Adverse Effects Associated with Long-Term Administration of Azole Antifungal Agents.

Azole antifungals are first-line options in the prophylaxis and treatment of invasive fungal infections. They are often used for prolonged (weeks to months) periods of time, particularly in patients with hematologic malignancies, or in those who have received a solid organ or hematopoietic stem cell transplant. Long-term use of azoles is associated with hepatotoxicity and hormone-related effects, including gynecomastia, alopecia, decreased libido, oligospermia, azoospermia, impotence, hypokalemia, hyponatremia, and (rarely) adrenal insufficiency. Voriconazole and posaconazole have been associa

Fate, behaviour and weathering of priority HNS in the marine environment: An online tool.

Literature data and data obtained with modelling tools were compiled to derive the physicochemical behaviour of 24 priority Hazardous and Noxious Substances (HNS), as a proxy to improve environmental, public health and political issues in relation to HNS spills. Parameters that rule the HNS behaviour in water and those that determine their distribution and persistence in the environment, such as fugacity, physicochemical degradation, biodegradation, bioaccumulation/biotransformation and aquatic toxicity, were selected. Data systematized and produced in the frame of the Arcopol Platform project

Testing a multimedia compartmental model with monitoring data.

Based on its geographic similarity and nested structure, a chemical transport and transformation model developed in The Netherlands was adopted to a nine-parish, 5,400-km2 area in southern Louisiana, USA, and tested for its ability to predict concentrations in the environment. SimpleBox 2.0 represents a class of models that compartmentalize the air, water, soil, sediment, and plants into boxes while maintaining a high degree of detail for processes within and between boxes. Past use has been in the evaluative mode, primarily where qualitative predictions of chemical behavior and distribution a

Examination of low-incidence brain tumor responses in F344 rats following chemical exposures in National Toxicology Program carcinogenicity studies.

Neoplasms in the brain are uncommon in control Fischer 344 (F344) rats; they occur at a rate of less than 1% in 2-yr toxicity/carcinogenicity studies. Furthermore, only 10 of nearly 500 studies conducted by the National Toxicology Program (NTP) showed any evidence of chemically related neoplastic effects in the brain. Generally, the brain tumor responses were considered equivocal, because the characteristics of potential neurocarcinogenic agents (such as statistically significant increased incidences, decreased latency and/or survival, and demonstration of dose-response relationships) were not

QSAR-Driven Discovery of Novel Chemical Scaffolds Active against Schistosoma mansoni.

Schistosomiasis is a neglected tropical disease that affects millions of people worldwide. Thioredoxin glutathione reductase of Schistosoma mansoni (SmTGR) is a validated drug target that plays a crucial role in the redox homeostasis of the parasite. We report the discovery of new chemical scaffolds against S. mansoni using a combi-QSAR approach followed by virtual screening of a commercial database and confirmation of top ranking compounds by in vitro experimental evaluation with automated imaging of schistosomula and adult worms. We constructed 2D and 3D quantitative structure-activity relat

Using membrane-water partition coefficients in a critical membrane burden approach to aid the identification of neutral and ionizable chemicals that induce acute toxicity below narcosis levels.

The risk assessment of thousands of chemicals used in our society benefits from adequate grouping of chemicals based on the mode and mechanism of toxic action (MoA). We measure the phospholipid membrane-water distribution ratio (DMLW) using a chromatographic assay (IAM-HPLC) for 121 neutral and ionized organic chemicals and screen other methods to derive DMLW. We use IAM-HPLC based DMLW as a chemical property to distinguish between baseline narcosis and specific MoA, for reported acute toxicity endpoints on two separate sets of chemicals. The first set comprised 94 chemicals of US EPA's acute

MS2Tox Machine Learning Tool for Predicting the Ecotoxicity of Unidentified Chemicals in Water by Nontarget LC-HRMS.

To achieve water quality objectives of the zero pollution action plan in Europe, rapid methods are needed to identify the presence of toxic substances in complex water samples. However, only a small fraction of chemicals detected with nontarget high-resolution mass spectrometry can be identified, and fewer have ecotoxicological data available. We hypothesized that ecotoxicological data could be predicted for unknown molecular features in data-rich high-resolution mass spectrometry (HRMS) spectra, thereby circumventing time-consuming steps of molecular identification and rapidly flagging molecu

Predicting skin sensitizer potency based on in vitro data from KeratinoSens and kinetic peptide binding: global versus domain-based assessment.

Three in vitro methods for the prediction of the skin sensitization hazard have been validated. However, predicting sensitizer potency is a key requirement for risk assessment. Here, we report a database of 312 chemicals tested in the KeratinoSens™ assay and for kinetic peptide binding. These data were used in multiple regression analysis against potency in the local lymph node assay (LLNA). The dataset covers the majority of chemicals from the validation of the LLNA to predict human potency and this subset was analyzed for prediction of human sensitization potency by in vitro data. Global ana

OncoSolidDB: An Oncology-Focused Curated Database of Ligand-Target Interactions for Precision Medicine Across Major Solid Cancers.

