Chagas disease cardiomyopathy (CCC) is one of the few examples of post-infectious autoimmunity, where infectious episodes with an established pathogen, the protozoan parasite Trypanosoma cruzi, clearly triggers molecular mimicry-related target organ immune damage. CD4+ T-cell clones infiltrating hearts from CCC patients cross-reactively recognize human cardiac myosin, the major heart protein, and the immunodominant B13 protein from T. cruzi. Moreover, in vitro priming with B13 leads to the recovery of cardiac myosin cross-reactive T-cell clones. In order to identify cross-reactive epitopes bet

Platelets are short-lived anucleate cells essential for primary hemostasis and recognized for their functions in thrombosis, immunity, antimicrobial defense, neurodegeneration, as well as cancer growth and metastasis. Their brief lifespan in circulation is controlled by the removal of sialic acid residues from the platelet surface (desialylation) and also the mitochondrial apoptosis pathway, with high expression of the anti-apoptotic protein BCL-XL being required for platelet survival. This dependence on BCL-XL has prevented the clinical deployment of recently developed small molecule inhibito

OBJECTIVES: The characteristics of patients with panic disorder in emergency department (ED) patient populations are unknown. This study compares demographic information and emergency care use among patients identified as having a high likelihood of having panic disorder with that of patients who tested negative on the screening test for panic disorder.
METHODS: Prospective cross-sectional study of a convenience sample of patients presenting to an urban ED. Patients were excluded if they were aged 18 years or younger, were unstable, or could not speak English or Spanish. Of 968 patients, 813

Current influenza vaccines predominantly induce immunity to the hypervariable hemagglutinin (HA) head, requiring frequent vaccine reformulation. Conversely, the immunosubdominant yet conserved HA stem harbors a supersite that is targeted by broadly neutralizing antibodies (bnAbs), representing a prime target for universal vaccines. Here, we showed that the co-immunization of two HA stem immunogens derived from group 1 and 2 influenza A viruses elicits cross-group protective immunity and neutralizing antibody responses in mice, ferrets, and nonhuman primates (NHPs). Immunized mice were protecte

In the recent past, practical blood flow restriction (pBFR) using non-pneumatic, usually elastic cuffs has been established as a cost-effective alternative to traditional blood flow restriction (BFR) using pneumatic cuffs, especially for training in large groups. This study investigated whether low-load resistance exercise with perceptually primed pBFR using an elastic knee wrap is suitable to induce similar motor performance fatigue as well as physiological and perceptual responses compared to traditional BFR using a pneumatic nylon cuff in males and females. In a randomized, counterbalanced

The potential of maternally derived cellular factors to mediate immunity to Trichinella spiralis in neonates during lactation was investigated in this study. Female FI rats, infected with T. spiralis, were able to transfer immunity to their suckling offspring, evidenced by a significant reduction in the intestinal parasite burdens of their neonates. When challenged between 2 and 3 weeks of age with 200 T. spiralis larvae, pups suckling on immune mothers harboured 28% and 26% (at 3 and 8 days post-challenge) of the worm numbers present in control neonates suckling on naive mothers. Immune imprinting, in which prior antigenic exposures biases recall toward dominant epitopes, constrains the breadth and durability of influenza vaccine protection. Here we show that targeted variation across multiple hemagglutinin (HA) head sites (A, B, and D) between sequential A(H3N2) vaccines reprograms epitope hierarchy-redirecting recall toward conserved, subdominant head and stem epitopes. In a controlled ferret model mimicking imprinting-like recall in humans, antigenically distant priming accelerates neutralizing antibody induction, broadens reactivity, enhances cross-protection, and

Psoriatic disease initially affects the skin and later extends to the joints. Here, we show a two-step process that orchestrates the spread of inflammation from the skin to the joints. Induction of psoriatic skin disease in photoconvertible mice, followed by sequencing and computational characterization of skin-derived cells in the joints, was used to identify a population of CD2+MHC-II+CCR2+ myeloid precursors that builds a skin-derived myeloid cell compartment in the joints. Single-cell cross-species reference mapping and mitochondrial variant tracing showed an orthologous human cell populat

Introduction: CD40 signaling activates dendritic cells leading to improved T cell priming against tumor antigens. CD40 agonism expands the tumor-specific T cell repertoire and has the potential to increase the fraction of patients that respond to established immunotherapies.Areas covered: This article reviews current as well as emerging CD40 agonist therapies with a focus on antibody-based therapies, including next generation bispecific CD40 agonists. The scientific rationale for different design criteria, binding epitopes, and formats are discussed.Expert opinion: The ability of CD40 agonists

