Plant hormone signaling and modulation of DNA repair under stressful conditions.

The role played by phytohormone signaling in the modulation of DNA repair gene and the resulting effects on plant adaptation to genotoxic stress are poorly investigated. Information has been gathered using the Arabidopsis ABA (abscisic acid) overly sensitive mutant abo4-1, defective in the DNA polymerase ε function that is required for DNA repair and recombination. Similarly, phytohormone-mediated regulation of the Ku genes, encoding the Ku heterodimer protein involved in DNA repair, cell cycle control and telomere homeostasis has been demonstrated, highlighting a scenario in which hormones mi

Fragrance material review on 3-methyl-5-(2,2,3-trimethyl-3-cyclopenten-1-yl)pent-3-en-2-one.

A toxicologic and dermatologic review of 3-methyl-5-(2,2,3-trimethyl-3-cyclopenten-1-yl)pent-3-en-2-one when used as a fragrance ingredient is presented. 3-Methyl-5-(2,2,3-trimethyl-3-cyclopenten-1-yl)pent-3-en-2-one is a member of the fragrance structural group Alkyl Cyclic Ketones. These fragrances can be described as being composed of an alkyl, R1, and various substituted and bicyclic saturated or unsaturated cyclic hydrocarbons, R2, in which one of the rings may include up to 12 carbons. Alternatively, R2 may be a carbon bridge of C2-C4 carbon chain length between the ketone and cyclic hyd

UVB radiation and 17-hydroxygeranyllinalool diterpene glycosides provide durable resistance against mirid (Tupiocoris notatus) attack in field-grown Nicotiana attenuata plants.

Depending on geographical location, plants are exposed to variable amounts of UVB radiation and herbivore attack. Because the role(s) of UVB in the priming and/or accumulation of plant defence metabolites against herbivores are not well understood, we used field-grown Nicotiana attenuata plants to explore the effects of UVB on herbivore performance. Consistent with previous reports, UVB-exposed plants accumulated higher levels of ultraviolet (UV)-absorbing compounds (rutin, chlorogenic acid, crypto-chlorogenic acid and dicaffeoylspermidine). Furthermore, UVB increased the accumulation of jasmo

Genetic variation in jasmonic acid- and spider mite-induced plant volatile emission of cucumber accessions and attraction of the predator Phytoseiulus persimilis.

Cucumber plants (Cucumis sativus L.) respond to spider-mite (Tetranychus urticae) damage with the release of specific volatiles that are exploited by predatory mites, the natural enemies of the spider mites, to locate their prey. The production of volatiles also can be induced by exposing plants to the plant hormone jasmonic acid. We analyzed volatile emissions from 15 cucumber accessions upon herbivory by spider mites and upon exposure to jasmonic acid using gas chromatography-mass spectrometry. Upon induction, cucumber plants emitted over 24 different compounds, and the blend of induced vola

Jasmonate- and salicylate-mediated plant defense responses to insect herbivores, pathogens and parasitic plants.

While extensive research has examined plant defense responses to attack by herbivores and pathogens, plant responses to parasitism by other plants are not well characterized. The expression of induced plant defenses is mediated by complex signaling networks in which the plant hormones jasmonic acid (JA) and salicylic acid (SA) play key roles. In general, JA-mediated signaling pathways are implicated in the regulation of antiherbivore defenses, while the SA pathway is associated with defense responses against pathogens. However, there are many exceptions to this basic framework, and recent work

Therapy against organophosphate poisoning: the importance of anticholinergic drugs with antiglutamatergic properties.

Potent cholinesterase inhibitors (e.g., soman, sarin), induce a wide range of deleterious effects including convulsions, behavioral impairments and ultimately, death. Due to the likelihood of various scenarios of military or terrorist attacks by these and other chemical weapons, research has to be aimed at finding optimal therapies. Early accumulation of acetylcholine in synaptic clefts was suggested to trigger an array of toxic events including an excessive release of glutamate, culminating in the activation of its receptors. Stimulation of the N-Methyl-D-Aspartate (NMDA) subtype of these rec

Parasitism by Cuscuta pentagona attenuates host plant defenses against insect herbivores.

