A case-control study of linear energy transfer and relative biological effectiveness related to symptomatic brainstem toxicity following pediatric proton therapy.

BACKGROUND AND PURPOSE: A fixed relative biological effectiveness (RBE) of 1.1 (RBE1.1) is used clinically in proton therapy even though the RBE varies with properties such as dose level and linear energy transfer (LET). We therefore investigated if symptomatic brainstem toxicity in pediatric brain tumor patients treated with proton therapy could be associated with a variable LET and RBE. MATERIALS AND METHODS: 36 patients treated with passive scattering proton therapy were selected for a case-control study from a cohort of 954 pediatric brain tumor patients. Nine children with symptomatic br

Determination of the N-H Bond Dissociation Free Energy in a Pyridine(diimine)molybdenum Complex Prepared by Proton-Coupled Electron Transfer.

The pyridine(diimine)molybdenum bis(imido) complex (iPrPDI)Mo(═NTol)2 (Tol = 4-methylphenyl) was synthesized by the addition of 2 equiv of 4-methylphenylazide to the corresponding molybdenum benzene derivative, (iPrPDI)Mo(η6-C6H6) [iPrPDI = 2,6-(2,6-iPr2C6H3N═CMe)2C5H3N]. Protonation of (iPrPDI)Mo(═NTol)2 with 2,6-lutinidum triflate yielded a cationic molybdenum amido complex, [(iPrPDI)Mo(NHTol)(═NTol)][OTf], which was further transformed into the neutral molybdenum amido (iPrPDI)Mo(NHTol)(═NTol) by reduction with zinc powder. A series of spectroscopic, synthetic, and pKa determination studies

Developing a ratiometric two-photon probe with baseline resolved emissions by through band energy transfer strategy: Tracking mitochondrial SO2 during neuroinflammation.

Sulfur dioxide (SO2) with the largest quantity and widest distribution in the atmosphere is closely related to many nervous system diseases via mitochondria respiration. It is of great significance to monitor this gaseous molecule during various physiological and pathological processes, but currently the task still remains challenging due to the lack of reliable tools. Through-bond energy transfer (TBET) is a relatively new strategy to fabricate ratiometric fluorescent probes, which does not need spectral overlap between the energy donor and acceptor while provides high energy-transfer efficie

Lanthanide MOFs for inducing molecular chirality of achiral stilbazolium with strong circularly polarized luminescence and efficient energy transfer for color tuning.

We present herein an innovative host-guest method to achieve induced molecular chirality from an achiral stilbazolium dye (DSM). The host-guest system is exquisitely designed by encapsulating the dye molecule in the molecule-sized chiral channel of homochiral lanthanide metal-organic frameworks (P-(+)/M-(-)-TbBTC), in which the P- or M-configuration of the dye is unidirectionally generated via a spatial confinement effect of the MOF and solidified by the dangling water molecules in the channel. Induced chirality of DSM is characterized by solid-state circularly polarized luminescence (CPL) and

Spatial correlation of linear energy transfer and relative biological effectiveness with suspected treatment-related toxicities following proton therapy for intracranial tumors.

PURPOSE: The enhanced relative biological effectiveness (RBE) at the end of the proton range might increase the risk of radiation-induced toxicities. This is of special concern for intracranial treatments where several critical organs at risk (OARs) surround the tumor. In the light of this, a retrospective analysis of dose-averaged linear energy transfer (LETd ) and RBE-weighted dose (DRBE ) distributions was conducted for three clinical cases with suspected treatment-related toxicities following intracranial proton therapy. Alternative treatment strategies aiming to reduce toxicity risks are

Surface lanthanide activator doping for constructing highly efficient energy transfer-based nanoprobes for the on-site monitoring of atmospheric sulfur dioxide.

The sensitive and on-site detection of sulfur dioxide (SO2) is in great demand in the fields of food safety and environmental protection. Here, we developed a novel upconversion nanoprobe based on the luminescence energy transfer mechanism for monitoring the atmospheric SO2 concentrations. The lanthanide emitters, Tm3+ ions, were optimized to be doped on the surface layer of the upconversion nanoparticles to improve their energy transfer efficiency by minimizing the distance between the emitters and the surface quencher, a cyanine dye. As a proof-of-concept, the optimal nanoprobe was utilized

A graphene oxide based fluorescence resonance energy transfer (FRET) biosensor for ultrasensitive detection of botulinum neurotoxin A (BoNT/A) enzymatic activity.

