Adult baboons (5 males and 5 females) were exposed to immobilization stress by being strapped to a table in a horizontal position for 2 h. In females the experiment was performed during both the follicular and luteal phase. Breast microcysts are considered to be a normal findings in the adult female breast without any increased risk of developing carcinomatous change. Breast cysts fluid contains steroid but not studies have been reported on the ability of breast microcysts to metabolise steroid hormones. Twenty-three women with moderately elevated plasma testosterone levels (77 to 240 ng/dl) received 10 mg of medroxyprogesterone acetate (MPA) orally, three times daily, for 3 months or more. The testosterone levels were reduced to normal (less than 70 ng/dl) in 21 patients, suppressed partially in 1 patient, and unchanged in 1 patient. The removal of an ovarian teratoma normalized the testosterone level in the patient whose levels had failed to suppress with MPA. Androgen insensitivity was demonstrated in two male siblings with partial masculinization of the external genitalia. They had a previously undescribed defect characterized postpubertally by high plasma testosterone and luteinizing hormone concentrations with low serum follicle-stimulating hormone levels. Studies in skin fibroblasts showed normal androgen receptor affinity and capacity for 5alpha-dihydrotestosterone (DHT), normal nuclear retention of the receptor-DHT complex, and normal conversion of testosterone to DHT.

Three retinol dehydrogenases (RDHs) were tested for steroid converting abilities: human and murine RDH 12 and human RDH13. RDH12 is involved in retinal degeneration in Leber's congenital amaurosis (LCA). We show that murine Rdh12 and human RDH13 do not reveal activity towards the checked steroids, but that human type 12 RDH reduces dihydrotestosterone to androstanediol, and is thus also involved in steroid metabolism. Furthermore, we analyzed both expression and subcellular localization of these enzymes.

After amniotic fluid and fetal skin biopsy cultures trisomy 18 was diagnosed and the steroid metabolism of tissues from this foetus was studied by double isotope methods and compared with results from foetuses of similar age. The metabolism of 14C-pregnenolone, the aromatization of androstendendione to oestrone and the 5alpha-reduction of testosterone to 5alpha-dihydrotestosterone in this foetus were similar as in other foetuses. The reason for a low oestriol excretion in pregnancy with 18 trisomic foetus is not evident from the data obtained.

In vitro transformation of precursors to androgens and oestrogens was studied in breast tissues from 7 phenotypic males with gynaecomastia and 3 normal females. All the tissues studied showed formation of dihydrotestosterone and oestrogens, thus demonstrating the presence of 5alpha-reductase and aromatizing enzymes. The results on the values of urinary oestrogens before and after surgical removal of the breast tissue substantiate the biosynthetic potential of gynaecomastic tissue. The aetiological significance of these findings vis-à-vis gynaecomastia is discussed.

In experiment on rats injected with aromatase inhibitor or with dihydrotestosterone (DHT) it has been revealed that increase of androgens level in blood stimulates ovarial folliculogenesis on its early stages. DHT as compared with testosterone accelerates small follicles growth more effectively. Androgens, particularly DHT (7 mkg/100 g), suppress preovarial follicules maturing and turn them into atretic ones. Suppressing of ovarial functional activity delays or blockages the first ovulation during rats puberty.

A mother and daughter with lichen sclerosus et atrophicus (LSA) are described. Both had lesions restricted to their lower legs. Both patients were positive for HLA-B40, which has been associated with LSA. Sex steroid hormones showed no decrease in dihydrotestosterone, free testosterone, or androstenedione levels in the daughter as was demonstrated in a previous study of this disease, but did show some decrease in androstenedione in the mother. This article provides further evidence for a possible familial predisposition for LSA.

The androgen status in fifty consecutive women with hirsutism in the reproductive age group referred to the endocrine clinic was studied. Mean BMI was 24.3 +/- 5.6 kg/M2, 42% had menstrual disturbances and 30%, had severe hirsutism. The hirsute patients had significantly higher estimates of testosterone (48%), dihydrotestosterone (38%) and androstenedione (86%). Based on clinical and hormonal data, polycystic ovarian disease was diagnosed in 20 patients, nonclassical 21 hydroxylase deficiency in one and idiopathic hirsutism in 29.