Considerable research has examined plant responses to concurrent attack by herbivores and pathogens, but the effects of attack by parasitic plants, another important class of plant-feeding organisms, on plant defenses against other enemies has not been explored. We investigated how attack by the parasitic plant Cuscuta pentagona impacted tomato (Solanum lycopersicum) defenses against the chewing insect beet armyworm (Spodoptera exigua; BAW). In response to insect feeding, C. pentagona-infested (parasitized) tomato plants produced only one-third of the antiherbivore phytohormone jasmonic acid (

Arabidopsis jasmonate signaling pathway.

Jasmonates control defense gene expression and male fertility in the model plant Arabidopsis thaliana. In both cases, the involvement of the jasmonate pathway is complex, involving large-scale transcriptional reprogramming. Additionally, jasmonate signaling is hard-wired into the auxin, ethylene, and salicylate signal networks, all of which are under intense investigation in Arabidopsis. In male fertility, jasmonic acid (JA) is the essential signal intervening both at the level of anther elongation and in pollen dehiscense. A number of genes potentially involved in jasmonate-dependent anther e

Staphylococcus aureus pathogenicity on Arabidopsis thaliana is mediated either by a direct effect of salicylic acid on the pathogen or by SA-dependent, NPR1-independent host responses.

Staphylococcus aureus is a ubiquitous gram-positive bacterium that can cause superficial to serious systemic infections in animals and humans. Here we report the development of a plant infection model to study the pathogenesis of this bacterium. Three global regulatory mutants, RN6911 (agr-), ALC 488 (sarA-) ALC 842 (sarA-/agr-) and an alpha-toxin mutant defective in biofilm formation (DU1090) which are attenuated in animal pathogenesis, were also attenuated in their ability to infect plants, suggesting that these regulators that mediate synthesis of virulence factors essential for animal path

Ion channel-forming alamethicin is a potent elicitor of volatile biosynthesis and tendril coiling. Cross talk between jasmonate and salicylate signaling in lima bean.

Alamethicin (ALA), a voltage-gated, ion channel-forming peptide mixture from Trichoderma viride, is a potent elicitor of the biosynthesis of volatile compounds in lima bean (Phaseolus lunatus). Unlike elicitation with jasmonic acid or herbivore damage, the blend of substances emitted comprises only the two homoterpenes, 4,11-dimethylnona-1,3,7-triene and 4,8,12-trimethyltrideca-1,3,7,11-tetraene, and methyl salicylate. Inhibition of octadecanoid signaling by aristolochic acid and phenidone as well as mass spectrometric analysis of endogenous jasmonate demonstrate that ALA induces the biosynthe

Quantification of jasmonic acid, methyl jasmonate, and salicylic acid in plants by capillary liquid chromatography electrospray tandem mass spectrometry.

Jasmonic acid, methyl jasmonate, and salicylic acid have been reported to occur in plants and are thought to be essential for the regulation of systemic defense responses. This work describes a method for the quantitation in plant tissue of these regulators by reverse-phase capillary liquid chromatography interfaced to an electrospray tandem mass spectrometer. Inclusion during sample preparation of hydrogenated and/or deuterated internal standards corresponding to analogs of the regulators compensated for sample loss and permitted quantitation using the multiple reaction monitoring mode of the

Herbivore-induced volatiles: the emission of acyclic homoterpenes from leaves of Phaseolus lunatus and Zea mays can be triggered by a beta-glucosidase and jasmonic acid.

The treatment of healthy, undamaged plants of the Lima bean Phaseolus lunatus with solutions of a beta-glucosidase from bitter almonds (at 5 U.ml-1) through the petiole results in an enhanced emission of volatiles to the environment. The compounds are identical with those emitted in response to infestation with the red spotted spider mite Tetranychus urticae. Dominant products are the two acyclic homoterpenes 4,8-dimethyl-1,3E,7- dimethylnonatriene (homoterpene I) and 4,8,12-trimethyl-1,3E,7E,11-tridecatetraene (homoterpene II) which are of sesquiterpenoid and diterpenoid origin. Evaluation of dextromethorphan and carbetapentane as anticonvulsants and N-methyl-D-aspartic acid antagonists in mice.

