Autoclaved cercarial vaccine (ACV) was found to be highly effective in eliciting protective immunity against experimental Schistosomal mansoni. So, the aim of this study was to analyse ACV biochemically and to study ultrastructural changes inflicted on the cercariae as a result of autoclaving, thus rendering it highly protective. Results of this study showed that approximately 100 microg protein and 44 microg carbohydrate were obtained from 10(3) cercariae. The predominant sugar was fucose. Galactose, glucose, manose, galactosamine and glucosamine were also detected. Threonine, glycine, serine…

The contribution of the ciliary body to the origin of vitreous proteins was investigated in rabbits by incubating explants of this eye component under novel conditions. At the end of incubations for up to 21 h, the tissues were processed histologically and were shown to be in an excellent state of morphological preservation. When radioactive amino acids and fucose were added to the culture medium, protein and glycoprotein synthesis and secretion were detected using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) plus fluorography. The origin of these secretory products was…

Surface properties of microfilariae (mf) and infective larvae of Brugia patei were investigated to compare them to previous studies with the other brugian species. Of all the lectins tested, only wheat germ agglutinin (WGA) binds to the sheath surface of mf indicating the presence of N-acetyl-D-glucosamine as a major surface carbohydrate. However, cuticle of infective larvae failed to show binding of these lectins. Enzyme treatment of mf with N-acetyl-D-glucosaminidase and L-fucosidase has exposed D-mannose, N-acetyl-D-galactosamine and L-fucose on the sheath surface. The binding of lectins to…

Invertebrates rely on post-translational modifications (PTMs) of immune effectors to regulate innate immunity, yet how hemocyanin fucosylation mediates intracellular antimicrobial signaling remains a critical knowledge gap in crustacean immunology. Although fucosylation enhances the in vitro antibacterial activity of Penaeus vannamei hemocyanin (PvHMC), its function in regulating intracellular stress-immune crosstalk remains unclear. Here, we identify a novel axis linking PvHMC fucosylation, endoplasmic reticulum stress (ERS), and antimicrobial peptide (AMP) production to bolster shrimp defens

BACKGROUND: Haptoglobin bifucosylated tetra-antennary glycan have been identified in patients with early stage hepatocellular carcinoma, but its specificity according to the presence or not of cirrhosis has never been assessed. The aims of this study were to determine if haptoglobin bifucosylated tetra-antennary glycan (1) could be a marker of HCC in patients without cirrhosis; (2) could increase the performance of standard alpha-fetoprotein (AFP) or recent blood tests for HCC detection, i.e., lectin-reactive alpha-fetoprotein (AFP-L3), des-gamma-carboxy prothrombin (DCP) and Liver-Cancer-Risk

Humanized monoclonal antibody HMMC-1 established by immunizing transchromosomal mice with a human uterine endometrial cancer cell line has been found to react with the H-antigen carried on core l O-glycans through cotransfection of glycosyltransferases for O-glycans and inhibition of antibody-binding with synthetic oligosaccharides. However, direct binding analysis of an antibody against glycosphingolipids from human erythrocytes with different ABO blood groups revealed that it was able to bind selectively with polar glycolipids in blood group O, but not blood group A, B and AB erythrocytes. BACKGROUND: Aberrant fucosylation observed in cancer cells contributes to an augmented release of fucosylated exosomes into the bloodstream, where miRNAs including miR-4732-3p hold promise as potential tumor biomarkers in our pilot study. However, the mechanisms underlying the sorting of miR-4732-3p into fucosylated exosomes during lung cancer progression remain poorly understood.
METHODS: A fucose-captured strategy based on lentil lectin-magnetic beads was utilized to isolate fucosylated exosomes and evaluate the efficiency for capturing tumor-derived exosomes using nanoparticle tracking ana…

Infection of cells with either ectromelia or lymphocytic choriomeningitis (LCM) virus in the presence of 2-deoxy-D-glucose (2-DOG) inhibited by up to 70% the extent to which the infected cells become susceptible to virus-specific cell-mediated lysis. The concentration of 2-DOG used had little effect on the extent of total protein synthesis (incorporation of [35S] methionine) but inhibited (up to 25%) glycoprotein synthesis, as measured by incorporation of [3H] fucose. This suggested that glycoprotein synthesis was a necessary event for infected cells to become susceptible to T-cell mediated ly…

3'-Sialyl-3-fucosyllactose (3'S3FL) is a representative fucose-containing sialylated human milk oligosaccharide with distinctive biological functions, but its in vitro synthesis is limited by the poor tolerance of α2,3-sialyltransferases toward fucosylated acceptors. Here, we report an engineered α2,3-sialyltransferase variant, R60V, with enhanced catalytic performance toward the 3-fucosyllactose (3-FL) substrate. R60V exhibited a 3.6-fold enhancement in transglycosylation activity and a nearly 5-fold lower apparent Km for 3-FL. Its half-life at 40 °C extended from 42.55 to 89.82 h. Mechanisti…

Enhancement of antibody-dependent cellular cytotoxicity is a promising adjunct approach to achieve HIV control in the absence of antiretroviral therapy but requires the development of potent antibody-dependent cellular cytotoxicity-eliciting antibodies that can recognize diverse HIV-infected cell types. A panel of broadly neutralizing antibodies targeting the HIV envelope was identified that specifically binds both HIV-infected CD4+ T cells and monocyte-derived macrophages. Afucosylated versions of these broadly neutralizing antibodies containing ≈30% less core fucose were generated and elicit…

Luobuma tea is made from the leaves of Apocynum hendersonii (Bt) and A. venetum (Ht) and has been used for a very long time in China and Japan as herbal tea. This study isolated water-soluble polysaccharides from the two species` teas. Physicochemical properties, structural properties, in vitro and in vivo antioxidant and immunomodulatory activities were determined for the first time. The results showed that the Bt and Ht polysaccharides with molecular weights of 31.21 and 49.11 kDa, respectively, composed of arabinose, galactose, rhamnose, glucose, xylose, fucose, and mannose. In this study, ACP-1, a water-soluble polysaccharide was isolated from the roots of Alocasia cucullata (AC) using freeze-thaw treatment (FTT). Structural characterization revealed ACP-1 (2.10×105 Da) to be a homogeneous heteropolysaccharide primarily consisting of glucose, galactose, and arabinose, and a trace of fucose, rhamnose, and glucuronic acid. Methylation analysis and nuclear magnetic resonance spectroscopy revealed that the backbone of ACP-1 consisted of →[3)-β-D-Galp-(1]4→3,6)-β-D-Glcp-(1→3,6)-β-D-Glcp-(1→, with a branch at C-3. In vitro experiments demonstrated that ACP-1 significan…
