellitus (GDM) represents a systemic metabolic disorder manifesting during pregnancy, with its global prevalence rising annually. This study aimed to systematically investigate the differences in the glycoproteome of human milk from healthy (H) mothers and mothers with GDM, focusing on the dynamic changes from colostrum to mature milk by using LC-MS/MS. In total, for healthy mothers, 2474 glycopeptides and 249 glycosites were identified in colostrum and 1064 intact glycopeptides and 163 glycosites were identified in mature milk. For mothers with GDM, 1309 glycopeptides and

Utilization of Dairy By-Products as a Source of Functional and Health Compounds-The Role of Ovine Colostrum and Milk Whey on Chronic Myeloid Leukemia Cells.

Nowadays, the search for food products that promote consumers' health has gained interest, and dairy by-products, due to their biological quality, could have a prominent position among products with health benefits. However, little is known about their activity on cancer cells. This study aimed to provide evidence about the effect of ovine colostrum and milk whey on K562 cells, a model of the human chronic myeloid leukemia cell line. The exposure of K562 cells to a single administration of sheep by-products at different concentrations for three days and three treatments for three days was carr

Short-term feeding of defatted bovine colostrum mitigates inflammation in the gut via changes in metabolites and microbiota in a chicken animal model.

BACKGROUND: Nondrug supplement strategies to improve gut health have largely focused on the effects of individual compounds to improve one aspect of gut homeostasis. However, there is no comprehensive assessment of the reproducible effects of oral, short-term, low-level colostrum supplementation on gut inflammation status that are specific to the ileum. Herein, a chicken animal model highly responsive to even mild gut inflammatory stimuli was employed to compare the outcomes of feeding a standard diet (CON) to those of CON supplemented with a centrifuge-defatted bovine colostrum (BC) or a nonf

Factors associated with passive immunity transfer in dairy calves: combined effect of delivery time, amount and quality of the first colostrum meal.

Despite the well-known importance of an adequate colostral immunoglobulin (Ig) transfer to calf health and survival, failed transfer of passive immunity (FTPI) remains a widespread problem in dairy farming. The aim of this study was to investigate the management factors associated with FTPI in newborn calves, evaluating particularly the combined effect of delivery time, amount and quality of the first colostrum meal. The study was conducted from March to August 2014 on 21 Italian dairy farms. Farmers were asked as first to answer a farm-level questionnaire on calf management. Blood sampling wa

Short communication: Supplementation of colostrum and milk with 5-hydroxy-l-tryptophan affects immune factors but not growth performance in newborn calves.

In ruminants, colostrum is the main source of immunoglobulins for the newborn animal, conferring immune protection until the immune system becomes active and able to synthesize its own immunoglobulins. Serotonin (5-HT), a biogenic amine derived from tryptophan, has stimulatory effects on many physiological processes, including components of the innate (mastocytes, eosinophils, and natural killer cells) and adaptive (T and B lymphocytes) immune systems. Based on the known effects of 5-HT on the immune system, we hypothesized that increased concentrations of 5-HT, through administration of its p

Fetal organ growth in response to oesophageal infusion of amniotic fluid, colostrum, milk or gastrin-releasing peptide: a study in fetal sheep.

The hypothesis of the present study was that the infusion of the biological fluids to which the developing gut is normally exposed (i.e. amniotic fluid, colostrum, milk) and a single growth factor (gastrin-releasing peptide), which is found in high concentrations in fetal fluids and milk, could ameliorate the altered growth induced by the elimination of swallowed input secondary to ligation of the oesophagus. At 108-110 days of gestation the fetal oesophagus was ligated and a catheter inserted towards the stomach (32 fetuses). At 117-119 days of gestation saline (n = 5), amniotic fluid (n = 5)

Whole-Course-Repair Wound Dressing Based on Bovine Colostrum and Hyaluronic Acid for Diabetic Wound Repair Through Immunomodulation, Angiogenesis, and Re-Epithelization.

Excessive immune response, obstruction of angiogenesis, degradation of collagen deposition, and delayed re-epithelialization are the primary factors obstructing diabetic wound healing. Adequate and inclusive strategies are needed to address these challenges comprehensively. In this study, a whole-course-repair wound dressing (BC/HA) was simply fabricated to enhance diabetic wound closure. The BC/HA wound dressing, created by combining bovine colostrum (BC) with hyaluronic acid solution (HA), demonstrated good cytocompatibility, endothelial cell migration, angiogenesis, and antibiofilm effects.

