Bypassing Glutamic Acid Decarboxylase 1 (Gad1) Induced Craniofacial Defects with a Photoactivatable Translation Blocker Morpholino.

γ-Amino butyric acid (GABA) mediated signaling is critical in the central and enteric nervous systems, pancreas, lungs, and other tissues. It is associated with many neurological disorders and craniofacial development. Glutamic acid decarboxylase (GAD) synthesizes GABA from glutamate, and knockdown of the gad1 gene results in craniofacial defects that are lethal in zebrafish. To bypass this and enable observation of the neurological defects resulting from knocking down gad1 expression, a photoactivatable morpholino oligonucleotide (MO) against gad1 was prepared by cyclization with a photocleav

Congenital anomalies of the hand: an overview.

Of the 1% to 2% of newborns that are born with congenital defects, 10% of these are born with upper extremity malformations. Although many classification systems have been developed, the most widely used classification system was developed by Swanson. This system categorizes the congenital upper extremity malformations according to the embryonic process that has failed to develop. Congenital hand malformation is a broad category that is broken down into 7 subclasses. This review will focus on congenital hand defects and their associated craniofacial syndromes. The topics will include failure o

Craniofacial surgery for craniometaphyseal dysplasia.

Craniometaphyseal dysplasia (CMD) is a rare congenital bone dysplasia with abnormal bony overgrowth leading to characteristic facial features and cranial nerve compression. We present a 10-year-old child with bony swelling at the nasal root since birth along with decreased hearing in both ears. She had normal developmental milestones and intelligence. On examination, she had bossing of forehead with very broad nasal root, short septum, hypertelorism and epicanthic folds. CT scan with 3D reconstruction revealed grossly thickened calvarium and hyperostosis and sclerosis of the cranial base. As t

[Syndromes 4. Sanfilippo syndrome].

The Sanfilippo syndrome belongs to the mucopolysaccharidoses. In this syndrome heparansulfate accumulates in the lysosomes because of an enzymatic disorder in the degradation of mucopolysaccharides. The result is a function disorder of the involved tissues. The Sanfilippo syndrome is characterized by a combination of mental deterioration from the third year of life, hepatosplenomegaly and a typical facial appearance. These characteristics and the implications for dental treatment are discussed.

The effects of phenytoin on rat development: an animal model system for fetal hydantoin syndrome.

An animal model system has been established which reproduces some of the features of the Fetal Hydantoin Syndrome. This pattern of altered growth and development includes growth retardation, craniofacial anomalies, distal phalangeal hypoplasia, and mental deficiency. Rats exposed in utero to phenytoin on gestational days 9, 11, and 13 exhibited fetal onset growth retardation, abnormalities of the craniofacial region and axial skeleton. In addition, the exposed offspring had significantly lower fetal weights, a shortened snout and a high-arched, irregular palate, and significant delays in skele

The oculo-auriculo-fronto-nasal syndrome (OAFNS) - description of a rare and complex craniofacial deformity and its interdisciplinary management before school age.

Craniofacial clefts are certainly among the most challenging congenital malformations with respect to functional, aesthetic and psychosocial … ular malformations the term oculo-auriculo-fronto-nasal syndrome (OAFNS) has been proposed. Extreme craniofacial abnormalities have to be expected in this rare disorder. Adequate correction is a surgical challenge and interventions have to be adapted individually to patient's needs with respect to general condition, age and growth. This case report describes both the underl …

Benzophenone-2 disrupts craniofacial cartilage development via esr2a-Dependent inhibition of endothelin signaling in zebrafish larvae.

Epidemiological evidence has linked early-life exposure to estrogenic endocrine-disrupting chemicals (e-EDCs) to craniofacial malformations. Benzophenone-2 (BP2), a representative e-EDC, is frequently detected in environmental media and biological samples. However, the developmental effects of BP2 on craniofacial chondrogenesis and the underlying mechanisms remain unclear. Here, we demonstrate that early exposure to environmentally relevant concentrations of BP2 induces orofacial cleft-like craniofacial defects in zebrafish larvae, characterized by jaw flattening and anterior neurocranium shor

Craniocervical anomalies in Dubowitz syndrome. Three cases and a literature review.

Dubowitz syndrome is a disorder involving craniofacial abnormalities, growth retardation and mental retardation. Approximately 142 cases have been reported, with various associated other anomalies. These include cardiovascular, urogenital and endocrine abnormalities, as well as a predisposition to infections and hematological malignancies. Scoliosis has been described in association with this syndrome, as have isolated vertebral abnormalities. There has, however, been no description of craniocervical abnormalities. We describe three Dubowitz patients with significant craniocervical abnormaliti…

Tessier number 7 cleft with oblique clefts of bilateral soft palates and rare symmetric structure of zygomatic arch.

Craniofacial clefts are very rare congenital deformities. Tessier's numbering classification is a simple system universally accepted. Excluding cleft lip and palate, the transverse or lateral cleft (Tessier number 7) is the most common type of craniofacial cleft, manifesting macrostomia without skeletal abnormalities. We present a case of Tessier number 7 cleft with oblique clefting of the soft palate bilaterally and rare symmetric structures of the zygomatic arch.
