Signal transducer and activator of transcription-6 (STAT6) inhibition suppresses renal cyst growth in polycystic kidney disease.

Autosomal-dominant (AD) polycystic kidney disease (PKD) is a leading cause of renal failure in the United States, and currently lacks available treatment options to slow disease progression. Mutations in the gene coding for polycystin-1 (PC1) underlie the majority of cases but the function of PC1 has remained poorly understood. We have previously shown that PC1 regulates the transcriptional activity of signal transducer and activator of transcription-6 (STAT6). Here we show that STAT6 is aberrantly activated in cyst-lining cells in PKD mouse models. Activation of the STAT6 pathway leads to a p

Reversal of regioselectivity and enhancement of rates of nitrile oxide cycloadditions through transient attachment of dipolarophiles to cyclodextrins.

The reactions of nitrile oxides with monosubstituted dipolarophiles, such as propiolamide, typically afford proportionally 80 % or more of the 3,5-disubstituted cycloadducts. By contrast, the reactions of 6(A)-deoxy-6(A)-propynamido-beta-cyclodextrin with 4-tert-butylbenzonitrile oxide and 4-phenylbenzonitrile oxide afford >90 % and approximately 85 % of the corresponding 3,4-disubstituted isoxazoles, respectively. As well as reversing the regioselectivity, the cyclodextrin increases the rates of these cycloadditions. The extent of the acceleration is up to more than three orders of magnitude

The Sensory Identity of Caatinga Honeys: From Pollen Profile to Consumer Response.

Honey presents distinct sensory characteristics influenced by geographical and botanical origin. This study aimed to characterize the sensory identity of Apis mellifera honeys produced in the Caatinga region of Paraíba (Brazil) and to investigate their relationship with consumer perception. Twelve honey samples (mono- and polyfloral) were evaluated using a combination of Focus Group, Check-All-That-Apply (CATA), and affective tests. Initially, 551 sensory terms were generated through Focus Group sessions and reduced, through semantic and frequency analyses, to a structured sensory lexicon used

An ethnobotanical review of medicinal plants used for treating hemorrhoids in Thailand.

Hemorrhoids is one of the most prevalent diseases affecting people's health both in Thailand and worldwide. The therapeutic use of medicinal plants for hemorrhoidal treatments is widespread across many countries, supported by evidence demonstrating the efficacy of their pharmacological activity. This study aims to (1) conduct an in-depth literature review on plants used in Thailand for hemorrhoid treatment and (2) identify the most promising species in traditional Thai medicine to effectively treat hemorrhoids. A total of 53 references were methodically reviewed. The important medicinal plants

Possible therapeutic effects of essential oils on voltage dependent Na+ channels in experimental diabetic neuropathy.

Diabetic neuropathy (DN) is the most frequent complication of diabetes mellitus (DM), characterized by derangements of somatic and autonomic nerves, predominantly the longest nerves. DN is clinically relevant because it may seriously damage the quality of life of the patient, may be life-threatening, and is of great epidemiological incidence. It causes severe pain, dysesthesia, and allodynia initially; with disease progression, it kills the nerve, leading to full anaesthesia, accidental feet, leg ulceration, diabetic foot, and amputation. Underlying these dysfunctional alterations, there are n

Phytochemistry and Evaluation of the Anti-Inflammatory Activity of the Hydroethanolic Extract of Virola elongata (Benth.) Warb. Stem Bark.

BACKGROUND: Previous studies of the hydroethanolic extract of Virola elongata inner stem bark (HEVe) have demonstrated its antioxidant, gastroprotective, and antiulcer properties, but have not evaluated its anti-inflammatory potential. METHODS: HEVe was obtained by maceration and phytochemically analyzed. Its systemic anti-inflammatory activity was assessed by its effect on lipopolysaccharide (LPS)-induced peritonitis in mice. HEVe gel (HEgVe) was employed to evaluate topical anti-inflammatory activity by measuring the ear edema resulting from croton-oil-induced dermatitis in mice. A cell via

The Recycling of Substandard Rocket Fuel N,N-Dimethylhydrazine via the Involvement of Its Hydrazones Derived from Glyoxal, Acrolein, Metacrolein, Crotonaldehyde, and Formaldehyde in Organic Synthesis.

"Heptil" (unsymmetrical dimethylhydrazine-UDMH) is extensively employed worldwide as a propellant for rocket engines. However, UDMH constantly loses its properties as a result of its continuous and uncontrolled absorption of moisture, which cannot be rectified. This situation threatens its long-term usability. UDMH is an exceedingly toxic compound (Hazard Class 1), which complicates its transportation and disposal. Incineration is currently the only method used for its disposal, but this process generates oxidation by-products that are even more toxic than the original UDMH. A more benign appr

Gas-phase reactivity of acyclic α,β-unsaturated carbonyls towards ozone.

We evaluated different approaches to discuss the reactivity of α,β-unsaturated carbonyls comparative to alkene analogues. It was found that the reactivity factors xr, defined as the relative ratio between the rate coefficient of the carbonyl and a core structure, allow a semi-quantitative estimation of substituent effects in α,β-unsaturated acids, aldehydes and esters when the carbonyl containing substituent is replaced by a hydrogen atom. By contrast, it can be shown that the reactivity of the corresponding ketones differs from the other carbonyls. A linear correlation is presented between th

LASSBio-1586, an N-acylhydrazone derivative, attenuates nociceptive behavior and the inflammatory response in mice.

Pain and inflammation are complex clinical conditions that are present in a wide variety of disorders. Most drugs used to treat pain and inflammation have potential side effects, which makes it necessary to search for new sources of bioactive molecules. In this paper, we describe the ability of LASSBio-1586, an N-acylhydrazone derivative, to attenuate nociceptive behavior and the inflammatory response in mice. Antinociceptive activity was evaluated through acetic acid-induced writhing and formalin-induced nociception tests. In these experimental models, LASSBio-1586 significantly (p<0.05) redu

Basophils regulate the recruitment of eosinophils in a murine model of irritant contact dermatitis.

BACKGROUND: Although eosinophils have been detected in several human skin diseases in the vicinity of basophils, how eosinophils infiltrate the skin and the role of eosinophils in the development of skin inflammation have yet to be examined. OBJECTIVE: Using murine irritant contact dermatitis (ICD) as a model, we sought to clarify the roles of eosinophils in ICD and the underlying mechanism of eosinophil infiltration of the skin. METHODS: We induced croton oil-induced ICD in eosinophil-deficient ΔdblGATA mice with or without a reactive oxygen species (ROS) inhibitor. We performed cocultivati
