Monoclonal antibodies specific for Corynebacterium sepedonicum, the causative agent of potato ring rot.

BALB/c mice were immunized with Corynebacterium sepedonicum, and spleen cells from the immunized animals were fused with cells of the mouse myeloma line P3-X63-Ag8.653. Several hybridoma cell cultures were selected for further study. Monoclonal CS-B-5 was specific for C. sepedonicum and did not react significantly with other closely related phytopathogenic corynebacteria in an enzyme-linked immunosorbent assay (ELISA). As few as 10(3) organisms could be detected. This approach should prove useful for developing improved diagnostic procedures for a number of bacterial plant pathogens.

Protection of mice against intestinal amoebiasis with BCG, Corynebacterium parvum and Listeria monocytogenes.

Treatment with Corynebacterium parvum or BCG, or infection with live Listeria monocytogenes was found to protect mice against subsequent infection with Entamoeba histolytica. Complete protection was obtained in mice treated with 10(7) (colony forming units) of BCG but not with 10(5). Partial protection was achieved with 10(6) of BCG. These data provide evidence that non-specific immunity plays an important role in the host defense against amoebic infection.

Anemia and thrombocytopenia from Corynebacterium parvum-stimulated resistance against malaria, trypanosomiasis, and babesiosis.

Nonspecific immunity (NSI) was manifested in rats injected intravenously with killed Corynebacterium parvum and challenged with Trypanosoma lewisi, Plasmodium chabaudi, or Babesia rodhaini. The NSI became evident some 5 days after infection as a suppressed parasitemia, a more rapid recovery from patent infection and as enhanced survival among rats infected with B. rodhaini. The C. parvum injections produced anemia and thrombocytopenia with splenomegaly and signs of glomerulonephritis in rats. The signs became evident about 5 days after injection and were accompanied by reduced titers of lytic …

Protection of mice against Babesia spp. and Plasmodium spp. with killed Corynebacterium parvum.

Mice which had been pre-treated with killed Corynebacterium parvum given intravenously or intraperitoneally, but not subcutaneously, were completely resistant to infection with Babesia microti or B. rodhaini, and were protected from death caused by Plasmodium vinckei or P. chabaudi infection. There is evidence that the parasites died within circulating erythrocytes. This occurred much too soon for a specific antibody response to be evoked, and no antibody could be detected by the indirect fluorescent-antibody technique. Thus it is suggested that a non-specific soluble mediator may play an impo…

Analysis of protein condensation for enzymes involved in extracellular glutamate overproduction in Corynebacterium glutamicum.

During extracellular glutamate overproduction in Corynebacterium glutamicum, an increase in metabolic flux toward glutamate synthesis occurs without an increase in the transcriptional or translational levels of relevant enzymes, suggesting the involvement of unprecedented post-translational regulatory mechanisms. Here, we investigated the relationship of enzyme condensation with the metabolic shifts that lead to glutamate overproduction. We systematically analyzed the subcellular localization of enzymes involved in glutamate overproduction using fluorescent protein tagging to identify enzyme c…

Prosthetic joint infection by Corynebacterium striatum: a case report on a rare and emerging pathogen.

Corynebacterium striatum, a non-diphtherial Gram-positive bacillus traditionally considered a skin commensal, is increasingly recognised as an opportunistic pathogen in device-associated infections. We report a polymicrobial prosthetic joint infection in a 73-year-old woman with prior bilateral total knee replacement. C. striatum was repeatedly isolated from synovial fluid and deep tissue samples with Pseudomonas aeruginosa and identified by MALDI-TOF mass spectrometry. The isolate was susceptible to glycopeptides and linezolid. The patient received intravenous teicoplanin and revision surgery…

Metabolic engineering of Corynebacterium glutamicum for production of myo-Inositol and D-chiro-Inositol.

D-chiro-inositol (DCI), a key signaling molecule in glucose and lipid metabolism and steroid synthesis, plays important roles in ameliorating human metabolic disorders. To achieve de novo DCI biosynthesis in Corynebacterium glutamicum, the inositol catabolism-blocked chassis strain DCI‑1 was engineered by dynamically downregulating pgi, pfkA, and zwf genes. The most effective strain DPa‑1 produced 2.89 g/L myo-inositol (MI), the direct precursor of DCI, and 0.83 g/L DCI, with the total inositol over four times the control. Subsequently, to promote the conversion of MI to DCI and the supply of …

Corynebacterium silvaticum in a game-handling butcher: A new zoonotic pathogen of human infections.

