Drug-induced suppression of ACTH secretion does not promote anti-depressive or anxiolytic effects.

Mammals respond to a real or perceived stress in an integrated physiological and psychological fashion. Psychiatric disorders like major depression and anxiety have been associated to stressful events. In a previous study we demonstrated that the stress-induced ACTH secretion can be robustly inhibited by the concurrent use of CRF1 (CP154,526 - Pfizer) and V1B (SSR149415 - Sanofi-Aventis) non-peptide antagonists. A proof of mechanism was offered by substituting CP154,526 by SSR125543 and obtaining the same results on three stress models: forced swimming, ether vapor inhalation and restraint. SS

Neuroprotective Effects of Alpha-Lipoic Acid Against Behavioral Toxicity, Oxidative and Inflammatory Damage Caused by Titanium Dioxide Nanoparticles.

Titanium dioxide nanoparticles (TiO₂-NPs) are used widely in various industries, but emerging evidence suggests their ability to elicit neurotoxicity. The present study evaluated the alpha-lipoic acid (ALA) neuroprotective potential against TiO₂-NP-induced cognitive and molecular impairments in male Sprague-Dawley rats. Twenty-four rats were allocated into four groups: negative control, TiO₂-NPs (150 mg/kg, i.p.), ALA (50 mg/kg, orally), and TiO₂-NPs + ALA. Treatments were administered on alternate days for 28 days. Neurobehavioral tests, including the open field test (OFT), elevated plus maze

Preventive and Therapeutic Effects of Astaxanthin on Depressive-Like Behaviors in High-Fat Diet and Streptozotocin-Treated Rats.

The comorbidity of diabetes and depression has a negative impact on both lifestyle and quality of life. Astaxanthin (AST) has been demonstrated to improve glucose metabolism and has antidepressant-like effects, but it is not clear whether AST has potential for preventing depression in diabetes. The aim of this study is to observe the preventive and therapeutic effects of AST on glucose metabolism or depressive-like behaviors in a diabetic rat model produced by feeding with a high-fat diet for 10 weeks followed by injection of 25 mg/kg streptozotocin (STZ). Preventive treatment with AST at dose

Potential effects of genistein in preventing progressive loss of cognitive functions in wild-type mice.

Mild cognitive impairment occurring with age is an intermediate stage between normal cognitive functioning and dementia. Precisely, because of the lack of unequivocal symptoms, the deterioration of cognitive functions that occurs with age is a growing concern for the aging population. Therefore, the aim of this study was to verify the prophylactic potential of genistein (5,7-dihydroxy-3-(4-hydroxyphenyl)-4H-1-benzopyran-4-one) in preventing memory loss of healthy individuals. The experiments were conducted using wild-type mice of both sexes that were not genetically modified. Analysis of cogni

Alcohol-Induced Early-Stage Liver Injury Contributes to Increases in Voluntary Alcohol Intake and Cognitive Deficits in Mice.

BACKGROUND: Liver-brain communication is increasingly recognized as a potential contributor to vulnerability to alcohol use disorder (AUD). However, the behavioral consequences of early alcohol-associated liver disease (ALD) remain underexplored. This study aimed to determine whether alcohol-induced hepatic injury influences subsequent alcohol consumption and cognitive outcomes. METHODS: Female C57BL/6J mice were fed a Lieber-DeCarli (LDC) ethanol diet to induce early-stage ALD-like pathology, followed by a voluntary two-bottle choice (TBC) paradigm measuring alcohol consumption. Behavioral a

Combined exposure to tobacco smoke and ethanol during adolescence leads to short- and long-term modulation of anxiety-like behavior.

BACKGROUND: Tobacco smoking is associated with alcohol drinking and consumption of both drugs typically begins during adolescence. Since anxiety is considered a relevant factor for both smoking and drinking due to its motivating force for a continued consumption, anxiety alterations shared by these two drugs could explain their co-use and co-abuse. METHODS: Here, we investigated the short- and long-term effects of adolescent tobacco smoke and/or ethanol exposure on anxiety levels. From postnatal day 30-45, Swiss mice were exposed to tobacco smoke (SMK--whole body exposure, 8 h/day) and/or eth

[Effect of a new triazinoindole derivative on the functional state of CNS in animals under normoxia and hypoxia conditions].

The influence of the new triazinoindole derivative encoded VM-606 on the individual behavior of rats in the open-field and elevated-plus-maze tests has been studied under normal conditions and after exposure to hypoxia with hypercapnia. It is established that VM-606 at a dose of 50 mg/kg under normoxia conditions reduces emotional anxiety, orientation-investigation activity, and mobility factor, while under hypoxic conditions this drug reduces the severity of behavioral changes in test animals. The experiments on mice showed that the compound studied potentiates the hypnotic effect of hexenal.

Dcf1 Affects Memory and Anxiety by Regulating NMDA and AMPA Receptors.

The hippocampus is critical for memory and emotion and both N-methyl-D-aspartate (NMDA) and α-amino-3-hydroxy-5-methyl- 4-isoxazolepropionic acid (AMPA) receptors are known to contribute for those processes. However, the underlying molecular mechanisms remain poorly understood. We have previously found that mice undergo memory decline upon dcf1 deletion through ES gene knockout. In the present study, a nervous system-specific dcf1 knockout (NKO) mouse was constructed, which was found to present severely damaged neuronal morphology. The damaged neurons caused structural abnormalities in dendrit

Standardised ginseng extract G115® potentiates the antidepressant-like properties of fluoxetine in the forced swim test.

