Nicorandil Affects Mitochondrial Respiratory Chain Function by Increasing Complex III Activity and ROS Production in Skeletal Muscle Mitochondria.

Adenosine triphosphate (ATP)-dependent potassium channels openers (KATP) protect skeletal muscle against function impairment through the activation of the mitochondrial KATP channels (mitoKATP). Previous reports suggest that modulators of the mitochondrial KATP channels have additional effects on isolated mitochondria. To determine whether the KATP channel opener nicorandil has non-specific effects that explain its protective effect through the mitochondrial function, chicken muscle mitochondria were isolated, and respiration rate was determined pollarographically. The activity of the electron

The XylC-mediated aromatic metabolic network modulates the metabolic response of Pseudomonas aeruginosa to deltamethrin.

The widespread emergence of pyrethroid resistance represents a major challenge in pest and vector management. This study examined the function of the xylC-encoded benzaldehyde dehydrogenase in Pseudomonas aeruginosa under deltamethrin stress. The metabolic response of a ΔxylC mutant cultured with 50 mg/L deltamethrin was compared to the wild-type strain, and functional complementation was achieved by adding purified XylC protein. Integrated metabolomic and proteomic analyses were used to characterize the associated metabolic adaptations. Deltamethrin exposure inhibited the production of 3-(2-c

Morphological and molecular characterization of ixodid tick vectors in Algeria, North Africa.

Ticks are hematophagous ectoparasites of veterinary and medical importance, serving as vectors for a wide array of pathogens that affect livestock, wildlife, and humans. Given their role in pathogen dissemination, this study aimed to provide an updated molecular characterization and phylogenetic analysis of hard tick species in Algeria using two mitochondrial markers: Cytochrome c oxidase subunit 1 (Cox1) and the 12S rRNA gene. We analyzed fourteen tick species from the archival collection of the Pasteur Institute of Algeria, originally collected from various provinces across the country. Thes

Genetic characterization of Angiostrongylus cantonensis in Rattus norvegicus in Hokkaido, Japan: A rediscovery record in Hokkaido after 44 years.

Angiostrongylus cantonensis is a zoonotic nematode causing eosinophilic meningitis in humans. This species was confirmed to exist in Hokkaido until 1982, its current status remained unknown. In the present study, we detected A. cantonensis in brown rats (Rattus norvegicus) in Hokkaido for the first time in 44 years. We conducted the first genetic characterization of A. cantonensis in Hokkaido by performing a phylogenetic analysis using two genes. We identified A. cantonensis using morphological and genetic analysis, which confirmed its current presence in Sapporo City. The small subunit of the

H2S remodels mitochondrial ultrastructure and destabilizes respiratory supercomplexes.

Mitochondrial form and function are intimately interconnected, responding to cellular stresses and changes in energy demand. Hydrogen sulfide, a product of amino acid metabolism, has dual roles as an electron transport chain substrate and complex IV (CIV) inhibitor, leading to a reductive shift, which has pleiotropic metabolic consequences. Luminal sulfide concentration in the colon is high due to microbial activity, and in this study, we demonstrate that chronic sulfide exposure of colonocyte-derived cells leads to lower Mic60 and Mic19 expression that is correlated with a profound loss of cr

Lack of Musashi-2 induces type IIa fiber-dominated muscle atrophy.

Skeletal muscle is a highly plastic tissue, adapting its structure and metabolism in response to diverse conditions such as contractile activity, nutrients, and diseases. Finding a novel master regulator of muscle mass and quality will provide new therapeutic targets for the prevention and treatment of muscle weakness. Musashi is an RNA-binding protein that dynamically regulates protein expression; it was originally discovered as a cell fate determination factor in neural cells. Here, we report that Musashi-2 (Msi2) is dominantly expressed in slow-type muscle fibers, fibers characterized by hi

Pathological Features in Paediatric Patients with TK2 Deficiency.

Thymidine kinase (TK2) deficiency causes mitochondrial DNA depletion syndrome. We aimed to report the clinical, biochemical, genetic, histopathological, and ultrastructural features of a cohort of paediatric patients with TK2 deficiency. Mitochondrial DNA was isolated from muscle biopsies to assess depletions and deletions. The TK2 genes were sequenced using Sanger sequencing from genomic DNA. All muscle biopsies presented ragged red fibres (RRFs), and the prevalence was greater in younger ages, along with an increase in succinate dehydrogenase (SDH) activity and cytochrome c oxidase (COX)-neg

Time course of cytochrome oxidase blob plasticity in the primary visual cortex of adult monkeys after retinal laser lesions.

We studied the time course of changes of cytochrome oxidase (CytOx) blob spatial density and blob cross-sectional area of deprived (D) and nondeprived (ND) portions of V1 in four capuchin monkeys after massive and restricted retinal laser lesions. Laser shots at the border of the optic disc produced massive retinal lesions, while low power laser shots in the retina produced restricted retinal lesions. These massive and restricted retinal lesions were intended to simulate glaucoma and diabetic retinopathy, respectively. We used a Neodymium-YAG dual frequency laser to make the lesions. We measur