Background/Objectives: The rapid expansion of targeted therapies has reshaped oncology by exploiting ligand-receptor interactions (LRI) to improve treatment specificity and patient outcomes. However, the data describing these ligands remain fragmented across multiple sources, limiting accessibility for researchers and clinicians. To address this gap, we developed the OncoSolidDB, the first curated and oncology-focused bioinformatics database dedicated to ligands associated with solid malignancies. Methods: OncoSolidDB integrates and harmonizes data from reliable repositories, including ChEMBL,

Integration of a virtual database-based precursor ion list data acquisition strategy combined with untargeted metabolomics for characterizing and distinguishing triterpenoids and flavonoids in Astragalus membranaceus var. mongholicus and Astragalus membranaceus.

Systematic characterization of natural product components and identification of chemical markers are important prerequisites for quality control and product development. Although data dependent acquisition (DDA) methods are widely used for the identification of natural product components, they have the limitation of potentially missing key active ingredients present at low abundance. This study proposes a comprehensive analytical strategy that integrates a virtual database-based precursor ion list (PIL) acquisition method with untargeted metabolomics to deeply characterize triterpenoids and fl

Targeting multiple pathways with virtual screening to identify the multi-target agent for Alzheimer's disease treatment.

Alzheimer's disease (AD) is a chronic neurodegenerative disorder characterized by progressive memory loss, posing a significant risk, particularly within aging populations. Effective treatments to prevent or cure AD have remained elusive with current treatments only able to slow disease progression. Moreover, challenges in drug development for AD therapies are a complex pathology. A multi-target agent gained considerable interest over the single therapy in complex diseases, providing possible insights into therapeutic development by simultaneously targeting multiple pathological mechanisms. Scoring multiple toxicological endpoints using a toxicogenomic database.

As information regarding microarray data sets and toxicogenomic biomarkers grows rapidly, the process of analyzing data and interpreting the results is increasingly complicated. To facilitate data analysis, a simple expression ratio-based scoring method called the TGP1 score was previously proposed [Kiyosawa, N., Shiwaku, K., Hirode, M., Omura, K., Uehara, T., Shimizu, T., Mizukawa, Y., Miyagishima, T., Ono, A., Nagao, T., Urushidani, T., 2006. The CXCL Family Contributes to Immunosuppressive Microenvironment in Gliomas and Assists in Gliomas Chemotherapy.

Gliomas are a type of malignant central nervous system tumor with poor prognosis. Molecular biomarkers of gliomas can predict glioma patient's clinical outcome, but their limitations are also emerging. C-X-C motif chemokine ligand family plays a critical role in shaping tumor immune landscape and modulating tumor progression, but its role in gliomas is elusive. In this work, samples of TCGA were treated as the training cohort, and as for validation cohort, two CGGA datasets, four datasets from GEO database, and our own clinical samples were enrolled. Consensus clustering analysis was first int

Improving dictionary-based named entity recognition with deep learning.

MOTIVATION: Dictionary-based named entity recognition (NER) allows terms to be detected in a corpus and normalized to biomedical databases and ontologies. However, adaptation to different entity types requires new high-quality dictionaries and associated lists of blocked names for each type. The latter are so far created by identifying cases that cause many false positives through manual inspection of individual names, a process that scales poorly. RESULTS: In this work, we aim to improve block list s by automatically identifying names to block, based on the context in which they appear. By c

Access to Interpretable Data to Support Disproportionate Health Risks from Industrial Releases: A Case Study on the Environmental Protection Agency's Datasets and Their Application to the Latinx Communities of Houston, Texas.

Latinx communities face disproportionate environmental injustices and are targeted due to systematic economic and political inequities. This research evaluates the ease at which links between industrial releases and risk of adverse health effects can be defined to influence policy change in Houston, TX. The Environmental Protection Agency (EPA)'s Toxic Release Inventory (TRI) is the most comprehensive public database on industrial facilities' toxic chemical releases in the US. TRI is presented within a risk-based context through the Risk Screening Environmental Indicators (RSEI) scores. TRI an

Assessing the effects of interventions for Aedes aegypti control: systematic review and meta-analysis of cluster randomised controlled trials.

BACKGROUND: The Aedes aegypti mosquito is the vector for dengue fever, yellow fever, chikungunya, and zika viruses. Inadequate vector control has contributed to persistence and increase of these diseases. This review assesses the evidence of effectiveness of different control measures in reducing Aedes aegypti proliferation, using standard entomological indices. METHODS: A systematic search of Medline, Ovid, BVS, LILACS, ARTEMISA, IMBIOMED and MEDIGRAPHIC databases identified cluster randomised controlled trials (CRCTs) of interventions to control Aedes aegypti published between January 2003

Chiral pesticides: identification, description, and environmental implications.

Of the 1,693 pesticides considered in this review, 1,594 are organic chemicals, 47 are inorganic chemicals, 53 are of biological origin (largely non chemical; insect, fungus, bacteria, virus, etc.), and 2 have an undetermined structure. Considering that the EPA's Office of Pesticide Programs found 1,252 pesticide active ingredients(EPA Pesticides Customer Service 2011), we consider this dataset to be comprehensive; however, no direct comparison of the compound lists was undertaken. Of all pesticides reviewed, 482 (28%) are chiral; 30% are chiral when considering only the organic chemical pestic