Botulinum neurotoxins (BoNTs) are among the most potent toxic bacterial proteins for humans, which make them potential agents for bioterrorism. Therefore, an ultrasensitive detection of BoNTs and their active states is in great need as field-deployable systems for anti-terrorism applications. We report the construction of a novel graphene oxide (GO)-peptide based fluorescence resonance energy transfer (FRET) biosensor for ultrasensitive detection of the BoNT serotype A light chain (BoNT-LcA) protease activity. Facile synthesis and photophysical characterization of luminescent CdTe quantum dots for Forster resonance energy transfer based immunosensing of staphylococcal enterotoxin B.

Aqueous phase synthesis of CdTe quantum dots (QDs) with surface functionalization for bioconjugation remains the best approach for biosensing and bioimaging applications. We present a facile aqueous phase method to prepare CdTe QDs by adjusting precursor and ligand concentrations. CdTe QDs had photoluminescence quantum yield up to ≈33% with a narrow spectral distribution. The powder X-ray diffraction profile elucidated characteristic broad peaks of zinc blende cubic CdTe nanoparticles with 2.5-3 nm average crystalline size having regular spherical morphology as revealed by transmission electro

Real-time fluorescence resonance energy transfer PCR with melting curve analysis for the detection of Opisthorchis viverrini in fish intermediate hosts.

A real-time fluorescence resonance energy transfer (FRET) PCR combined with a melting curve analysis was developed for the detection of Opisthorchis viverrini in its fish intermediate host, cyprinoid fishes. Real-time FRET PCR is based on a fluorescence melting curve analysis of a hybrid between an amplicon generated from a family of repeated DNA elements, the pOV-A6 specific probe sequence (Genbank Accession No. S80278), a 162 bp repeated sequence specific to O. viverrini, and specific fluorophore-labeled probes. The real-time FRET PCR could detect as little as a single metacercaria artificia

Fluorescence resonance energy transfer detection of synaptophysin I and vesicle-associated membrane protein 2 interactions during exocytosis from single live synapses.

To investigate the molecular interactions of synaptophysin I and vesicle-associated membrane protein 2 (VAMP2)/synaptobrevin II during exocytosis, we have used time-lapse videomicroscopy to measure fluorescence resonance energy transfer in live neurons. For this purpose, fluorescent protein variants fused to synaptophysin I or VAMP2 were expressed in rat hippocampal neurons. We show that synaptophysin I and VAMP2 form both homo- and hetero-oligomers on the synaptic vesicle membrane. When exocytosis is stimulated with alpha-latrotoxin, VAMP2 dissociates from synaptophysin I even in the absence

Improving the luminescence properties of the near-infrared phosphor Ca0.8Sr0.2O:Eu2+ via energy transfer and its application in the concentration detection of ethanol solutions.

In this work, a near-infrared (NIR) phosphor Ca0.8Sr0.2O:Eu2+ was prepared via a solid-state method, and its luminescence properties were improved by co-doping with Ce3+ ions. The decay curves of Ca0.8Sr0.2O:Ce3+, Eu2+ confirmed that there was an obvious energy transfer from Ce3+ to Eu2+, which enhanced the quantum efficiency of Ca0.8Sr0.2O:Eu2+. The thermal stability can also be enhanced by introducing Ce, which reduces the probability of non-radiative transitions at elevated temperatures, thereby minimizing electron loss. An NIR light emitting diode (LED) was fabricated by combining a blue L

Dynamics of the Energy Transfer Process in Eu(III) Complexes Containing Polydentate Ligands Based on Pyridine, Quinoline, and Isoquinoline as Chromophoric Antennae.

