Biocompatible antibiotic-loaded mesoporous silica/bioglass/collagen-based scaffolds as bone drug delivery systems.

Local delivery of antibiotics has gained increasing interest in the treatment of osteomyelitis due to its effectiveness and safety. Since the regeneration of bone tissue at the site of infection is as important as bacterial eradication, implantable drug delivery systems should not only release the drugs in a proper manner but also exert the osseointegration capability. Herein, we present an implantable drug delivery system in a scaffold form with a unique set of features for local treatment of osteomyelitis. For the first time, collagen type I, ciprofloxacin-loaded mesoporous silica, and biogl…

Systematic review of studies on drug-delivery systems for management of temporomandibular-joint osteoarthritis.

INTRODUCTION: Temporomandibular-joint osteoarthritis (TMJOA) management is a major challenge. Minimally invasive therapies (based mainly on injections) have been developed to increase local efficacy and limit adverse systemic effects. However, the requirement for repeat injections due to a short duration of action and expensive healthcare costs have pushed researchers to develop, via tissue engineering, drug-delivery systems (DDSs). In this literature systematic review, we aim to provide an overview of studies that tested DDSs on a TMJOA model. MATERIAL AND METHODS: We searched on PubMed for

Surface functionalization of zeolite-based drug delivery systems enhances their antitumoral activity in vivo.

Zeolites have attractive features making them suitable carriers for drug delivery systems (DDS). As such, we loaded the anticancer drug 5-fluorouracil (5-FU), into two different zeolite structures, faujasite (NaY) and Linde Type L (LTL), to obtain different DDS. The prepared DDS were tested in vitro using breast cancer, colorectal carcinoma, and melanoma cell lines and in vivo using the chick embryo chorioallantoic membrane model (CAM). Both assays showed the best results for the Hs578T breast cancer cells, with a higher potentiation for 5-FU encapsulated in the zeolite LTL. To unveil the endo…

Implantable drug delivery systems with a bioinspired zwitterionic nanocoating resist foreign body reaction-induced obstruction and enable sustained delivery.

Implantable drug delivery systems (IDDS) hold great promise for sustained therapeutic administration, particularly for deep tissues like the inner ear. However, the obstruction of delivery systems induced by foreign body reactions (FBRs) remains a significant challenge to long-term implantation. Here, we developed a bio-inspired zwitterionic nanocoating (PDA-PSB) for IDDS. Experimental results showed that the PDA-PSB coating significantly improved the hydrophilicity, reduced protein and cell adhesion, and effectively suppressed inflammatory responses. To evaluate the long-term performance, we …

Sublingual Drug Delivery Systems: Quality by Design Principles, Applications, Market Landscape, and Innovative Technologies.

One promising alternative to the oral approach is drug administration via the oral mucosa. Sublingual refers to the administration of drugs through the mouth that are rapidly absorbed by the blood vessels "under the tongue." In terms of efficacy and patient compliance, the sublingual route is better than the oral route of delivery. Sublingual drug delivery systems provide a rapid onset of action, bypass first-pass liver extraction in the gastrointestinal route, and achieve high bioavailability through enzymatic degradation. This article highlights the quality-by-design philosophy, the everchan…