The recently described Corynebacterium (C.) silvaticum is a diphtheria toxin (tox-) gene bearing species so far mainly found in wild boar and deers. Due to its close relationship to C. ulcerans a zoonotic potential has been proposed. To date, only two cases of human C. silvaticum infections have been reported in the literature. Here we present the case of a 63-year-old male game-hunting butcher with recurrent and difficult-to-treat axillary lymphadenitis and abscess formation. This case underlines the zoonotic potential of this novel member of the C. diphtheriae species complex and suggests zo…

Here, we reinvestigated the function of TmaT in Cglu and found that it and several factors encoded in the tmaT locus are specifically required for mycolate transport via the CmpL4 transporter pathway. Notably, one of these addit …

Epidemiological and molecular characterization of multidrug-resistant Corynebacterium striatum in a tertiary hospital in China.

Corynebacterium striatum has emerged as an important opportunistic nosocomial infection; however, epidemiological and antimicrobial resistance data from China remain limited. This study explored the epidemiological distribution, antimicrobial susceptibility profiles, resistance genes, virulence genes, plasmid characteristics, and phylogenetic relationships of clinical C. striatum isolates from a tertiary hospital in northern China. A retrospective study was conducted on C. striatum isolates recovered between February 2019 and December 2021 from patients with respiratory tract infections, urina…

Corynebacterium minutissimum as a Rare Cause of Tibial Osteomyelitis: A Case Report and Literature Review.

Osteomyelitis is commonly caused by pathogens like Staphylococcus aureus, but rare organisms such as Corynebacterium minutissimum, typically associated with superficial skin infections, can also be implicated. Recognizing these atypical pathogens presents diagnostic and therapeutic challenges, especially in the presence of orthopedic hardware. We conducted a literature review yielding 25 studies and encompassing 797 patient cases, which highlights the emerging role of Corynebacterium species in osteomyelitis, particularly following trauma or surgical interventions. A 72-year-old man with a his…

Multiple subcutaneous abscesses by Corynebacterium amycolatum in a patient with severe idiopathic aplastic anaemia.

Corynebacterium amycolatum is a part of the normal skin flora and has been underestimated as a pathogen. However, in recent years, the species has gained recognition as an important pathogen causing severe infections, particularly in immunocompromised patients. Nevertheless, identifying these organisms at the species level is difficult in routine clinical microbiology, leading to limited knowledge of their clinical manifestations in infectious diseases. In this study, we report a rare case of multiple subcutaneous abscesses in a patient with severe neutropenia, wherein C.amycolatum was identif…

A multiomic approach to defining the essential genome of the globally important pathogen Corynebacterium diphtheriae.

Diphtheria is a respiratory disease caused by Corynebacterium diphtheriae. While the toxin-based vaccine has helped control outbreaks of the disease since the mid-20th century there has been an increase in cases in recent years, including systemic infections caused by non-toxigenic C. diphtheriae strains. Here we describe the first study of gene essentiality in C. diphtheriae, providing the most-dense Transposon Directed Insertion Sequencing (TraDIS) library in the phylum Actinobacteriota. This high-density library has allowed the identification of conserved genes across the genus and phylum w…

Potential in vitro action of an adenosine analog and synergism with penicillin against Corynebacterium pseudotuberculosis.

Caseous lymphadenitis is a well-known disease caused by Corynebacterium pseudotuberculosis affecting small ruminants with small significance to human health because of its minor zoonotic potential. In both cases, few treatment options are available and conventional antimicrobial therapy is commonly refractory due to development of pyogranulomatous reactions, bringing great interest in discovering novel therapeutics for more suitable approaches. Dideoxynucleotides presented antibacterial action against various bacteria but were never described for C. pseudotuberculosis. Hypothesizing the antimi…

Antimicrobial Resistance and Molecular Epidemiology of Corynebacterium striatum Isolated in a Tertiary Hospital in Turkey.

Although Corynebacterium striatum is part of the human flora, it has recently drawn attention both for its multidrug resistance and its role as an invasive infection/outbreak agent. This cross-sectional study aimed to determine the antimicrobial resistance and clonal relationships among C. striatum strains. In total, 81 C. striatum strains were identified using Phoenix-100TM (BD, Sparks, MD, USA). The antimicrobial resistance of the strains was determined using the Kirby-Bauer disk diffusion method. Clonal relatedness among the strains was performed via arbitrarily primed polymerase chain reac…
