Galectin-8 promotes migration and proliferation and prevents apoptosis in U87 glioblastoma cells.

BACKGROUND: Glioblastoma is one of the most aggressive cancers of the brain. Malignant traits of glioblastoma cells include elevated migration, proliferation and survival capabilities. Galectins are unconventionally secreted glycan-binding proteins that modulate processes of cell adhesion, migration, proliferation and apoptosis by interacting with beta-galactosides of cell surface glycoproteins and the extracellular matrix. Galectin-8 is one of the galectins highly expressed in glioblastoma cells. It has a unique selectivity for terminally sialylated glycans recently found enhanced in these hi…

Rouleaux mimicking anti-H in molecularly confirmed para-Bombay siblings: a diagnostic pitfall.

The para-Bombay phenotype is a rare H-deficient blood group variant characterized by markedly reduced or absent expression of H on red blood cells (RBCs) because of mutations in FUT1, with variable secretor status influenced by FUT2. The Bombay phenotype is relatively more common in the Indian population, but the para-Bombay phenotype remains exceedingly rare and is frequently underrecognized because of diagnostic challenges during routine serologic testing. We report molecularly confirmed para-Bombay siblings from India in whom rouleaux-induced pseudoagglutination mimicked anti-H reactiv ity.

Fcer1g and St3gal1: Macrophage-associated angiogenesis biomarkers and therapeutic targets in sepsis-induced acute lung injury.

BACKGROUND: Acute lung injury (ALI) involves the release of growth factors and inflammatory mediators from damaged pulmonary tissues, fostering endothelial cell proliferation, migration, and vascular lumen formation, thereby driving pathological angiogenesis. Macrophages contribute to angiogenesis and vascular homeostasis, but their dysregulation in pathological states worsens vascular dysfunction. This study aims to identify macrophage-associated angiogenesis-related genes as novel diagnostic biomarkers and therapeutic targets for sepsis-associated ALI (SALI). METHODS: Transcriptomic dataset

The silent threat: a true Bombay phenotype with unexpected tolerance to group O-positive incidental transfusion.

INTRODUCTION: Bombay (Oh) blood group individuals are typically transfused with Bombay group red blood cells because transfusion with other blood groups can lead to life-threatening hemolysis. The mutations in the FUT1 gene prevent the expression of the H antigen on red blood cells. METHODS: A 35-year-old man presented with severe anemia (hemoglobin, 4.8 g/dL) and was transfused with 3 units of O RhD-positive packed red blood cells at an outside hospital without adverse reaction. One month later, his blood group at our center was revealed as O RhD positive with panagglutination in reverse gro

CCR3-dependent eosinophil recruitment is regulated by sialyltransferase ST3Gal-IV.

Eosinophil recruitment is a pathological hallmark of many allergic and helminthic diseases. Here, we investigated chemokine receptor CCR3-induced eosinophil recruitment in sialyltransferase St3gal4-/- mice. We found a marked decrease in eosinophil extravasation into CCL11-stimulated cremaster muscles and into the inflamed peritoneal cavity of St3gal4-/- mice. Ex vivo flow chamber assays uncovered reduced adhesion of St3gal4-/- compared to wild type eosinophils. Using flow cytometry, we show reduced binding of CCL11 to St3gal4-/- eosinophils. Further, we noted reduced binding of CCL11 to its ch

Intestinal commensal microbiota and cytokines regulate Fut2+ Paneth cells for gut defense.

Paneth cells are intestinal epithelial cells that release antimicrobial peptides, such as α-defensin as part of host defense. Together with mesenchymal cells, Paneth cells provide niche factors for epithelial stem cell homeostasis. Here, we report two subtypes of murine Paneth cells, differentiated by their production and utilization of fucosyltransferase 2 (Fut2), which regulates α(1,2)fucosylation to create cohabitation niches for commensal bacteria and prevent invasion of the intestine by pathogenic bacteria. The majority of Fut2- Paneth cells were localized in the duodenum, whereas the maj

[The clinical diagnostic significance of auto antibodies to CD74 at axial spondylarthritis.].