Coxiella burnetii seroconversion in neonatal calves in an infected dairy herd attributable to ingestion of imported colostrum replacer rather than vertical transmission.

In April-June 2017, a cluster of 4 third-trimester abortions caused by Coxiella burnetii was diagnosed on a dairy farm with 356 Holstein cows in Uruguay. We investigated 1) the seroprevalence of C. burnetii in cows delivering normal calves in this herd, 2) whether non-aborting cows gave birth to seropositive and/or PCR-positive newborn calves in the year of the abortion cluster, and 3) whether seronegative newborn calves seroconverted after ingesting colostrum replacer. In March-September 2017, serum samples were collected from 133 calving cows and their calves immediately after birth and befo

Colostrum insulin supplementation to neonatal Holstein bulls affects small intestinal histomorphology, mRNA expression, and enzymatic activity with minor influences on peripheral metabolism.

The objectives of this study were to evaluate how varying colostral insulin concentrations influenced small intestinal development and peripheral metabolism in neonatal Holstein bulls. Insulin was supplemented to approximately 5× (70.0 μg/L; n = 16) or 10× (149.7 μg/L; n = 16) the basal colostrum insulin (12.9 μg/L; BI, n = 16) concentration to maintain equivalent macronutrient intake (crude fat: 4.1 ± 0.06%; crude protein: 11.7 ± 0.05%; and lactose: 1.9 ± 0.01%) among treatments. Colostrum was fed at 2, 14, and 26 h postnatal and blood metabolites and insulin concentration were measured at 0,

Peptidomics-guided 3D structural-mechanistic investigation of bovine colostrum peptides reveals multi-targeted in silico and in vitro antidiabetic and antioxidant potential.

Multi-target inhibition of α-amylase, α-glucosidase, and human aldose reductase can significantly address postprandial hyperglycemia and subsequent diabetic complications, yet bovine colostrum peptides remain largely unscreened against these enzymes. Using a peptidomics-guided, integrated in silico and in vitro approach, we identified three lead peptides and highlighted Peptide P2 (GPAGPQGPR) as a common lead inhibitor, showing favorable predicted binding and interaction profiles relative to the reference inhibitors. A dedicated 3D-structural analysis, including residue mapping, RMSF profiles,

Unveiling peptidomic and metabolomic profile from camel milk colostrum and mature camel milk: Insights into cholesterol esterase and pancreatic lipase inhibitory effect.

This study investigated cholesterol esterase (CE) and pancreatic lipase (PL) inhibitory properties of camel colostrum (CC) and mature camel milk (MCM) following simulated gastrointestinal digestion (SGID), based on the hypothesis that SGID releases bioactive peptides that may enhance CE and PL inhibitory activities. Both simulated gastric digestion (SGD) and SGID digesta exhibited potent CE and PL inhibitory activities. Peptide sequences with peptide ranker scores >0.5 were considered as potentially bioactive peptides. Molecular docking demonstrated strong interactions between specific peptide

Characterization and comparison site-specific N-glycosylation profiling of milk fat globule membrane proteome in donkey and human colostrum and mature milk.

Milk fat globule membrane (MFGM) proteins are highly glycosylated and involved in various biological processes within the body. However, information on site-specific N-glycosylation of MFGM glycoproteins in donkey and human milk remains limited. This study aimed to map the most comprehensive site-specific N-glycosylation fingerprinting of donkey and human MFGM glycoproteins using a site-specific glycoproteomics strategy. We identified 1,360, 457, 2,617, and 986 site-specific N-glycans from 296, 77, 214, and 196 N-glycoproteins in donkey colostrum (DC), donkey mature milk (DM), human colostrum

Graduate Student Literature Review: Reducing mortality and morbidity in transported preweaning dairy calves: Colostrum management and pretransport nonsteroidal anti-inflammatory drug administration.

Currently, mortality and morbidity rates for preweaning calves in the US dairy industry are high, with the major cause being digestive disorders and respiratory diseases. One of the most important management practices that can reduce calf mortalities and morbidities is the feeding of colostrum, provided its quantity, quality, and cleanliness, and timing of feeding are according to recommendations. However, other management practices similar to transportation, can also compromise calf health and production performance. When preweaning calves are transported, stressors similar to physical restra