Two antitussives, dextromethorphan and carbetapentane, which have been reported to bind to a common binding site in brain tissue and produce anticonvulsant effects in rats, were evaluated for their anticonvulsant effects against maximal electroshock-induced seizures, for their neurological impairing effects on the horizontal screen test, and their protective effects against N-methyl-D-aspartic acid (NMDA)-induced lethality in mice. Both compounds protected animals against maximal electroshock-induced seizures in a dose-related fashion after either intraperitoneal or oral administration. The ne

Pre-harvest potassium and postharvest cold storage extend vegetable soybean shelf life by inhibiting lipid oxidation and Jasmonate signaling.

Postharvest dehydration and senescence limit the shelf life of vegetable soybean. This study investigated the effects of pre-harvest potassium (K) fertilization and postharvest low-temperature storage on quality preservation, using physiological assays, widely targeted metabolomics, and transcriptomics in pods produced under K-deficient (K0) and K-sufficient (K120) conditions and stored at 4 °C or 25 °C. K120 seeds contained significantly higher K than K0 seeds (15.0 vs. 12.0 mg g-1 DW), confirming differential nutritional status. After 9 days of storage, low temperature combined with optimal

Jasmonic acid signaling in plants: regulatory mechanisms in development and stress responses.

Jasmonic acid (JA), one of the most pivotal plant hormones, has emerged as a central focus in plant biology research due to its extensive and diverse biological functions. This review synthesizes the significant advancements made over the past decade in understanding JA's role in regulating plant development and mediating responses to environmental stresses, areas that lacked systematic review in previous years. We provide a concise overview of JA's biosynthetic and signal transduction pathways, with particular emphasis on the key regulatory complex comprising CORONATINE INSENSITIVE 1 (COI1),

Hirschmanniella mucronata Effector HmENG1 Interacts with OsGAPDH to Promote Nematode Infection in Rice.

β-1,4-Endoglucanase (ENG) is a canonical effector protein secreted by the esophageal glands of plant-parasitic nematodes. Homology analysis identified a β-1,4-endoglucanase homolog in Hirschmanniella mucronata, designated as HmENG1. In situ hybridization demonstrated the specific localization of Hmeng1 transcripts within the esophageal glands of H. mucronata. Recombinant HmENG1 exhibited cellulase activity in vitro. In planta RNAi experiments confirmed that silencing Hmeng1 significantly reduced the infection capability of H. mucronata. In addition to its potential role in degrading plant cell

Blue or far-red light supplementation induced pre-hardening in the leaves of the Rht12 wheat dwarfing line: hormonal changes and freezing tolerance.

Reduced height (Rht) genes have revolutionised wheat cultivation, but they can compromise freezing tolerance, and only a few alleles are in use. Thus, evaluating the role of other Rht alleles in stress responses is crucial. Far-red supplementation of white light (W+FR) can induce pre-hardening in cereals at 15°C. However, the relevant effect of blue light enrichment (W+B) is poorly described. This study investigates the influence of W+FR or W+B exposure in young winter wheat leaves of a tall (wild-type, rht12) and a dwarf, gibberellin-deficient (near-isogenic line, Rht12) genotype in cv. In Vitro Inhibition of Cryptosporidium parvum Infection by the Olive Oil Component Oleocanthal.

Human cryptosporidiosis caused by the zoonotic apicomplexan parasite Cryptosporidium parvum represents a neglected and re-emerging poverty-related disease. C. parvum possesses minimalistic metabolic capacities and highly depends on its intestinal epithelial host cell for intracellular replication. Based on previous results showing that glycolysis and glutaminolysis inhibition diminished C. parvum replication in vitro, we here investigated the impact of the olive oil component oleocanthal on C. parvum infection in HCT-8 cells under physioxia (5% O2) and hyperoxia (21% O2). Oleocanthal targets a