OBJECTIVE: Ginsenosides, biologically active components of the root of Panax ginseng, have been reported to have therapeutic benefits in a number of disease states including psychiatric conditions such as major depressive disorder. Our objective was to determine if a standardised commercial ginseng extract, G115®, could reduce the signs of behavioural despair commonly observed in animal models of depression either alone or in combination with the selective serotonin reuptake inhibitor (SSRI) fluoxetine. METHODS: Male Sprague-Dawley (SD) rats (N = 51) were divided into four groups: vehicle con

Activation of Calcium/Calmodulin-Dependent Kinase II in the Medial Prefrontal Cortex Mediates Spinal Cord Injury-Related Cognitive and Affective Changes.

PURPOSE: Research has documented a high prevalence of cognitive and affective impairments in individuals with spinal cord injury (SCI). However, the molecular mechanisms underlying these deficits remain poorly understood. In this study, to investigate the molecular basis of cognitive and affective dysfunctions following SCI, we examined the role of calcium/calmodulin-dependent kinase II (CaMKII) activation, with a specific focus on its phosphorylated form (pCaMKII), using a rat model of SCI. METHOD: Experimental results demonstrated that SCI led to spatial memory deficits as well as depressio

Behavioral effects of flibanserin (BIMT 17).

Flibanserin is a 5-HT1A agonist that, in contrast to other 5-HT1A receptor agonists, is capable of activating 5-HT1A receptors in frontal cortex. Flibanserin also behaves as an antagonist at 5-HT2A receptors. This compound has been described to be a putative fast-acting antidepressant owing to these properties. In the present study, the effect of flibanserin was investigated in several behavioral paradigms different from animal models of depression. Intraperitoneal flibanserin, at doses of 4-8 mg/kg, antagonized d-amphetamine- and (+)SKF-10047- induced hypermotility in mice and rats. At doses

The anxiolytic effect of cinnabar involves changes of serotonin levels.

The aim of this study was to explore the neuropharmacological mechanism of cinnabar. The anxiolytic effect of cinnabar on anxiety-like behaviors in mice was investigated using the elevated plus maze test. The changes in the levels of monoamine neurotransmitters and their metabolites and the activity of monoamine oxidase (MAO) in the brain of mice were determined. The results indicate that cinnabar possessed anxiolytic effects after chronic administration (p.o.) at effective doses in association with the declined brain serotonin (5-HT) level. The cinnabar showed no effects on 5-HT metabolism pa…

Role of different neurotransmitter systems in the cholecystokinin octapeptide-induced anxiogenic response in rats.

The possible involvement of different neurotransmitter systems in the anxiogenic action of cholecystokinin octapeptide sulphate ester (CCK-8) was investigated in rats. Intracerebroventricularly (i.c.v.) administered CCK-8 induced an anxiogenic response in an elevated plus-maze test. Pretreatment with dopaminergic, muscarinergic acetylcholine receptor blockers and an opiate receptor antagonist blocked the anxiogenic response to CCK-8. The alpha and beta adrenoreceptor, the GABA receptor and the 5-hydroxytryptamine (5-HT) receptor blockers were not able to modulate the 'anxiogenic-like' effect o

Energizing effects of bupropion on effortful behaviors in mice under positive and negative test conditions: modulation of DARPP-32 phosphorylation patterns.

Motivational symptoms such as anergia, fatigue, and reduced exertion of effort are seen in depressed people. To model this, nucleus accumbens (Nacb) dopamine (DA) depletions are used to induce a low-effort bias in rodents tested on effort-based decision-making. We evaluated the effect of the catecholamine uptake blocker bupropion on its own, and after administration of tetrabenazine (TBZ), which blocks vesicular storage, depletes DA, and induces depressive symptoms in humans. Male CD1 mice were tested on a 3-choice-T-maze task that assessed preference between a reinforcer involving voluntary p

Overexpression of FoxO3a in the dentate gyrus alleviates CUS-induced anxiety- and depression-like behaviors and cognitive impairment.

BACKGROUND: Excessive stress leads to injury and dysfunction, but the underlying mechanism remains unclear. As a human longevity gene, forkhead box O3a (FoxO3a) is a transcription factor that regulates various cellular processes, including the response to oxidative stress, apoptosis, and autophagy. This study aims to explore whether FoxO3a in the dentate gyrus (DG) of the hippocampus is involved in the formation of anxiety- and depressive-like behavior and cognitive impairment in stressed rats and to investigate the detailed mechanism. METHODS: This study was conducted using the 6-week chroni

Female rats exposed to stress and alcohol show impaired memory and increased depressive-like behaviors.

Exposure to daily life stressors is associated with increases in anxiety, depression, and overall negative affect. Alcohol or other psychoactive drugs are often used to alleviate stress effects. While females are more than twice as likely to develop mood disorders and are more susceptible to dependency than males, they are infrequently examined. In this study, female rats received no stress/no alcohol control (CON), alcohol alone (ALC), stress alone (STR), or stress plus alcohol (STR+ALC). Stress consisted of restraint for 6h/day/7days, and alcohol was administered immediately following restra

Sickness behavior of rats with abdominal sepsis can be improved by antibiotic and G-CSF prophylaxis in clinic modeling randomized trials.

BACKGROUND AND AIM: In clinical sepsis research nearly all immune-modulators have demonstrated no benefit in regard to the 28-day mortality rate. Other endpoints such as quality of life have become more attractive, but clinically relevant animal models analyzing an equivalent to quality of life by measurement of sickness behavior are extremely rare. The concept of clinic modeling randomized trials was used in an animal trial to model clinical complexity and conditions of a randomized clinical trial. METHODS: 80 adult male Wistar rats were randomly assigned to (1) control: anesthesia and sham