In this work, we investigated from a theoretical point of view the dynamics of the energy transfer process from the ligand to Eu(III) ion for 12 isomeric species originating from six different complexes differing by nature of the ligand and the total charge. The cationic complexes present the general formula [Eu(L)(H2O)2]+ (where L = bpcd2- = N,N'-bis(2-pyridylmethyl)-trans-1,2-diaminocyclohexane N,N'-diacetate; bQcd2- = N,N'-bis(2-quinolinmethyl)-trans-1,2-diaminocyclohexane N,N'-diacetate; and bisoQcd2- = N,N'-bis(2-isoquinolinmethyl)-trans-1,2-diaminocyclohexane N,N'-diacetate), while the n

Electrochemiluminescent aptasensor based on resonance energy transfer system between CdTe quantum dots and cyanine dyes for the sensitive detection of Ochratoxin A.

In this work, an innovative aptasensor based on electrochemiluminescence resonance energy transfer (ECL-RET) from CdTe quantum dots (QDs) to a cyanine dye (Cy5) fluorophore for the determination of Ochratoxin A (OTA) was fabricated. A strong cathodic ECL emission was obtained by the CdTe QDs modified glassy carbon electrode (GCE). After the immobilization with the capture DNA (cDNA) and the sequential hybridization with the probe DNA-modified Cy5 (pDNA, the aptamer of OTA), the ECL signal enhanced obviously through the ECL-RET. Meanwhile, the spectrum- and distance-related ECL enhancement effe

A theoretical study of energy transfer in Ar(1S) + SO2( X ̃ 1 A ') collisions: Cross sections and rate coefficients for vibrational transitions.

Vibrational transitions, induced by collisions between rare-gas atoms and molecules, play a key role in many problems of interest in physics and chemistry. A theoretical investigation of the translation-to-vibration (T-V) energy transfer process in argon atom and sulfur dioxide molecule collisions is presented here. For such a purpose, the framework of the quasi-classical trajectory (QCT) methodology was followed over the range of translational energies 2 ≤ Etr/kcal mol-1 ≤ 100. A new realistic potential energy surface (PES) for the ArSO2 system was developed using pairwise addition for the fo

Intramolecular energy transfer to S-nitrosylated Cys residues is at the basis of fluorescence sensitivity to nitric oxide in the blue fluorescent protein mTagBFP2.

Fluorescent protein (FP) variants have recently emerged as promising intracellular nitric oxide (NO) sensors based on NO-induced fluorescence loss due to cysteine S-nitrosylation - the covalent addition of a nitroso group to a cysteine thiol within a protein to form an S-nitrosothiol. Here, we investigate the mechanisms underlying this fluorescence loss using a combined experimental and computational approach. We focus on mTagBFP2, a blue fluorescent protein that undergoes a 70% reduction in fluorescence quantum yield and lifetime upon exposure to micromolar NO concentrations. We discriminate,

Substitution of the sole tryptophan of the cupredoxin, amicyanin, with 5-hydroxytryptophan alters fluorescence properties and energy transfer to the type 1 copper site.

Amicyanin is a type 1 copper protein with a single tryptophan residue. Using genetic code expansion, the tryptophan was selectively replaced with the unnatural amino acid, 5-hydroxytryptophan (5-HTP). The 5-HTP substituted amicyanin exhibited absorbance at 300-320 nm, characteristic of 5-HTP and not seen in native amicyanin. The fluorescence emission maximum in 5-HTP substituted amicyanin is redshifted from 318 nm in native amicyanin to 331 nm and to 348 nm in the unfolded protein. The fluorescence quantum yield of 5-HTP substituted amicyanin mutant was much less than that of native amicyanin.

Proton Treatment Techniques for Posterior Fossa Tumors: Consequences for Linear Energy Transfer and Dose-Volume Parameters for the Brainstem and Organs at Risk.

PURPOSE: In proton therapy of posterior fossa tumors, at least partial inclusion of the brainstem in the target is necessary because of its proximity to the tumor and required margins. Additionally, the preferred beam geometry results in directing the field distal edge toward this critical structure, raising concerns for brainstem toxicity. Some treatment techniques place the beam's distal edge within the brainstem (dose-sparing techniques), and others avoid elevated linear energy transfer (LET) of the proton field by placing the distal edge beyond it (LET-sparing techniques).