The modern diagnostic approaches permit to diagnose axial spondylarthrosis (axSpA) at roentgenologic stage corresponding to ankylosing spondylitis (AS). While early diagnostic of non-roentgenologic axSpA (nr-axSpA) is still complicated. This situation conditions a need in searching new laboratory biomarkers for early diagnostic of spondylarthrosis, including auto-antibodies to antigen CD74 described recently. The purpose of study is to evaluate clinical diagnostic significance of auto-antibodies to antigen CD74 in case of axSpA. The technique of quantitative enzyme-linked immunosorbent assay w

Different effects of androgen on the expression of Fut1, Fut2, Fut4 and Fut9 in male mouse reproductive tract.

The α-(1,2) fucosyltransferases (Fut1 and Fut2) and α-(1,3) fucosyltransferases (Fut4, Fut9) are responsible for the synthesis of Lewis X (LeX) and Lewis Y (LeY) conjugated to glycoproteins. We recently reported that these fucosyltransferases were differentially expressed in the reproductive tract of male mouse. Here, we studied the effect of androgen on fucosyltransferase expression through the use of mouse castration models. We found that Fut1 mRNA and Fut4 mRNA were upregulated, while Fut2 mRNA and Fut9 mRNA were downregulated by androgen in the caput epididymis. However, in the vas deferen

Genetic manipulation of blood group carbohydrates alters development and pathfinding of primary sensory axons of the olfactory systems.

Primary sensory neurons in the vertebrate olfactory systems are characterised by the differential expression of distinct cell surface carbohydrates. We show here that the histo-blood group H carbohydrate is expressed by primary sensory neurons in both the main and accessory olfactory systems while the blood group A carbohydrate is expressed by a subset of vomeronasal neurons in the developing accessory olfactory system. We have used both loss-of-function and gain-of-function approaches to manipulate expression of these carbohydrates in the olfactory system. In null mutant mice lacking the alph

A homozygous nonsense mutation (428G-->A) in the human secretor (FUT2) gene provides resistance to symptomatic norovirus (GGII) infections.

Noroviruses (formerly Norwalk-like viruses) are a major cause of acute gastroenteritis worldwide and are associated with a significant number of nosocomial and food-borne outbreaks. In this study we show that the human secretor FUT2 gene, which codes for an alpha(1,2)-fucosyltransferase synthesizing the H-type 1 antigen in saliva and mucosa, is associated with susceptibility to norovirus infections. Allelic polymorphism characterization at nucleotide 428 for symptomatic (n = 53) and asymptomatic (n = 62) individuals associated with nosocomial and sporadic norovirus outbreaks revealed that homo

Gene delivery by viral vectors in primary cultures of lacrimal gland tissue.

PURPOSE: To test the feasibility of gene transfer into lacrimal gland tissue in primary culture, using different viral vectors. METHODS: Lacrimal glands were dissected from adult Sabra rats and divided by pincers to 0.3-0.4 mm fragments. Tissue was maintained under primary organ culture conditions using the "raft" technique. The ability of three different viral vectors to conduct beta-galactosidase (beta-gal) gene delivery was examined: adenovirus (Ad5CMVLacZ), vaccinia (VSC9), and herpesvirus (tkLTRZ(1)). Tissue fragments were incubated for 60 minutes with one of the viral vectors and transf

Galectin-3-A Multifunctional Molecule and a Key Player in Health and Disease.

Galectins are a family of proteins that belong to one of the most widespread classes of lectins found in all organisms. They are found intracellularly in various structures or secreted into the extracellular space, where they are involved in mediating cellular biological processes such as growth, function, interaction and response. They are known for their ability to bind to carbohydrates, specifically those containing beta-galactose. This narrative review outlines the human galectins, emphasizing Galectin-3, a β-galactoside-binding protein with numerous pleiotropic regulatory activities; it m

ST6GAL1 Glycoengineering Rewires Cytokine Signaling and Preserves Metabolic Fitness in CAR-T Cells Under Galectin-3-Mediated Immunosuppression.

