Lipogenetic and glycolytic enzyme activities in carcinoma and nonmalignant diseases of the human breast.

Activities of some enzymes associated with carbohydrate and lipid metabolism were determined in 48 human breast carcinomas and compared with those found in 35 nonmalignant breast tumours and also in 13 normal breast tissues. In fibrocystic disease only the activity of citrate lyase was markedly higher (14-fold) than in normal tissue. The activities of the remaining enzymes did not differ significantly from those in normal tissue. Enzyme activities in breast carcinoma were 4--160 x those determined in normal tissue according to the following sequence : phosphofructokinase less than malate NADP

[Activity of gluconeogenetic enzymes of rat kidney cortex during acute hypoxia].

The activities of gluconeogenic enzymes of the rat kidney cortex was studied after exposure to lowered atmospheric pressure (200 mm Hg) for 3 hours. The hypoxic stress was found to cause an increase in the activities of phosphoenolpyruvate carboxykinase and alanine aminotransferase, but failed to affect significantly the activities of fructose-1,6-diphosphatase, glucose-6-phosphatase, and aspartate aminotranspherase. The ratio of glucose-6-phosphatase/hexokinase activities was increased under these conditions.

[Effect of retinol deficiency on the activity of phospholipase A lysosomal enzymes in rat testes].

In the testes homogenates of rats kept on a A-deficient ration and receiving additionally retinyl-acetate (control) and retinoic acid (experiment) the activity of the phospholipases A1 and A2, with 1-acyl-2(1-14C)-oleoyl-SN-glycero-3-galactosidase and acid phosphatase employed as a substrate, was investigated. In the testes of rats receiving retinyl-acetate the activity of the phospholipases A1 and A2 was greatly declining. An addition of retinyl to the testes homogenates of test rats contributed to re-establishing the activity of the phospholipases up to the control level. In the testes of ra

Species differences in activating and inactivating enzymes related to the control of mutagenic metabolites.

Microsomal monooxygenases catalyze the biosynthesis of epoxides from olefinic and aromatic compounds whilst microsomal epoxide hydratase and cytoplasmic glutathione S-transferases are responsible for their further biotransformation. Although catalytically very efficient the cytoplasmic glutathione S-transferases play, due to their subcellular localization, a minor role in the inactivation of epoxides derived from large lipophilic compounds and were, therefore, not included in this study. It was shown with such a lipophilic compound, benzo(a)pyrene, as a model substance and with liver enzyme me

Effect of development on the activity of microsomal drug-metabolizing enzymes in rat liver.

1. The activity of rat liver microsomal drug-metabolizing enzymes was determined at various ages between 6 and 100 days post natum. The enzymes studied were: aromatic hydroxylases by using as substrate biphenyl, which is metabolized by oxidation to 2- and 4-hydroxybiphenyl; nitroreductase by using p-nitrobenzoate as substrate, which is metabolized by reduction to p-aminobenzoate; glucuronyl synthetase by using 4-methylumbelliferone as the substrate, which is conjugated to give 4-methylumbelliferone glucuronide, and cytochrome P-450, which is regarded as the major terminal mixed-function oxidas

Probing Acetylcholinesterase Activity and Enzyme Inhibitor Evaluation by Electrostatic-Gating Nanopores.

Acetylcholinesterase (AChE) plays a crucial role in neuron function and is closely associated with neurodegenerative diseases. Probing AChE-mediated catalytic reactions in biomimetic scenarios (e.g., micro- and nanospace) is of fundamental significance. Here, we present an artificial nanopore to investigate AChE activity by catalyzing acetylcholine (ACh) hydrolysis. In this assay, a predesigned glass nanopore facilitated the electrostatic assembly of the substrate, ACh. Under optimized conditions, AChE exhibited a high catalytic efficiency of up to 94% in a confined environment. The surface ch

Applications of enzyme activity assays in clinical development for rare disease therapeutics.

Many rare diseases, such as lysosomal storage diseases, are caused by genetically driven enzyme deficiencies. Treatment strategies for these diseases have expanded from substrate reduction and enzyme replacement therapies to more recent gene- and cell-based approaches. As therapeutic modalities for treating rare diseases evolve, bioanalytical assays for measuring enzyme activity have been developed in parallel to support a variety of contexts of use (COUs). In this review, we examine the major contexts in which enzyme activity assays are applied to generate reliable data supporting patient sel