According to recent analyses, Bumby's RAPE scale of rape-supportive cognitions about women and sexual assault comprises two factors. Excusing rape serves to reduce abusers' culpability for their offending, and ascribing blame to victims, while justifying rape is associated with a sense of sexual entitlement. The distinct effects of these factors on rape judgments have not yet been investigated. We examined whether these belief clusters differentially explained judgments of perpetrator innocence after priming cues related to each of them. We used a cross-sectional design (N = 217) to test our h

Cytoplasmic antineutrophil cytoplasmic antibodies (cANCA) that accompany the neutrophilic vasculitis seen in Wegener's granulomatosis (WG), are directed against proteinase-3 (PR-3), a serine proteinase which is located in azurophilic granules of neutrophils and monocytes. PR-3, when expressed on the surface of TNFalpha-primed neutrophils, can directly activate neutrophils by complexing cANCA and promoting concomitant Fcgamma receptor (FcgammaR) cross-linking. Although the neutrophil's pathogenic role in WG has been studied, the role of the monocyte has not been explored. The monocyte, with its

The eye is increasingly recognized as a tissue shaped by systemic immune and metabolic states rather than an isolated immune-privileged organ. Translocator protein (TSPO), an 18-kDa outer mitochondrial membrane protein enriched in myeloid and glial cells, links mitochondrial homeostasis, redox signaling, lipid metabolism, and immune polarization. This review synthesizes evidence for TSPO involvement in systemic-ocular immunometabolic crosstalk and organizes it into a three-stage working model: peripheral immunometabolic priming, blood-retinal barrier vulnerability, and intraocular glial amplif

Exposure to microbes is essential to promote the development of the host's immune system. Commensal microbes (i.e. the microbiota) which are acquired early in life play a vital role in immune priming. Whilst many organisms within the microbiota are harmless, some can be considered opportunistic pathogens. Examples include Staphylococcus aureus, Streptococcus pneumoniae and Pseudomonas aeruginosa, and these organisms can also contribute to the development of a healthy host immune system. At the extreme end of the spectrum, pathogens which typically do not form part of the microbiota (e.g. Mycob

In a study of production of the common enterobacterial antigen (CA) by members of the Aeromonas group, ten strains of A. shigelloides, nine strains of A. hydrophila, and nine strains of A. salmonicida were used. Passive hemagglutination and hemolysis tests as well as the hemagglutination-inhibition procedure revealed that all strains of A. shigelloides, in contrast to the strains of the other two species, produce this antigenic determinant. The antigen of A. shigelloides was demonstrated even when the supernatant fluids of agar-grown cultures were used in a dilution of 1:1,000, whereas 10-time

The failure of M. bovis BCG to induce long-term protection has been endowed to its inability to escape the phagolysosome, leading to mild activation of CD8+ mediated T cell response. Induction of apoptosis in host cells plays an important role in potentiating dendritic cells-mediated priming of CD8+ T cells, a process defined as "cross-priming." Moreover, IL-10 secretion by infected cells has been reported to hamper BCG-induced immunity against Tuberculosis (TB). Previously, we have reported that apoptosis of BCG-infected macrophages and inhibition of IL-10 secretion is FOXO3 dependent, a tran…

Regulation of insulin-mediated resumption of meiotic maturation in catfish oocytes was investigated. Insulin stimulation of post-vitellogenic oocytes promotes the synthesis of cyclin B, histone H1 kinase activation and a germinal vesicle breakdown (GVBD) response in a dose-dependent and duration-dependent manner. The PI3K inhibitor wortmannin abrogates recombinant human (rh)-insulin action on histone H1 kinase activation and meiotic G2-M1 transition in denuded and follicle-enclosed oocytes in vitro. While the translational inhibitor cycloheximide attenuates rh-insulin action, priming with tran

BACKGROUND: To compare the efficacy of the Newcastle infant dialysis and ultrafiltration system (Nidus) with peritoneal dialysis (PD) and conventional haemodialysis (HD) in infants weighing <8 kg.
METHODS: We compared the urea, creatinine and phosphate clearances, the ultrafiltration precision, and the safety of the Nidus machine with PD in 7 piglets weighing 1-8 kg, in a planned randomised cross-over trial in babies, and in babies for whom no other therapy existed, some of whom later graduated to conventional HD.
RESULTS: Two babies entered the randomised trial; 1 recovered rapidly on PD, t

This study was done to examine the type and severity of disciplinary practices adopted by parents in Kerala. 1668 mothers, aged 18-49, participated in face-to-face interviews as part of a cross-sectional, population-based survey covering rural, urban and coastal areas. One child from each family was randomly selected as the index child. The interview focused primarily on disciplinary practices. 62% of the mothers reported using severe verbal discipline and 50% of mothers reported using severe physical abuse. Results suggest a high prevalence of normative and abusive practices in the community
