Epigenetics in the Primary Biliary Cholangitis and Primary Sclerosing Cholangitis.

Epigenomics, the study of modifications to genetic material that do not alter the underlying DNA sequence, is generating increasing interest … tion to this phenomenon, as it can be modified by both internal and external exposures (coined the “exposome”). This may explain differences in disease presentation, treatment response, and rates of progression between individuals. Epigenetic changes may also provide a framework for discovering potential biomarkers for diagnosis and screening of PBC and PS …

A proposed organization of multifaceted knowledge in data repositories: structuring record collections and evidence sets for data lakehouses and moonshots.

BACKGROUND: We are developing a cloud-based neurohealth data platform to aggregate and store data from large, complementary historical and ongoing studies focused on traumatic brain injury (TBI), a condition shaped by complex interactions among biological, environmental and contextual factors, and to enable secure, flexible querying across studies. These multifaceted datasets span clinical, physiological, behavioral, and environmental domains, mirroring the complexity of comparable multisite efforts such as the exposome moonshot. OBJECTIVE: Define a scalable and interoperable framework for or…

[Effects of air pollution on cardiovascular health].

Ambient air pollution is a leading modifiable environmental determinant of cardiovascular morbidity and mortality worldwide. Long-term and s … duce exposure and vulnerability. We argue that contemporary cardiovascular prevention must adopt an exposome-oriented perspective and engage with transport, energy, housing and urban planning policies to effectively protect vulnerable patients and communities.

IRON DEFICIENCY PROMOTES HELICOBACTER PYLORI-INDUCED GASTRIC CARCINOGENESIS BY ENABLING RECIPROCITY BETWEEN CARCINOGENIC SECONDARY BILE ACIDS AND PROTECTIVE LONG-CHAIN FATTY ACIDS.

Helicobacter pylori (Hp) is the strongest known risk factor for gastric adenocarcinoma. We previously demonstrated that iron deficiency, a condition of the exposome, augments Hp-induced carcinogenesis. To define mechanisms driving this phenotype, we performed targeted metabolomics in mice which revealed that Hp significantly increased levels of deoxycholic acid (DCA), a carcinogenic secondary bile acid, exclusively under iron-deficient conditions. Further, DCA directly promoted Hp-induced dysplasia. Because bile acids and fatty acids can exert opposing effects on disease, fatty acids were also…

Antagonistic stem cell fates under stress govern decisions between hair greying and melanoma.

The exposome, an individual's lifelong environmental exposure, profoundly impacts health. Somatic tissues undergo functional decline with age, exhibiting characteristic ageing phenotypes, including hair greying and cancer. However, the specific genotoxins, signals and cellular mechanisms underlying each phenotype remain largely unknown. Here we report that melanocyte stem cells (McSCs) and their niche coordinately determine individual stem cell fate through antagonistic, stress-responsive pathways, depending on the type of genotoxic damage incurred. McSC fate tracking in mice revealed that McS…

Circulating miRNAs as Potential Biomarkers for Patient Stratification in Bipolar Disorder: A Combined Review and Data Mining Approach.

Bipolar disorder is a debilitating psychiatric condition that is shaped in a concerted interplay between hereditary and triggering risk factors. Profound depression and mania define the disorder, but high clinical heterogeneity among patients complicates diagnosis as well as pharmacological intervention. Identification of peripheral biomarkers that capture the genomic response to the exposome may thus progress the development of personalized treatment. MicroRNAs (miRNAs) play a prominent role in of post-transcriptional gene regulation in the context of brain development and mental health. They…

Environmental chemicals and endogenous metabolites in bile of USA and Norway patients with primary sclerosing cholangitis.

Primary sclerosing cholangitis (PSC) is a complex bile duct disorder. Its etiology is incompletely understood, but environmental chemicals likely contribute to risk. Patients with PSC have an altered bile metabolome, which may be influenced by environmental chemicals. This novel study utilized state-of-the-art high-resolution mass spectrometry (HRMS) with bile samples to provide the first characterization of environmental chemicals and metabolomics (collectively, the exposome) in PSC patients located in the United States of America (USA) (n = 24) and Norway (n = 30). First, environmental chemi…

Atherosclerosis and the Bidirectional Relationship Between Cancer and Cardiovascular Disease: From Bench to Bedside, Part 2 Management.

The first part of this review highlighted the evolving landscape of atherosclerosis, noting emerging cardiometabolic risk factors, the growing impact of exposomes, and social determinants of health. The prominent role of atherosclerosis in the bidirectional relationship between cardiovascular disease and cancer was also discussed. In this second part, we examine the complex interplay between multimorbid cardio-oncologic patients, cardiometabolic risk factors, and the harmful environments that lend a "syndemic" nature to these chronic diseases. We summarize management strategies targeting disor…

Assessment of Exposure to Di-(2-ethylhexyl) Phthalate (DEHP) Metabolites and Bisphenol A (BPA) and Its Importance for the Prevention of Cardiometabolic Diseases.

Exposomics analyses have highlighted the importance of biomonitoring of human exposure to pollutants, even non-persistent, for the prevention of non-communicable diseases such as obesity, diabetes, non-alcoholic fatty liver disease, atherosclerosis, and cardiovascular diseases. Phthalates and bisphenol A (BPA) are endocrine disrupting chemicals (EDCs) widely used in industry and in a large range of daily life products that increase the risk of endocrine and cardiometabolic diseases especially if the exposure starts during childhood. Thus, biomonitoring of exposure to these compounds is importa

Environmental endocrine disruptors in cardio-oncology: emerging modifiers of cardiovascular vulnerability in patients with cancer.

BACKGROUND: Cardio-oncology has traditionally focused on treatment-related cardiovascular toxicity and conventional cardiovascular risk factors. However, increasing evidence suggests that environmental exposures may contribute to both cancer development and cardiovascular disease through shared biological mechanisms. Endocrine-disrupting chemicals (EDCs) are ubiquitous environmental contaminants capable of interfering with hormonal signaling, metabolic homeostasis, vascular function, and inflammatory pathways. Despite growing evidence linking EDCs to cardiometabolic disorders and hormone-sensi

Air pollution, dementia, and lifespan in the socio-economic gradient of aging: perspective on human aging for planning future experimental studies.

Air pollution (AirPoll) accelerates human aging, as assessed by increased adult mortality and earlier onset of cardiovascular diseases, and dementia. Socio-economic strata (SES) of wealth and education have parallel differences of mortality and these diseases. Children from impoverished homes differ in brain development at birth and in risk of early fat excess and hypertension. To further enhance the healthspan, biogerontologists may consider a wider range of environmental exposures from gestation through later life morbidity that comprise the Gero-Exposome. Experimental studies with rodents a…