Chimeric antigen receptor (CAR)-T cell therapy has demonstrated remarkable efficacy in hematologic malignancies; however, durable responses remain limited by tumor microenvironment (TME)-mediated immunosuppression. Galectin-3 (Gal-3), a β-galactoside-binding lectin enriched in the TME, contributes to CAR-T cell dysfunction by impairing cytotoxicity, promoting apoptosis, and altering cellular signaling. While we previously demonstrated that enforced expression of the α2,6 sialyltransferase ST6GAL1 reduces galectin binding and improves CAR-T cell function, the mechanistic basis underlying this p

Covalent Anchoring of Enzyme-Activatable Fluorescent Signals for in Situ Imaging and Longitudinal Staging of Cellular Senescence in Pulmonary Fibrosis.

Idiopathic pulmonary fibrosis (IPF) is a fatal interstitial lung disease caused by the pathological accumulation of senescent cells. In this fibrotic microenvironment, senescence-associated β-galactosidase (SA-β-Gal) serves as a key biomarker. However, existing SA-β-Gal molecular probes suffer from signal diffusion and rapid clearance due to the extracellular leakage, compromising imaging fidelity. To address this issue, an enzyme-activated covalent labelling strategy is proposed, and as a design paradigm developing TCFM-Gal, a imaging tool integrating β-galactosidic fluorophore with an ortho-

A glycosaminoglycan microarray identifies galectin-4 binding to sulfated glycosaminoglycans.

Lectin-glycosaminoglycan (GAG) interactions remain incompletely understood despite their potential roles in cell signaling and immune regulation. In this study, we performed a comprehensive profiling of the binding specificity of 49 types of human endogenous lectins-including SIGLECs, C-type lectins, and galectins-for GAGs using GAG microarrays. GAGs containing 6-O-sulfation, such as heparin (HP), chondroitin sulfate C (CSC), and chondroitin sulfate E (CSE), exhibited broad binding to multiple SIGLECs and C-type lectins, whereas non-sulfated or low-sulfated GAGs exhibited minimal interactions,

Mechanism Underlying the Color Differences in Apples Based on Metabolomic and Transcriptomic Analyses.

Apple peel color is a key determinant of fruit quality and consumer preference. To elucidate the molecular basis underlying the peel color difference between 'Jinhong' (JH) apples and 'Hongjinhong' apples (HJH, a deep red bud sport of JH apples), we integrated physiological assays, widely targeted metabolomics, transcriptomics, and quantitative real-time PCR (qRT-PCR) to analyze peel pigment contents, metabolite profiles, and gene expression dynamics during fruit development. The deep red phenotype of HJH peel was primarily attributed to enhanced anthocyanin accumulation rather than alteration

Online HPLC-DPPH-Guided Isolation and Free Radical-Scavenging Evaluation of C-4″-Epimeric Quercetin Diglycosides From Saxifraga tangutica.

Activity-guided separation of medicinal plants requires linking activity signals and chromatographic resolution in complex matrices. In this study, based on a previously established HPLC-DPPH platform, medium-pressure prefractionation and preparative chromatographic purification were applied for the targeted isolation of free radical scavengers from Saxifraga tangutica (S. tangutica). Medium-pressure prefractionation reduced sample complexity and enriched active fractions, while Click XION chromatography concentrated the target fraction. Subsequent purification on the ReproSil-Pur C18 AQ colum

Galectins as stress-integrating regulators of neuroimmune signaling and proteinopathy in the central nervous system.

Galectins are β-galactoside-binding lectins that play increasingly mechanistic functions in central nervous system (CNS) physiology and disease. Over the past decade, a rapidly expanding literature has identified galectins as regulators of microglial activation, misfolded protein pathology, vesicle damage sensing, autophagy, synaptic plasticity, myelination, vascular repair, and neuroimmune communication. Galectins operate across intracellular and extracellular compartments to integrate cellular stress and innate immune signaling. Here, we review CNS studies of galectin-1, galectin-3, galectin
